Abstract

Sinar ultraviolet memicu terbentuknya radikal bebas penyebab kerutan. Daun binahong (Anredera cordifolia) memiliki aktivitas antioksidan yang dapat menstabilkan radikal bebas. Penggunaan ekstrak daun binahong dalam bentuk gel lebih praktis dan nyaman dikulit. Penelitian ini bertujuan untuk mengetahui aktivitas anti-wrinkle pemberian gel ekstrak binahong pada mencit yang diinduksi sinar UV-B. Penelitian ini menggunakan 25 ekor mencit jantan, dibagi 5 kelompok. Kelompok normal (perlakuan apapun), kelompok UV-B (hanya paparan sinar UV-B), kelompok tretinoin (paparan sinar UV-B dan gel tretinoin), kelompok perlakuan 1 (paparan sinar UV-B dan gel ekstrak binahong satu kali sehari), kelompok perlakuan 2 (paparan sinar UV-B dan gel ekstrak binahong dua kali sehari. Paparan dilakukan dengan lampu UV-B Kernel KN-4003 selama 2 minggu. Pengamatan kerutan secara visual scoring dengan metode Bissett. Data yang diperoleh dianalisis menggunakan uji statistik SPSS. Skor kerutan pada kelompok normal adalah 0, kelompok UV-B 2,66±0,48, kelompok tretinoin 0,44±0,38, kelompok perlakuan 1 1,06±0,61, dan kelompok perlakuan 2 0,80±0,84. Kelompok UV-B memberikan perbedaan yang signifikan terhadap tretinoin, perlakuan 1, dan 2 (P<0,05). Kelompok tretinoin tidak berbeda signifikan terhadap skor kerutan pada perlakuan 1 dan 2 (P>0,05). Pemberian gel ekstrak binahong memiliki aktivitas sebagai anti-wrinkle dalam mencegah terjadinya kerutan pada mencit yang diinduksi sinar UV-B. Penambahan frekuensi memberikan perbedaan yang tidak signifikan. Kata kunci: anti-wrinkle, ekstrak daun binahong, gel

Abstract

Hypertriglyceridemia occurs when the triglyceride levels are increasing from the normal (>150 mg/dL) and it causes pancreatitis at a dose of more than 885 mg/dL. The triglyceride-lowering medicines consumption has several side effects on various organ systems. The plant-based medicine (phytomedicine) alternatives have been observed as the synthetic medicine substitution. Purple-fleshed sweet potato (Ipomoea batatas [L.]) contains anthocyanin that has proven to carry out the antioxidant mechanism and the capability to be used as anti-hypertriglyceridemia. This study aims to identify the activity of purple-fleshed sweet potato extracts on triglyceride levels and the histopathological profile of rats' pancreas-induced Triton X-100. Twenty-five male Wistar rats of week 8th were divided into five groups. The normal (N) group was treated with saline solution, and the treatment (T) groups were a single dose of 100 mg/kg of body weight (BW) Triton X-100 and purple-fleshed sweet potato extracts of 175 (T1); 350 (T2); and 700 (T3) mg/kg BW, while the negative (C-) control group was mere induced by a single-dose Triton X-100 of 100 mg/kg BW. The blood serum was isolated and the pre-and post-test was subsequent conducted. The triglyceride serum levels and histopathological data were analyzed by the Kolmogorov-Smirnov normality test and Kruskal-Wallis post-test statistical analysis. The results showed that Triton X-100-induced treatment could significantly increase the triglyceride levels compared to the normal group (P<0.05). The purple-fleshed sweet potato extracts started to lower the triglyceride level at a dose of 700 mg/kg BW (51.83 ±19.15(%)). The extract doses of 175 and 350 mg/kg BW could not reduce the triglyceride level significantly (3.03±2.77 and 5.63±4.24 (%), respectively). A dose of 700 mg/kg BW did not damage the exocrine gland and Langerhans islet of hyperlipidemia rat pancreas treated by Triton X-100.

Abstract

There are many plants that have the potential to be developed into anti-diabetic herbal medicine. One of them is yellow root (Fibraurea tinctoria Lour.) that contains chemical compounds including alkaloids, flavonoids, tannins, saponins, steroids/triterpenoids. Its known that the administration of ethanolic extract of yellow root at doses of 50, 100 and 200 mg/kg BW can reduce blood glucose levels of rats with induced with alloxan. To increase the use of yellow root in food medicine, preparations were developed by formulating the extract in tablet dosage form. In this study, tablets were made with three formulas with different fillers, namely Avicel PH 101 and pregelatinized starch. The research was conducted experimentally using three formulas with varying concentrations of fillers with a ratio between Avicel PH 101 and and pregelatinized starch, namely formula I (1:0), formula II (0.5:0,5) and formula III (0:1). Furthermore, evaluation of granules which includes evaluation of flow time, angle of repose, compressibility, mass density and moisture content. The granules were compressed and evaluated for tablets including evaluation of weight uniformity, hardness, friability and tablet disintegration time. Based on the results of the study, it can be concluded that the variation of the filler did not affect the physical properties of the yellow root extract granules, but did affect the physical properties of the tablets. The results showed that the tablet that met all the requirements for the physical properties of the yellow root extract tablet was formula II with a combination of Avicel PH 101 and Pregelatinized Starch as fillers.
