Abstract

This paper reports an optimization technique of effervescent tablet formulation from the ethanolic extract of “asam kandis” dried rinds (Garcinia cowa Roxb) and an assay of its reconstituted solution effect on rat’s body weight and feeding behavior. Formulation optimization was done by factorial design of 2 factors (extract concentration; composition of tablet disintegrant) and 3 levels. Nine formulas (FI – FIX) were prepared and subjected to evaluations including granule’s physicochemical and tablet’s pharmaceutical properties. The best formula was then chosen and given orally to 4 groups of rats: that was given standard food, high fat diet, high fat diet with reconstituted tablet 250 mg and high fat diet with reconstituted tablet 500 mg respectively. All groups were given 10 g of standard food every day for 10 days followed by high fat diet for the next 22 days. Two treatment groups were given reconstituted tablet at day 11th. The amount of food consumed by each group was weighed and rat’s body weight was measured. The optimization technique showed that both factors influenced the physicochemical properties of granules and the pharmaceutical properties of tablets significantly (p < 0.05). FV (extract concentration 500 mg, disintegrant: sodium bicarbonat 500 mg + citric acid 200 mg + tartaric acid 200mg) was chosen as the best.  Results showed that administration of reconstituted tablet FV to rats increased the bodyweight of the treated groups for the first week which then decreased during the next 3 week observation, while the body weight of control group kept increasing. Reconstituted tablet at dose 500 mg significantly influenced rat’s bodyweight and the feeding behavior (p<0.05). The group given reconstituted tablet 500 mg had the lightest body weight and consumed the least food during 3 weeks observation.

Abstract

A study on the use of pregelatinized glutinous rice starch in combination with sodium carboxy methyl cellulose as sustained release matrix and sodium diclofenac as a model has been conducted. Two formulas were prepared with these ratio of pregelatized glutinous rice starch to Na CMC of (42,70% : 14,25 %) (F1) and (28,5% : 28,5 %) (F2) respectively. Evaluation of pharmaceutical properties of the tablets showed that all formulas fulfilled the requirement in term of weight uniformity, content uniformity, friability, hardness and disintegration time. Dissolution profile showed that F1 showed down the release of drug at 86% in 12 hours, while F2 80,16 % in 12 hours.

Abstract

Balutan luka primer dari film yang mengandung kolagen kulit ikan gabus (Channa striata) dapat menciptakan lingkungan luka dengan kelembapan seimbang (moisture balance) dikenal sebagai metode modern dressing, sehingga akan mempercepat fibrinolisis, mempercepat angiogenesis, menurunkan risiko infeksi, mempercepat pembentukan growth factor, mempercepat pembentukan sel aktif. Kolagen mempunyai peranan penting dalam proses penyembuhan luka karena kolagen mempunyai kemampuan dalam hemostasis, interaksi dengan trombosit, interaksi dengan fibronektin dan meningkatkan eksudasi cairan, meningkatkan komponen selular, meningkatkan faktor pertumbuhan dan mendorong proses fibroplasia. Telah dilakukan pembuatan film untuk balutan primer luka bakar yang mengandung kolagen yang diisolasi dari kulit ikan gabus (Channa striana) 1%, dengan bahan tambahan PVA 4%, propilenglikol 5%, Gliserin 10%, dan pengawet nipagin dan nipasol. Hasil evaluasi gel dan film kolagen diperoleh pH gel 5,44, viskosita gel 1,0378 Poise, tebal film 0,7378 mm, berat jenis film 0,6427 g/cm3 dan water uptake film 47, 41 % (w/w). Dari penelitian yang telah dilakukan dapat disimpulkan hasil evaluasi gel dan film memenuh syarat, tapi perlu penyempurnaan untuk cara pengeringan film, karena film yang mengandung kolagen tidak dapat dikeringkan pada suhu diatas 40oC karena akan merubah kolagen jadi gelatin.

Abstract

Solid dispersion made with the weight ratio (b/b) using the dissolving method. DTA analysis of the solid dispersion obtained in Eudragit L 100 affects the position and sharpness of the peak. X-ray diffraction analysis showed that of clarithromycin - Eudragit L 100 is in amorphous form and did not produce a new crystalline phase. SEM analysis showed that crystalline of Eudragit L 100 is smaller (amorphous) and stick to the surface of the clarithromycin crystal. IR spectrophotometer analysis showed that there is no chemical interaction between clarithromycin and Eudragit L 100. Clarithromycin assay was done by HPLC method, using mobile phase methanol and 0.067 M KH2PO4 (13:7) plus fospat acid pH 4.0. The dissolution profile by the time of 60 minutes are clarithromycin 45.73%, CF (1:1) 49.86%, DP (1:1) 51.53%, DP (2:1) 55.87% and DP (1:2) 58.97% respectively. Solid dispersion 1:2 (w/w) have an increased dissolution rate compared to clarithromycin.

