Abstract

One of the approachs to understand the structure-function of the gene is by studying its mutants. A few of sup45 ts mutants was isolated from Saccharomyces cerevisiae strain BSC483/1a induced by EMS. The concentration EMS of 1.5% (v/v) was found to be the most effective concentration on this study. Eight of sup45 ts mutants were selected from 515 allosupressor mutants. All of the mutants showed lower growth rate and maximum growth compared to that of the wild type. In addition of allosuppression phenotype, six of the mutant showed temperature sensitive and the other two exhibited temperature and paromomycine sensitive. Quantitation of an allosuppression phenotype using plasmid-borned gene fusion, PGK-termination codon-LACZ, showed that the mutants have a variation level of allosuppression. Temperature sensitivity analysis of the mutants showed that one, three, and one mutants were unviable at 32, 34, and 37OC, respectively. Further analysis on two of the mutants showed that the allosuppression level increased by increasing temperature growth. All the data suggested that the mutants were allelic specific mutants.

Abstract

Penelitian ini bertujuan untuk mengungkapkan pengaruh pembelajaran IPA berbasis SETS terhadap cross disciplinary knowledge peserta didik kelas VII SMP di Kota Makassar. Penelitian ini merupakan penelitian kuasi eksperimen dengan populasi SMP di kota Makassar yang menerapkan Kurikulum 2013. Sampel diambil sebanyak 3 sekolah yang dipilih berdasarkan stratifikasi sekolah dimana SMPN 12 Makassar mewakili sekolah kategori tinggi, SMP Islam Athirah Makassar mewakili sekolah kategori sedang, serta SMPN 3 Makassar mewakili sekolah kategori rendah. Penentuan kelas eksperimen dan kontrol pada masing-masing sekolah dilakukan dengan teknik cluster random sampling. Instrumen yang digunakan adalah tes dengan bentuk soal pilihan majemuk beralasan. Analisis data menggunakan independent sample t- test serta uji U-mann whitney. Hasil penelitian menunjukkan bahwa pada sekolah SMP kategori tinggi dan sedang, capaian cross disciplinary knowledge kelompok eksperimen yang mendapatkan pembelajaran IPA berbasis SETS rata-rata lebih tinggi secara signifikan dibandingkan dengan kelompok kontrol yang mendapatkan pembelajaran IPA konvensional, sedangkan pada sekolah kategori rendah capaian cross disciplinary knowledge kelompok eksperimen dan kontrol tidak berbeda secara signifikan.Kata kunci: SETS, cross disciplinary knowledge

Abstract

The virus called HIV can infect human cells andcause the disease called AIDS. HIV infection is principallythe integration between the DNA of the HIV andthat of the host cell so that the DNA of the HIV undergoesreplication at the time the host cell undergoes fission.The HIV is unable to conduct a metabolism processwithout any host cell. In order to survive, the HIVmakes use of the energy, precursor molecules, and essentialenzymes of the host cell. The competition occurringbetween the HIV and the host cell will result inthe host cell's growth b,eing disturbed. Human cellswhch can be infected by the HIV include the T helperlymphocyte cells, the macrophage cells, and the dendriticcells. The T helper lymphocyte cells play a partin the biosynthetic process of antibodies. The macrophagecells are very useful in the protection of cellsfrom infection caused by a number of pathogenousmicroorganisms as well as toxic chemical substances.The dendritic cells function as the cells providing antigensas well as the cells complementingthe T cytotoxiccells. The three types of cells above are responsible forthe body's immunity system. The occurrence of H V-infection on these cells causes a decline in the humanbody's immunity system and results in AIDS. The contagonof HIV can happen via sexual intercourse (heterosexualor homosexual), blood transfusion, the use ofthe same needle for injections to different persons, andsaliva. Prevention of HTV infection can be done byavoiding the behaviors described above whle for curativetreatment can possibly be done by gving antibioticswhich can hinder the biosynthetic process of thereverse transcriptase enzyme protein and decrease theactivity of the enzymes playing a part in the modificationprocess of a foreign DNA. In social life, ostracismon the IW carrier is not approved because the conta-@on of ttus virus is not via air, the slun, or clothes.

