Abstract

 ABSTRACT Passive smoking has higher health risk than active smoking like coronary heart diseases, lung cancer and mortality level. Nicotine is main contain in cigarettes smokes that caused addictive. The nicotine level of the side stream smoke from Indonesia non-filter cigarettes brand X was analyzed by Gas chromatography-Mass spectrophotometer and Cundiff-Markunas methods for the level of nicotine employing modified smoking standard, namely two puff/min and then were trapping in HCl 0.1 N. The total alkaloid result from 13.745 g burned tobacco (10 cigarettes) was 0.130 g. The nicotine as total alkaloids levels with Cundiff-Markunas methods was 66.860% and the nicotine level with gas chromatography-mass spectrophotometer was 10.05%. Keywords: nicotine, cigarettes smokes, passive cigarettes smokes.

Abstract

Ekstrak kulit manggis telah banyak digunakan sebagai obat herbal untuk pengobatan berbagai penyakit dan pemeliharaankesehatan. Senyawa kimia utama dari ekstrak kulit manggis adalah α-mangostin. Bioaktivitas senyawa alfa mangostin telah diuji secara ekstensif pada berbagai organisme in vitro. Namun, informasi tentang efek keamanannya belum diketahui. Penelitian ini bertujuan untuk meninjau keamanan alfamangostin dengan mengevaluasi aktivitas sitotoksik dari alfa mangostin pada kultur sel leukosit normal dan aktivitas antioksidan dari α-mangostin. Evaluasi sitotoksik dilakukan dengan 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT assay)dan aktivitas antioksidan dilakukan menggunakan1,1-difenil-2-pikrilhidrazil (DPPH assay). Konsentrasi alpha mangostin yang diuji yaitu: 3.125 μg/mL; 6,25 μg/mL; 12,5 μg/mL; 25 μg/mL; 50 μg/mL dan 100 μg/mL. Hasilnya menunjukkan bahwa alfa mangostintidak memiliki efek toksik padakulturselleukosit. Alfamangostin mampu memicu respon proliferasi sel leukosit pada 50 μg/mL dengan viabilitas sel 208,485 ± 21,21% dan pada 100 μg/mL dengan viabilitas sel 361.818 ± 86,37%. Alpha mangostinjuga menunjukkan aktivitas antioksidan yang kuat dengan nilai IC5013,57 μg/mL. Hasil penelitian ini membuktikan bahwa alfa mangostin tidak toksikpada sel leukosit manusia sehingga aman untuk dikonsumsi. Sebagai senyawa antioksidan alfamangostin dapat memicu proliferasi sel leukosit dalam kultur sel leukosit.

Abstract

Mesenchymal Stem Cells (MSC) are cells that are able to differentiate into other cells and have immunomodulatory properties that are used to treat inflammatory diseases. MSCs have the ability to repair damaged tissue by regenerating damaged tissue. MSCs produce a secretome, where the MSC secretome has various cytokines, chemokines, growth factors, anti-inflammatory factors and proteins that are produced in molecular form. One of the molecules secreted by the secretome is Interleukin-10.IL-10 is an anti-inflammatory cytokine which functions to inhibit the production of several other types of cytokines such as TNF, IL-1, chemokine and IL-12). Apart from that, IL-10 is able to inhibit the function of macrophages in assisting T cell activity. One of the products from Secretome MSC is a cream preparation. To find out whether the MSC cream contains IL-10 protein, Interleukin-10 levels were analyzed in the MSC secretome cream with 3 different MSC cream concentration formulations (2%, 3% and 5%). Analysis of IL-10 levels was carried out using ELISA to detect IL-10 antigen. The results of the examination of 9 samples showed that all samples were positive for IL-10 with a concentration of 2% dilution 1:2 showing the best results with an Optical Density (OD) of 0.069 and followed by a concentration of 5% dilution 1:2 showing an OD value of 0.065.

Abstract

Acne vulgaris is a chronic inflammatory skin condition influenced by excess sebum production, bacterial colonization, and host immune responses. Microorganisms, particularly Cutibacterium acnes and Staphylococcus aureus, play a key role in acne pathogenesis and contribute to persistent inflammation. With increasing antibiotic resistance, innovative therapeutic approaches that are effective, safe, and immunomodulatory are urgently required. This literature review aims to evaluate the role of adipose-derived mesenchymal stem cell (ADMSC) secretome in inhibiting acne-causing bacteria. The review was conducted using databases including ResearchGate, PubMed, Google Scholar, and ScienceDirect, using keywords such as secretome, MSC secretome, antibacterial, C. acnes, S. aureus, and acne vulgaris. A total of 13 relevant research articles were identified. The ADMSC secretome contains antimicrobial peptides (e.g., LL-37), cytokines, growth factors, and extracellular vesicles that can disrupt bacterial membranes, enhance phagocytosis, and inhibit biofilm formation. Several studies demonstrate strong inhibitory effects against S. aureus, including resistant strains, while data on C. acnes remains limited but still supports its therapeutic relevance.
