Abstract

Cellulase enzyme activity of Penicillium sp. LBKURCC20 Isolate of Riau can beincreased by concentrating the enzyme using 80% saturated solid ammonium sulfate(NH 4 ) 2 SO 4 anhydrous. Precipitated enzyme was redissolved by the addition of acetatebuffer solution pH 5.5 to 1/70 times its original volume. Cellulase enzyme activity wasdetermined using substrates CMC and avicel. CMCase and avicelase enzyme activity ofthe crude enzyme was (0.0164 ± 0.0036) U/mL and (0.003 ± 0.0025) U/mL,respectively. CMCase and avicelase enzyme activity after concentration was (0.219 ±0.0579) U/mL and (0.03 ± 0.03) U/mL, respectively. The test results showed that theactivity of concentrated CMCase increased 13.5 times and avicelase enzyme activityincreased 10 times after concentrating.

Abstract

This study aims to analyze students' perceptions of the implementation of science laboratory practicums at public middle school 020 Pekanbaru and public middle school 7 Pangkalan Kuras, Riau Province. Using a quantitative survey method, this study involved 75 student respondents. The instrument used was a questionnaire consisting of 26 statements, analyzed descriptively. The results show that students' perceptions of the preparation, implementation, and conclusion stages of the practicum fall into the "good" category, with urban schools having a higher percentage. In conclusion, the implementation of practicums in urban schools is better than in rural schools, bringing attention to the need for quality improvements in rural school practicums.

Abstract

Perkembangan zaman yang semakin pesat memerlukan dunia pendidikan untuk berinovasi dan beradaptasi dengan kemajuan teknologi. Pendidikan sains, khususnya di tingkat SMP, perlu disesuaikan dengan keterampilan abad ke-21 seperti berpikir kritis dan keterampilan komunikasi. Penelitian ini bertujuan untuk menganalisis kebutuhan pembelajaran berbasis Case-Based Learning (CBL) yang dibantu media poster comment dalam pembelajaran IPA di SMP. Melalui metode analisis kebutuhan, penelitian ini menggali permasalahan dalam pembelajaran serta kebutuhan siswa untuk meningkatkan keterampilan berpikir kritis dan komunikasi tulisan. Hasil survei menunjukkan adanya kebutuhan yang tinggi terhadap penerapan model pembelajaran CBL berbantuan media poster comment, yang dapat membuat pembelajaran lebih menarik dan efektif. Dengan demikian, langkah selanjutnya adalah mengembangkan perangkat pembelajaran berbasis CBL dan media poster comment untuk memenuhi kebutuhan tersebut dan meningkatkan kualitas pembelajaran IPA.
