Abstract

Gurami (Osphronemus gouramy Lac.) is a popular fish species among Indonesian people. Several Gurami strains have been cultivated by fish farmer, one of which is Gurami Soang. This strain is belived to have a faster growth rate compared to other strains. However, like other strains, the fingerling of Soang strain have also a low survival and suceptible to disease, especially that caused by Aeromonas hydrophila infection. It has been proved that seeds from a single spawning event show varibale disease resistance. The difference in resistance among individuals is suggested related to the difference in their genetic component. One of the genes responsible for resistance is Major Histocompatibility Complex (MHC) class II B gene. Variability in resistance can be analyzed by using PCR - RFLP technique. PCR-RFLP is a technique that can produce a specific DNA fragments by PCR, followed by cutting the PCR product using restriction enzymes to describe the presence or absence of restriction sites in DNA fragments. This research aims to determine genetic marker to differiantiate between resitant and irresistant individual of Gurami Soang infected by A. hydrophila which die at a different time priod based on PCR-RFLP MHC class IIB gene. The study used survey method with purposive random sampling. The Data of PCR-RFLP band patterns were analyzed descriptively. The result indicated that cutting of the MHC class II B gene using HinfI produce two RFLP bands with 300 bp and 100 bp length in all samples. Meanwhile, the MHC IIB gene was not cuted by PstI, HindIII, BamHI and EcoRI enzymes forall samples. These mean that MHC II gene in all individuals were monomorphic. Therefore,it can be concluded that there is no specific genetic marker to differentiate gurami soang individulas which was dying in different time periods.

Abstract

Kompetensi vokasional siswa SMK perikanan perlu ditingkatkan melalui pendekatan pembelajaran yang adaptif terhadap keberagaman kemampuan dan gaya belajar. Program pengabdian ini bertujuan menguatkan kompetensi siswa dalam teknik pemijahan induksi melalui pelatihan berbasis pembelajaran adaptif di SMKN 1 Bawang Banjarnegara pada periode Juni–Oktober 2025. Mitra kegiatan adalah guru dan 35 siswa kelas XI Program Agribisnis Perikanan. Metode pelaksanaan meliputi persiapan, sosialisasi, pelatihan teori–praktik, monitoring evaluasi, dan keberlanjutan. Pembelajaran dirancang adaptif dengan penyesuaian materi, metode, dan evaluasi sesuai karakteristik dan kebutuhan individual siswa. Hasil menunjukkan seluruh siswa (100%) berhasil melaksanakan prosedur pemijahan induksi dengan tingkat keberhasilan pemijahan dan kelangsungan hidup larva yang tinggi. Peningkatan kompetensi signifikan tercermin dari kenaikan skor rata-rata dari 50% (pre-test) menjadi 87,2% (post-test), dengan N-gain 0,71 (kategori tinggi). Pembelajaran adaptif terbukti meningkatkan motivasi, partisipasi aktif, keterampilan kolaborasi, dan problem solving siswa. Program ini berkontribusi nyata dalam memperkuat relevansi SMK perikanan dengan kebutuhan industri dan membuka peluang replikasi di sekolah kejuruan lainnya

Abstract

Freshwater fish populations are highly sensitive to habitat variation, yet information on how habitat characteristics shape fish diversity in Central Java rivers remains limited. The Ijo River is an important natural system for evaluating ecological factors that determine fish communities. This study aims to assess the physical and chemical characteristics of the Ijo River and evaluate their influence on fish diversity. Research was conducted using survey methods and purposive random sampling at nine stations across three zones: upstream, middle, and downstream. The results obtained are that the environmental parameters varied across river zones. The downstream zone recorded the highest temperature (29.14°C), greatest depth (2.54 m), widest channel (32.97 m), and highest free CO₂ (12.64 ppm). The upstream zone showed the greatest light penetration (74.43 cm), fastest Current Velocity (0.75 m/s), highest dissolved oxygen (5.28 ppm), and highest pH (8.31). Fish diversity also differed between zones. The downstream zone had the highest abundance (4,885 individuals) and greatest dominance (0.17), whereas the upstream zone exhibited the highest Shannon-Wiener diversity index (2.46) and evenness (0.36). These results indicate that upstream zones, with lower temperatures, faster currents, higher dissolved oxygen, and stable pH, support greater species diversity and evenness. Downstream zones, with higher temperatures, elevated free CO₂, and wider river width, show greater abundance but are dominated by a few adaptive species. This pattern, consistent with other rivers, confirms that physical and chemical water characteristics shape distribution, abundance, and dominance of freshwater fish. Thus, fish diversity is a key indicator of river ecosystem health.
