Abstract

The aim of this research is to know the respon of humic acid and some growth media on the contain of active biological substance in Centella asiatica (L.) Urb. Hopefully, through the application of humic acid in the combination of media, the growth and plant metabolism will improve to produce active biological substance as primar metabolism. The growth media used were : soil & sand (1 : 1) ; soil & husk (1 : 1 ) ; and soil & zeolite (1 : 2). The humic acid treatment were 4 concentration, 0 g/kg media, 4 g/kg media, 8 g/kg media and 12 g/kg media. The research method was Completely Randomized Design, with factorial as treatment design. The variable observed were alkaloid content, flavonoid content, the presence of terpenoid, and biomass weight. The research result showed that the high weight of biomass and alkaloid contain got in the addition of 8 g humic acid in media soil and sand. The high flavonoid contain got in addition of 4 g/kg media in media soil and zeolite. There was brawn ring in the extract analysis dissolved in chloroform, acetic acid, and strong sulfate acid.

Abstract

A research has been carried out with the aims to: 1. study the influence of both the kind and concentration of cytokinins on palea and lemma growth in in vitro culture; 2. study the influence of palea and lemma age towards external application of cytokinin in in vitro culture; and3. determine the best palea-lemma age, kind and concentration of cytokinin which resulted the best growth of palea and lemma in in vitro culture. The used experimental method was Completely Randomised Design (CRD) with factorial treatment pattern. The applied treatment consisted of three factors i.e. palea-lemma age (U): (55, 60, and 65 days after planting); kind of cytokinin (S): (BAP and Kinetin); and Cytokinin Concentration (K): (0, 5, 10, and 15 M) with 3 replications. The observed variables were the palea and lemma growth with the parameters were palea-lemma length and width. The results showed that the kind of cytokinin had influenced the growth of palea-lemma in in vitro culture, and kinetin has better influence on the palea-lemma growth. The age of the palea-lemma determined the responsiveness of the palea-lemma towards external application of cytokinin. Older palea-lemma showed less responsive than younger ones towards external application of plant growth regulators. Moreover, the treatment combination (U1S2K2) (10 µM Kinetin applied to 55-day-old rice palea-lemma) had the best effect on increasing the size of palea-lemma of IR 64 rice.

Abstract

Pandanus tectorius Park., tree of life, which almost all of its parts are used by humans. Industrial fiber made from screw pine leaves has been successfully exported to several countries, while the fruit is consumed as an alternative staple food. Fruit is only produced by female plants, because screw pine is dioecious plant. Farmers obtain screw pine seedling using stem cuttings from female trees, but the number of seedling produced is limited and this technique can damage the plant. Alternative seedling production using seeds has constraints in the form of uncertainty over the sex of the seedling. One of the best ways to provide screw pine seedling is to use a shoot-tip culture technique. Shoot-tip culture is able to reproduce plants efficiently and quickly and does not damage the plant. Seedling will also have the same genetic traits as the selected plant. However, efforts to develop an efficient and fast screw pine shoot-tip culture protocol have not been carried out massively. The induction and shoot multiplication stages still had a low success rate. Another problem in seedling production through tissue culture is the certainty seedlings are true-to-type. This review article aims to reveal the potential of shoot-tip culture for in vitro screw pine seedling production and to test the genetic stability of the screw pine seedling.
