Abstract

Brown seaweed contains fucoidan, a large molecular weight sulfate polysaccharide (about 100,000 Da) which has platelet antiagregation activity. This activity is achieved if the fucoidan has a small molecular weight (3900-7600 Da) so this activity can increase by hydrolized with sellulase Enzym. The purpose of this study was to obtain extract nanoparticles that meet physical quality requirements and have a higher platelet antiagregation activity than brown seaweed extract both before and after hydrolysis. Extraction was using kinetic maseration method using 80% ethanol after that using 2% calcium chloride solution. The results were dried and hydrolyzed with cellulase enzyme and nanoparticles were made by ionic gelation method. Nanoparticle characterization results in particle size of 552.8, polydispersity index of 0.569, potential zeta of +53.5 mV, and spherical shape. In-vitro testing results for platelet antiagregation activity showed the percentage of platelet aggregation inhibition of brown seaweed extract with a concentration 500µg / mL is 22.19% and extracts after hydrolysis was 57.94% and nanoparticles extract after hydrolysis was 72.93%. Extract nanoparticles meet physical quality requirements and extracted nanoparticles after hydrolysis have the highest platelet antiagregation activity compared to brown seaweed extract both before and after hydrolysis.

Abstract

Acne is an inflammatory condition that was caused by Propionibacterium acnes. Sunfl owerseeds (Helianthus annuus L.) with terpenoids and linoleic acid could prevent and treat acne. In theprevious study, sunfl ower seed oil had a Minimum Inhibitory Concentration (MIC) value for P. acnesbacteria ≥1.5% with a Diameter Inhibitory Zone 15.27 mm which was classifi ed as a strong antibacterialagent. This study was intended to develope cream formula (F1-F4) using variations of cetyl alcoholconcentrations of 8, 10, 12% and blanko respectively. The creams were formulated by heating the waterphase and the oil phase separately at 70˚C, then stirred to form the cream. The prepared formulations wereevaluated for organoleptic, homogeneity, type of cream, viscosity and rheology, spread ability, globulesize, centrifugation, and pH. The organoleptic results showed that the cream was white, odorless, softin texture and homogeneous with the type of oil in water (O/W) cream. The viscosity test showed thatthe viscosity of F1-F3 formulas were 490.29; 3886.29; 22510.67 cP and F4 of 111.04 cP with plasticthixotropic fl ow properties. Formula F1-F4 had a spread ability of 8.9; 5,4; 4,4; 7.3 cm2, the globulesizes were 137.78; 133,33; 151.11; 94.44 μm, did not show phase separation during centrifugation andthe pH of the F1-F3 formula were 6.3 and F4 was 5.93. It may be concluded that the evaluation of thephysical and chemical quality of sunfl ower seed oil cream are within the acceptable standard.

Abstract

Minyak biji bunga matahari memiliki kandungan terpenoid dan asam linoleat, sehingga dapat menghambat Propionibacterium acnes dan dapat diformulasikan menjadi sediaan krim. Penelitian ini bertujuan untuk memperoleh formula terbaik dari sediaan krim minyak biji bunga matahari. Minyak biji bunga matahari ditentukan nilai KHM dan didapat hasil sebesar 1,5%. Dilakukan formulasi sediaan krim menggunakan desain faktorial 22 dengan variasi konsentrasi gliseril mono stearat dan setil alkohol. Sediaan krim dibuat dengan cara memanaskan fase air dan fase minyak masing-masing pada suhu 70?C, lalu dicampur dan diaduk sampai terbentuk sediaan krim. Terhadap sediaan dilakukan evaluasi baik secara fisika, kimia, maupun mikrobiologi. Dari hasil evaluasi dilakukan overlaid counter plot untuk mendapatkan formula terbaik sesuai syarat mutu fisika, kimia, dan mikrobiologi. Formula terbaik didapat pada formula III dengan konsentrasi gliserilmonostearat sebesar 9% dan setil alcohol sebesar 1%. Dari hasil evaluasi pada formula III didapat sediaan krim berwarna putih, tidak berbau, dan tekstur lembut dengan nilai yield value sebesar 90,77 dyne/cm2, memiliki ukuran globul sebesar 59,90 µm, viskositas sebesar 32705,33 cP dengan sifat alir tiksotropi plastis, kemampuan menyebar sebesar 33,62 cm2, dan pH sebesar 5,95, dan DDH sebesar 15,28 mm.