Abstract

Scientific writing skills are a very urgent need for students. This study aims to show the effect of the implementation of disciplinary literacy instruction as a strategy for scientific writing skills. The population of the study is the senior high school in Sleman region with  quasi-experimental method. The results of two replications were analyzed and showed normal and homogeneous results. This indicates that there is no difference in the learning outcomes of the two classes and shows the similarity of the cognitive abilities of the two classes, so research may proceed. The treatment given was in the form of disciplining literacy habits in students by using articles that contained problem-solving. Data collection using authentic assessment was in the form of student laboratory work reports. The analysis technique used to analyze the effect of the implementation of disciplinary literacy instruction on scientific writing skills was ANOVA test analysis at 0.05 significance level. The results showed that the sig value below 0.05 that is 0.01. This number indicates that there is an effect of the implementation of disciplinary literacy instruction to the students' writing skills on the material of electrolyte and non-electrolyte solution in the first grade of senior high school.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh waktu kontak dan pHmedia terhadap efisiensi biosorpsi ion Pb2+ oleh sel ragi S. cerevisiae denganadanya pengaruh interferensi ion Cd2+. Subjek penelitian ini adalah ragi S.cerevisiae dan objek penelitian ini adalah biosorpsi ragi S. cerevisiae terhadap ionPb2+. Penelitian ini dilakukan secara bertahap untuk mengetahui kondisi biosorpsimeliputi: (1) Penentuan profil pertumbuhan S. cerevisiae pada rentang waktu 0, 2,4, 6, 8, 16, 24 dan 48 jam, (2) pengukuran terhadap pertumbuhan ragi S.cerevisiae dengan konsentrasi Pb2+ 0, 5, 10, 15, 20, dan 25 ppm, (3) pengukuranterhadap pertumbuhan ragi S. cerevisiae pada variasi waktu kontak 0, 2, 4, 6, 8dan 10 jam tanpa dan dengan interferensi Cd2+, (4) pengukuran terhadappertumbuhan ragi S. cerevisiae dengan pH media 3, 5, 7 dan 9 tanpa dan denganinterferensi Cd2+. Karakterisasi sampel dengan menggunakan Spectronic 20 danSpektrofotometer Serapan Atom (SSA). Hasil penelitian menunjukkan bahwavariasi waktu kontak dan pH media berpengaruh terhadap pertumbuhan ragi S.cerevisiae. Pada waktu kontak 6 jam menunjukkan efisiensi pertumbuhan sebesar45,31%. Pada pH 5 ragi S. cerevisiae menunjukkan biosorpsi optimum yaitu43,78%.

Abstract

Penelitian ini merupakan penelitian pengembangan dibidang pendidikan kimia. Tujuan penelitian ini adalah mengembangkan dan menentukan kualitas Lembar Kerja Siswa materi kimia larutan berbasis potensi lokal pengolahan limbah Daerah Bantul.Penelitian ini menggunakan model pengembangan prosedural yang mengadopsi model Borg Gall. Pada produk awal Lembar Kerja Siswa diberi masukan oleh ahli media, ahli materi, dan peer review. Selanjutnya produk Lembar Kerja Siswa dinilai kualitasnya oleh 5 orang guru kimia. Instrumen penilaian berupa angket yang berisi 6 aspek penilaian dengan butir penilaian sebanyak 38. Penilaian dilakukan oleh 5 orang guru kimia SMA di Kabupaten Bantul.Hasil penelitian ini adalah produk berupa Lembar Kerja Siswa materi kimia larutan berbasis potensi lokal pengolahan limbah Daerah Bantul. Berdasarkan penilaian dari 5 orang guru kimia SMA, Lembar Kerja Siswa ini memiliki kategori kualitas sangat baik (SB) dengan skor rata-rata 161 (persentase keidealan 84,74%). Oleh karena itu, Lembar Kerja Siswa ini layak digunakan sebagai sumber belajar materi kimia larutan untuk siswa SMA.
