Abstract

Sugarcane (Saccharum officinarum L.) is one of the plantation commodities that have high economic value because it is the main raw material in the manufacture of sugar. Generally, sugarcane propagation was done by vegetative cuttings, but this method has drawbacks. In vitro plant, culture is an alternative technique for producing quality sugarcane seed, a high number of seeds with shoot multiplication. This research aims to review the influence of concentration NAA and BAP on shoot multiplication of sugarcane (Saccharum officinarum L.). The study was conducted in the Laboratory of Superior Farm Seed Management Unit, Indonesian Agency for Agricultural Research and Development from June until August 2021. Explant material used was young rolled leaves collected from AMS variety sugarcane. The experiment was arranged in Factorial Completely Randomized Design (RAL), with 12 treatments, and five replication. Explants were cultured during 8 weeks in MS media with concentration NAA ( 0 ppm, 0.5 ppm, and 1 ppm), and BAP (0 ppm, 1 ppm, and 2 ppm, 3 ppm). The parameters measured were the date of shoot initiation, number of shoots, length of shoot, number of leaves, date of root initiation, number of the root, length of root, and color of the shoot. The data were analyzed using ANOVA at the confidence level of 95%. The results showed that interaction between NAA and BAP in promoting Saccharum officinarum L. Micro cutting growth was observed. The combination between NAA of 0 ppm and BAP of 2 ppm was found to be the best in date of shoot initiation, number of shoots, and number of the leaf. The combination between NAA 1 ppm and BAP 0 ppm was recommended to promote the root number and length of the root.

Abstract

Research is conducted in Dayeuhluhur Subdistrict, Cilacap County, Central Java from September to in November 2020. The study aims to gain the best influence of planting medi and fertilizer dose of compound NPK of tubo bala plant (Globba leucantha var. bicolor Holttum). The research method using the experimental method of Rancangan Acak Kelompok (RAK) there are 12 treatments repeated five times and consisting of 60 experimental units. The observed parameters namely the beginnings appear flowers, the beginnings of bloom flowers, the presentages of flower formation, the presentases of blooming flowers, the amount of fruit and the number of bulbils. The results of the study showed in treatment I (Tanah:pasir:bokashi (1:1:1) + NPK 0 gram) giving the best result in early mumculnya of 25th day interest after planting, early blooming flower of 36th day after planting, presentation of benign interest of 80%, presentage of blooming interest of 72.22%, fruit amount of 4 fruits and number of bulbils of 13 bulbils.

Abstract

Straw mushroom is one of the horticultural commodities that is in great demand by the community. The successful cultivation of straw mushrooms is determined by the quality of the seeds of the molten mushroom. Tissue culture is one of the efforts to get quality seeds. Quality seedlings are seen from the brood of the merang fungus and the quality of pure culture. Making pure culture is still constrained in making in vitro media, especially alternative media materials. This study aims to obtain the best results from the growth of G2 FP005 mycelia on alternative media materials in vitro. This research was conducted at the Biotechnology and Plant Breeding Laboratory, Faculty of Agriculture, Singaperbangsa University, Karawang from April to May 2023. The research method used was a single factor Complete Randomized Design (CRD) with 7 treatments and 5 replications: A (100% PDA); B (100% Charcoal Husk Solution); C (100% Husk Solution); D (80% PDA + 20% Husk Charcoal Solution), E (75% PDA + 25% Husk Solution), F (50% Husk Charcoal Solution + 50% Husk Solution); G (50% PDA + 25% Husk Charcoal Solution + 25% Husk Solution). The effect of the treatment was analyzed using variance and if the F test was significant at 1% level, then to find out which treatment was the best it was continued with the DMRT (Duncan Multiple Range Test) advanced test at the 1% level of significance. The results achieved from this study were that there was a significant effect of using alternative media materials on the growth of the straw mushroom mycelia. Alternative media 80% PDA + 20% rice husk charcoal gave the highest mycelia colony diameter of 7.88 cm and the highest mycelial colony growth rate of 2,878 cm/day. Keywords: Unsika Faperta Isolate, Straw Mushroom, Tissue Culture.
