A Comparison Study for Agrobacterium-mediated transformation Method in Sugarcane (Saccharum spp L.) Bambang Sugiharto; Hilda Safitri

Abstract

In order to compare transient expression of gus gene driven by CaMV 35S or rice ubiquitin RUBQ2 promoter, Agrobacterium-mediated transformation was conducted using embryogenic callus and suspension cultures of sugarcane. Histochemical observation of GUS activity after co-cultivation showed that rice ubiquitin promoter produced high level of clear blue spots both in embryogenic callus and suspension cultures, while the CaMV35S promoter was not detected. Regenerated shoots from the infected materials were found higher in suspension cultures than embryogenic callus. The results showed that the rice ubiquitin promoter as well as suspension cultures are more efficient for the Agrobacterium-mediated transformation. However, PCR analysis found there was no amplification of DNA neither for CaMV35 nor rice ubiquitin in genome DNA of regenerated shoot. Lacking of integration of the DNA into the genome showed that the regenerated shoots were not truly transformants might due to the presence of somaclonal variation that was common phenomenon in the regeneration from callus. To avoid the presence of somaclonal variation the transformation was then conduced using in vitro plant without intervening callus phase. Basal segment of in vitro plants prepared from axillary buds of sugarcane were used as the explants for the transformation. The histochemical observation of GUS activity showed that almost all of the infected materials partially exhibited blue color on the basal region. Infected in vitro plants showed rapidly grow and multiplied in theselection medium. Further investigation of the transformation using in vitro plants and Agrobacterium harboring SoSUT1 DNA construct driven by rice actin promoter resulted in development of antibiotic resistant sugarcane shoots. Interestingly the PCR analysis found an insertion of the antibiotic DNA fragment into the sugarcane genome DNA. This result suggests that in vitro plant is an effective target tissue for Agrobacterium-mediated transformation in sugarcane.

Abstract

The economic thinking of contemporary Muslim scholars covers various aspects, such as the Islamic financial system, zakat, waqf, and trade based on Islamic principles. The purpose of writing this article is to describe the Economic Thought of Contemporary Muslim Scholars from the Figures Ahmad Adzhar Basyir, K.H. Sahal Mahfud, K.H. Abdullah bin Nuh, Muhammad Amin Azis. This writing method uses a literature review approach sourced from various articles, journals, and books relevant to the research topic. Many challenges and opportunities faced by contemporary Islamic economic thinkers in disseminating their thoughts in a diverse global society. Contemporary Islamic economic thought has an important value in creating a more just and sustainable economic system based on Islamic principles. Their efforts in integrating religious values with economic science can provide valuable insights for the development of an inclusive and sustainable global economy.

Abstract

This study aims to analyze the influence of product reviews, halal labels, prices, and service quality with product categories as moderating variables on purchasing decisions for cosmetic products on Shopee. The objectives of this study are product reviews, halal labels, prices, service quality, product categories, and Purchasing Decisions. The research was conducted in a quantitative descriptive study using data collection techniques through the distribution of questionnaires or surveys using Google Forms—distributed questionnaires to users of online shopping services, namely Shopee. The type of sample uses non-probability sampling. The data type used is primary data obtained directly from the studied object. The data analysis technique uses Moderated Regression Analysis (MRA). The study results show that the variables of product reviews, prices, and service quality have a positive and significant effect on purchasing decisions, and the presence of product categories as moderators strengthens this influence. While the halal label variable does not have a positive and significant impact on buying decisions, moderating variables further weaken the influence of the halal label.