Abstract

Daun sosor bebek (Kalanchoe pinnata P.) merupakan tanaman obat yang diketahui memiliki aktivitas antiseptik. Tujuan dari penelitian ini untuk melihat pengaruh konsentrasi Carbophol 980 dan Cremophor RH 40 terhadap mutu fisik dan pH sediaan gel ekstrak kental daun sosor bebek yang dianalisa dengan metode ANOVA satu arah. Daun sosor bebek diekstrasi dengan metode maserasi menggunakan etanol 96%, kemudian diformulasikan menjadi sediaan gel dengan kombinasi Carbophol 980 sebagai gelling agent dan Cremophor RH 40 sebagai wetting agent. Sediaan gel dibuat 8 formula dengan variasi konsentrasi Carbophol 980 0,5% dan 1,0%, dan Cremophor RH 40 0,5%; 1,0%; 1,5%; dan 2.,%. Sediaan gel yang diperoleh dilakukan evaluasi meliputi pemeriksaan organoleptik, homogenitas, viskositas dan sifat alir, kemampuan menyebar, dan pH. Sediaan gel yang diperoleh berwarna coklat, berbau parfum, dan jernih, viskositas pada rpm 0,5 berkisar 430,67–1015, 33 dPa.s, sifat alir plastis, kemampuan menyebar 2491,20–2111,77 mm2, pH 5,0–5,6. Hasil uji statistic menggunakan ANOVA satu arah dengan α 5%, didapat bahwa variasi konsentrasi Carbophol 980 dan Cremophor RH 40 memberikan perbedaan yang bermakna terhadap viskositas, kemampuan menyebar, dan pH. Berdasarkan hasil evaluasi didapatkan formula 5 yang menggunakan Carbophol 980 dan Cremophor RH 40 pada konsentrasi 0,5% sebagai formula terbaik.       Kata Kunci: Variasi onsentrasi, Carbophol 980, Cremophor RH 40, Gel, Ekstrak Daun Sosor Bebek  Duck bill leaf (Kalanchoe pinnata P.) is a medicinal plant known to have antiseptic activity. The purpose of this study was to examine the effect of the concentration of Carbophol 980 and Cremophor RH 40 on the physical quality and pH of the thick extract gel preparations of duck bill of leaf extract which was analyzed by one way ANOVA method. Duck bill leaves were extracted by maceration method using 96% ethanol, then formulated into a gel preparation with a combination of Carbophol 980 as a gelling agent and Cremophor Rh 40 as a wetting agent. The gel preparations were made of 8 formulas with various concentrations of Carbophol 980 0.5% and 1.0%, and Cremophor RH 40 0.5%, 1.0%, 1.5%, and 2.0%. The obtained gel preparations were evaluated including organoleptic examination, homogeneity, viscosity and flowability, dispersibility, and pH. The gel preparation obtained was brown in color, smelled of perfume, and was clear, viscosity at rpm 0.5 ranged from 430.67–1015, 33 dPa.s, plastic flowability, dispersibility 2491.20–2111.77 mm2, pH 5.0–5.6. The results of statistical tests using one-way ANOVA with 5%, it was found that variations in the concentration of Carbophol 980 and Cremophor RH 40 gave significant differences in viscosity, dispersibility, and pH. Based on the evaluation results obtained formula 5 which uses Carbophol 980 and Cremophor RH 40 at a concentration of 0.5% as the best formula.
