Abstract

Expression of Mycobacterium tuberculosis Ag85B Antigen in Mammalian Cell CultureTuberculosis (TB) continues to be a major health problem worldwide, affecting millions of people each year. The only vaccine approved for the prevention of TB is Bacillus Calmette-Guérin (BCG). However, one of the limitations of BCG is that its preventive effect against pulmonary TB varies from person to person. Therefore, there arises a need for a new TB vaccine to replace BCG. This study aims to obtain the Ag85B recombinant protein which has characteristics similar to the native Ag85B antigen from Mycobacterium tuberculosis. In this study, we cloned and expressed recombinant Ag85B in mammalian cell culture. In the initial step, we cloned synthetic Ag85B into mammalian expression vector pFLAG-CMV4 and expressed the gene in CHO-K1 cells. Interestingly, a specific band around 30 kDa was observed in the culture media of transfected cells by Western blot analysis. The results from our research showed the potency of mammalian expression system to produce recombinant protein Ag85B for new TB vaccine candidate.Keywords: Ag85B, mammalian cells, tuberculosis, vaccine, expression ABSTRAKTuberkulosis (TB) terus menjadi salah satu masalah kesehatan dunia yang mempengaruhi jutaan manusia setiap tahun. Satu-satunya vaksin untuk TB yang ada adalah Bacillus Calmette-Guérin (BCG). Namun demikian, vaksin BCG ini memiliki kelemahan berupa terjadi efek preventif yang bervariasi dari satu individu terhadap individu lainnya.  Oleh sebab itu diperlukan pengembangan vaksin TB yang dapat menggantikan vaksin BCG yang sudah ada. Penelitian ini bertujuan memperoleh protein rekombinan Ag85B yang memiliki karakteristik mirip dengan antigen Ag85B native dari Mycobacterium tuberculosis. Pada penelitian ini, telah dilakukan kegiatan pengklonaan dan ekspresi gen Ag85B pada galur sel mamalia.  Pada tahap awal dilakukan pengklonaan gen sintesis Ag85B ke dalam plasmid pada sel mamalia pFLAG-CMV4 dan diekspresikan gennya pada sel CHO-K1. Hasil analisis Western blot menunjukan tersekresinya gen target berukuran 30 kDa pada media kultur dari sel mamalia yang ditransfeksi. Hasil dari penelitian ini menunjukkan potensi dari sistem ekpresi untuk protein rekombinan Ag85B pada galur sel mamalia sebagai kandidat vaksin TB yang baru.

Abstract

The rapid population growth in Central Lampung Regency has increased the demand for equitable access to educational facilities. This study aims to analyze the spatial distribution pattern of educational facilities to determine whether their placement is clustered, random, or dispersed. The analysis uses the Average Nearest Neighbor (ANN) method, a spatial analysis technique that measures the average distance between each facility and its nearest neighbor to assess spatial patterns. A total of 397 educational facilities, including junior high schools, senior high schools, and Islamic boarding schools, were analyzed using shapefile data obtained from the official geoportal of the Republic of Indonesia. The ANN results show an index value of 0.520875 with a z-score of -17.1969, indicating that educational facilities are clustered in specific regions. This clustering can lead to accessibility challenges for populations in underserved or rural areas. Therefore, it is recommended that the local government and the Central Lampung Education Office prioritize the development of new educational facilities in sparsely served districts to ensure more balanced spatial access to education services.

Abstract

Indicators of sustainable development include equal access to education. However, when administrative region and population density are taken into account, educational resources are still unevenly distributed throughout Indonesia's numerous provinces. Lampung was chosen as the research location due to its rapid population growth over the last two decades, which has influenced demand for educational services and the distribution of other public resources. Lampung has 9.52 million people and covers 33,570 km². The importance of this study stems from the necessity to understand the geographical link between these variables in order to facilitate spatial data-based regional planning that is more equitable and efficient in terms of educational distribution. This study used statistical analysis, namely Pearson correlation analysis, to establish the degree of relationship between variables. The study discovered that the area has a Pearson correlation coefficient of 0.674 on the number of educational institutions. The Pearson correlation between population density and the number of educational facilities is -0.139, indicating a very weak relationship. The multiple correlation calculation of the relationship between area and population density with educational facilities provides a strong result of 0.745, suggesting that the larger the area in a region and the denser the population, the more educational facilities are required. Lampung's major districts are Way Kanan, Tulang Bawang, and Pesisir Barat, which rank third, fourth, and sixth respectively. However, in terms of educational facilities, Way Kanan, Tulang Bawang, and Pesisir Barat rank sixth, ninth, and fourteenth, respectively. Lampung province's population facilities and administrative areas remain significantly different.