Abstract

In the study, re-testing factors that are thought to have an impact on the accountability of village fund management was carried out. The purpose of the study was to test the influence of Community Participation, Use of Information Technology, Competence of Village Apparatus, and Internal Control System on the Accountability of Village Fund Management. The object of the study was a village device in Astambul Subdistrict of Astambul District. The population in the study numbered 308. Sampling using judgment sampling. The number of samples analyzed was 150 respondents. Primary data was collected through questionnaires that were directly directed to the respondents. Test the hypothesis using multiple linear regression. The results of the study show that community participation has no impact on the accountability of village management. However, the use of technology, the competence of the village apparatus, and the internal control system have a positive impact on the accountability of the management of the Village Fund. The research is expected to be input, information and consideration for village administration, for example by adding means of community aspiration, maximum use of information technology, procurement of seminars and training, and carrying out an inventory of village assets.

Abstract

This study aims to describe the types and functions of modality in Malay language used by the people of Kampung Tua Tanjung Riau, Batam, in daily interaction and communication. The approach used is qualitative with descriptive method. The data is in the form of community speech containing modality, obtained through free listening, recording, and note-taking techniques. Data validity was tested through triangulation, and analysis was conducted through data reduction, data presentation, and conclusion drawing. The results show that people use four types of modality, namely intensional, epistemic, deontic, and dynamic. Intentional modality is the most dominant type, especially in the meaning of volitional. In addition, three main functions of modality in community communication were found. This finding indicates that people consciously utilize modality as an important part of interaction. The conclusion of this study states that modality has a central role in the communication system of the community of Kampung Tua Tanjung Riau. Tanjung Riau Malay language has a distinctive and rich modality system, which reflects the culture and way of thinking of its speakers. This research is expected to be a contribution to the preservation and development of regional linguistic studies, as well as a reference for further research in the field of local language and culture.

Abstract

This study aims to describe the role of the teacher in strengthening the character of reading in class III students through the school literacy movement at SD Negeri 14 Masalima. The method used is descriptive analysis method using data collection techniques namely observation, interviews and documentation. The research results are as follows: strengthening the character of fond of reading, strengthening curiosity, planning, implementing, and evaluating GLS, program implementation has reached 3 stages, namely the habituation, development, and learning stages. The obstacles are: Some students' motivation to read is still low, lack of initiative in students to read, lack of collection of books in the reading corner, books in the reading corner are rarely replaced and there are still some students who are still not fluent in reading. Abstract

Penelitian ini bertujuan untuk menguji pengaruh Fear of Missing Out (FOMO), Celebrity Endorser dan Kepercayaan Merek Pada Aplikasi Tiktok Terhadap Keputusan Pembelian Produk Viva Kosmetik Pada Mahasiswa Universitas Teknologi Sumbawa. Teknik penentuan sampling gmenggunakan non probability sampling dengan pendekatan porpusive sampling. Populasi pada penelitian ini adalah konsumen atau mahasiswa Universitas Teknologi Sumbawa yang pernah menggunakan dan membeli produk viva kosmetik di aplikasi tiktok. Pengambilan sampel sebanyak 150 sampel dengan menggunakan rumus Hair et al. Jenis data yang digunakan adalah kuantitatif. Sumber data dari penelitian ini adalah data primer dan data sekunder. Metode pengumpulan data yang digunakan adalah kuesioner. Teknik analisis data dalam penelitian ini yaitu analisis regresi linier berganda. Hasil dari penelitian ini menyatakan bahwa Fear of Missing Out tidak berpengaruh signifikan terhadap keputusan pembelian, Celebrity Endorser berpengaruh positif dan signifikan terhadap keputusan pembelian, Kepercayaan Merek berpengaruh positif dan signifikan terhadap keputusan pembelian dan Fear of Missing Out, Celebrity Endorser dan Kepercayaan Merek secara bersama-sama berpengaruh positif dan signifikan terhadap keputusan pembelian. Salah satu penyebab FOMO tidak berpengaruh dikarenakan konsep viral marketing Viva kosmetik lebih fokus pada konten yang lebih menekan pada edukasi dan riview produk dari influencer yang membuat konsumen lebih fokus pada kualitas dan manfaat produk, bukan pada FOMO atau tekanan sosial untuk segera membeli.