Abstract

Improvement of the dissolution rate of ketoprofen as a model for a poorly water-soluble drug by solid state co-grinding technique with hydrophilic polymer hydroxypropyl methylcellulose E6 (HPMC) using mortar grinder apparatus have been investigated. Three different formulations prepared with varying drug : carrier ratio 1:2, 1:1, and 2:1 (w/w). Physical mixture prepared with ratio 1:1 (w/w). X-ray diffraction showed decrease of peak intensity of ketoprofen. DSC analysis showed decrease of melting point of ketoprofen. FTIR analysis showed no chemical interaction between drug and polymer. In this investigation, it was found that solubility tended to increase with increase in the amount of polymer used. Solid dispersion ratio 1:2 showed the highest solubility (6,5 fold). However, unlike the solubility results, the dissolution rate increased with decreases in polymer concentration due to thick diffusion layer formation of the polymer. Solid dispersion ratio 2:1 showed the highest dissolution (2,1 fold).

Abstract

Floating tablet is one of the most suitable dosage forms that used for delivering long term drug release. The objective of this study was to evaluate Hydroxypropyl cellulose (HPC), Hydroxypropyl Methyl Cellulose (HPMC) K100M, and the combination as matrix in manufacturing floating tablets. Metformin HCl, an anti-diabetic, was used as a drug model. Metformin HCl floating tablet was manufactured by wet granulation method in three formulas using variation of matrix, which were 40% of HPC (F1), 40% of HPMC K100M (F2), combination of 20% HPC and 20% HPMC K100M (F3). Prior to tablet compaction, evaluation for granules were done which included moisture content, angle of repose using, bulk and tapped density, Hausner ratio, and compressibility. The evaluations of floating tablet were physical properties, floating ability, and in vitro drug release. The average of floating lag time for F1, F2, F3, were 7 minutes 13 seconds; 5 minutes 27 seconds; and 14 minutes 5 seconds, respectively. In addition, the floating time for F1 was 3 hours 16 minutes whereas F2, F3 were more than 48 hours. F2 showed the best floating ability to retain the drug release, which was 84.68% over 8 hours, while F1 and F3 were completely dissolved less than 6 hours.

Abstract

Ranitidine HCl is a histamine H2-antagonist which is used in the treatment of benign gastric and duodenal ulceration. In order to improve its bioavailability and half life time, microcapsule is prepared to extend the dose regiment. The aim of this study was to formulate and optimize ranitidine HCl containing microcapsule to prepare a suitable sustained release delivery system using factorial design. Microspheres were prepared using ethylcellulose by solvent evaporation method. The effect of different formulation variables, including stabilizer concentration (1-2 %) and drug/polymer ratio (2:2-1:2) on appearance, and entrapment efficiency was investigated. Data analysis showed that microspheres with optimum entrapment efficiency could be prepared using 2 % span 80, and 1:2 drug/polymer ratio. The results showed spherical shaped microcapsules, and porous, with realeasing time up to 10,55 hours.

Abstract

Kemajuan di bidang teknologi dan transportasi menyebabkan produk farmasi dengan cepat menyebar. Tingginya kebutuhan obat keras oleh masyarakat, murahnya harga obat di sarana tidak berwenang, masih rendahnya pengetahuan dan kesadaran masyarakat akan keamanan, kemanfaat dan mutu obat yang disediakan sarana yang berwenang mengakibatkan fenomena tingginya peredaran obat keras di sarana yang tidak berwenang. Penelitian ini bertujuan untuk mengetahui kinerja BBPOM di Padang, tingkat keberhasilan penegakan hukum dan tingkat keberhasilan peningkatan kesadaran masyarakat. Penelitian bersifat evaluatif dengan pendekatan triangulasi. Responden kuesioner sejumlah 70 orang yang berasal dari 7 kabupaten/kota di Sumatera Barat. Responden wawancara terstruktur sejumlah 10 orang yang berasal dari pejabat struktural BBPOM di Padang dan Jayapura. Sebanyak 5 jenis kuesioner divalidasi dengan nilai reliabilitas 0.8427; 0.8507; 0.7493; 0.7399 dan 0.8272. Hasil penelitian menunjukkan bahwa terjadi trend peningkatan peredaran obat keras di TOB sebesar 24%; 45% dan 49%, sedang di swalayan terjadi penurunan sebesar 17%, 17% dan 8%. Penegakan hukum di bidang obat terjadi peningkatan sebesar 33.3%; 70.0% dan penurunan 18.2%. Pemberdayaan konsumen melalui peningkatan pemahaman obat keras (92.9% dan 100.0%). Kesimpulan penelitian ini adalah kinerja BBPOM di Padang telah cukup optimal dengan strategi pengawasan rutin, pemberdayaan konsumen, penegakan hukum dan sinergitas dengan stakeholder terkait
