Abstract

The use of biocontrol agent in aquaculture is being adapted as an effective alternative to antibiotics which can lead to the elaboration of antibiotic-resistant bacteria and confer unpleasant impacts to aquatic organisms. Aquatic bacteria have been discovered as biocontrol agents and potential probiotic candidates to improve the health of aquatic organisms, feed efficiency, and disease resistance to aquaculture pathogens. However, local isolate has not intensively been explored and used to increase aquaculture sector productivity. Therefore, this research aimed to determine minimum inhibitory concentrations of their antibacterial compounds against aquaculture pathogens and to characterize aquatic bacteria by their viability in the feed. Four isolates from several aquatic environments in Indonesia (Pseudomonas sp. S1.1, Pseudomonas sp. S1.2, Pseudomonas sp. SL1.1, and Bacillus subtilis KM16) were used to characterize of antibacterial compound and to determine the viability in feed. Ethyl acetate extracts from all isolates showed better antibacaterial activity against Aeromonas hydrophila and Vibrio vulnificus than chloroform and dichloromethane extracts, in which ethyl acetate extract from Bacillus subtilis KM16 showed the strongest antibacterial activity. Pseudomonas spp. were more effective against V. vulnificus (40 mg/mL) and Bacillus subtilis KM16 was more effective against A. hydrophila (20 mg/mL), as proved by the minimum inhibitory concentrations of their ethyl acetate extracts. In this research, Bacillus subtilis KM16 had stable viability in feed than Pseudomonas sp. isolates.

Abstract

Bacteria can interact with each or other microorganisms by releasing, sensing, and reacting to small diffusible chemical signal molecules to alter their community behavior. This process, termed quorum sensing, is influenced by density of other bacteria that present in the environment. One example as a result of this process is the formation of biofilm. Biofilm consists of bacterial communities that attach to a surface and envelope themselves in secreted polymers. This formation can be beneficial to pathogenic bacteria because they become highly resistant to antibiotics and human immunity. Thus, antibiofilm agents that can inhibit biofilm formation are needed. The objective of this study were to screen and evaluate bacteria from hot spring and crater lakes that have antibiofilm activity against pathogenic bacteria. In this study, 26 isolates were successfully obtained and tested for quorum sensing and quorum quenching activities. Based on the result, two isolates, which were KM16 and PAP26, were found to have quorum quenching activity. Further research showed that KM16 and PAP26 had antibiofilm activity against more than six pathogenic bacteria. From characterization of the bioactive compounds, it is known that different compound from KM16 and PAP26 have different activity against each pathogen. In molecular identification, isolates KM16 and PAP26 were identified as Bacillus subtilis and Pseudomonas sp. through molecular identification.

Abstract

Aquaculture is currently experiencing massive loss due to the outbreak of pathogenic bacteria. One of the outbreak causes is the development of pathogenic bacterial resistance to the antibacterial. The problem can be solved using microorganisms that can produce new antibacterial compounds. The purpose of this research was to obtain bacteria from fishpond water that could produce antibacterial compounds. About two out of 81 isolates could produce antibacterial compounds. Those two isolates were obtained from saltwater fishponds in North Jakarta (TS2) and Harapan Island (PHY). All fishpond water was grown in marine broth or Luria broth. Extraction of antibacterial compounds was performed using four types of solvents: chloroform, dichloromethane, ethyl acetate, and methanol. Each of the solvents showed a different result. The extraction can only be successfully performed using chloroform and dichloromethane. Extraction using dichloromethane showed a larger inhibitory clear zone than chloroform. Based on 16S rRNA gene sequencing, PHY isolate was identified as Bacillus sp. and TS2 as Acinetobacter sp. In conclusion, isolate TS2 and PHY, which produced antibacterial compounds, showed potential use as aquaculture probiotics.

Abstract

Tempe bungkus daun jati merupakan produk pangan fermentasi tradisional Indonesia yang kaya protein, serat, dan vitamin yang sangat popular di Kabupaten Blora, Jawa Tengah. Penelitian ini bertujuan mengetahui pengaruh penambahan daun kelor pada tahapan perendaman dan peragian dalam proses produksi tempe terhadap profil bakteri asam laktat (BAL) dan tingkat kesukaan. Daun kelor segar dan serbuk disuplementasikan dengan variasi konsentrasi pada proses produksi tempe kacang kedelai. Hasil penelitian menunjukkan bahwa daun kelor tidak menghambat aktivitas pertumbuhan dari kapang Rhizopus. Penambahan serbuk daun kelor sebanyak 1x dan 2x (b/b) dari total jumlah ragi pada proses peragian menghasilkan tekstur tempe yang kompak. Semakin tinggi konsentrasi air perasan daun kelor dalam proses perendaman, jumlah total bakteri asam laktat menurun hingga 103 CFU/g. Profil BAL dari tempe bungkus daun jati dengan ataupun tanpa suplementasi daun jati mengandung BAL dari genus Weissella. Tempe bungkus daun jati dengan suplementasi 1x (b/b) serbuk daun kelor sangat berpotensi untuk disosialisasikan sebagai pangan fungsional kepada masyarakat.Lactic Acid Bacteria Profile and Sensory Evaluation of Teak Leaf-Wrapped Tempeh Supplemented with Moringa LeavesAbstractTempeh wrapped in teak leaves is a traditional Indonesian fermented food product that is rich in protein, fiber, and vitamins which is very popular in Kabupaten Blora, Central Java. This study aimed to investigate the effect of Moringa leaves addition in the soaking and fermentation stages of tempeh production towards the profile of lactic acid bacteria (LAB) and the level of respondent acceptance. Fresh and powdered leaf was supplemented in various doses to soybean tempeh production. As results, Moringa leaves did not inhibit the growth of Rhizopus. The addition of Moringa leaf powder as much as 1x and 2x (w/w) of the total amount of yeast in the fermentation process produced a compact tempeh texture. The higher the concentration of Moringa leaf juice in the soaking process reduced total number of lactic acid bacteria to 103 CFU/g. LAB profile of teak leaf-wrapped tempeh with or without teak leaf powder supplementation contained LAB that belongs to the genus Weissella. Teak leaf-wrapped tempeh supplemented by 1x (w/w) Moringa leaf powder has a potential value to be socialized as functional food to the community.
