Abstract

Tomato (Lycopersicon esculentum L.) is an important horticultural crops but very susceptive in their growth. It is known that the plant growth can be influenced by microorganisms, such as symbiosis between plants and endophytic fungi. Endophytic fungi are fungi that live in the tissues of living plants and will not cause any negative effects on its host. This study aims to determine the growth of tomato plants affected by endophytic fungi isolated from Mangrove root collected from Pulau Dua Sanctuary Serang Banten, both in vitro and in vivo. There were five isolates endophytic fungi of mangrove root (CEM), collection of biology education laboratory UNTIRTA, which is used in this study: CEM 2, CEM 3, CEM 4, CEM 7, and CEM 9. The research conducted by inoculating tomato seedlings on MS medium that has been grown by endophytic fungi for in vitro growth, while the in vivo growth carried out by inoculating tomato seedlings on zeolite medium with corn as a fungi carrier. Morphological observation and plant growth parameters measured after 5 weeks incubation. The result showed that CEM 9 is the potential endophytic fungi from roots of Mangrove Pulau Dua Sanctuary Serang to increase tomato growth.

Abstract

ABSTRAKCallose, β-1,3-glukan, merupakan senyawa alami yang terdapat pada tanaman dan terlibat dalam sistem pertahanan tanaman. Callose merupakan bentuk pertahanan tanaman yang penting karena dapat menjadi penghalang awal masuknya patogen ke dalam tanaman. Deposisi callose termasuk ke dalam sistem pertahanan aktif karena akan terinduksi jika tanaman terserang patogen. Molekul patogen yang dilaporkan dapat menginduksi deposisi callose adalah lipopolisakarida (LPS), komponen utama membran luar bakteri gram negatif. Telah dilakukan penelitian untuk mempelajari induksi deposisi callose pada tanaman tembakau oleh LPS yang diekstraksi dari bakteri Pseudomonas syringae pv. tabaci (Pta). Bakteri Pta merupakan fitopatogen penyebab penyakit wildfire pada tembakau. Untuk pengamatan deposisi callose, daun tembakau diinfiltrasi dengan LPS Pta 400 μg/ml dan 800 μg/ml serta diinkubasi selama 24 jam dan 48 jam. Selanjutnya, klorofil daun diluruhkan menggunakan larutan laktofenol dan daun diwarnai dengan aniline blue. Hasil pengamatan menunjukkan deposisi callose yang telah diwarnai dengan aniline blue tampak berpendar warna hijau ketika diamati menggunakan mikroskop fluoresensi. Intensitas deposisi callose, hasil induksi LPS bakteri Pta, dalam satu daun tampak tidak merata di semua bagian daun, namun dan cenderung terdeposisi di sekitar pertulangan daun.

Abstract

This research aimed to identify the Zingiberaceae diversity in Pandeglang which can be used as learning content for biodiversity concept in Pandeglang senior high school. The method was part of a research and development. Based on exploration, there were seven species of Zingiberaceae diversity that was utilized in Pandeglang, namely Zingiber officinale, Alpinia galanga, Curcuma domestica, Curcuma aeruginosa, Kaempferia pandurata, Kaempferia galanga, Zingiber zerumbet. The contents were developed for genetic-level biodiversity, species-level biodiversity, and ecosystem-level biodiversity learning concept.

Abstract

Stevia (Stevia rebaudiana Bert. M.) is known as a natural non-caloric sweetener. This plants contain glycoside such steviosida type, mainly on the leave contain sweetness level between 200-300 cane sugar but the calorie is very low. This research was aimed to determine difference effects of PEG (Polyethylene Glycol) concentrations and the light conditions on Stevia (Stevia rebaudiana Bert. M.) callus induction in vitro. This research was conducted from April to June 2016 at Biotechnology Laboratory Faculty of Agriculture, Sultan Ageng Tirtayasa University. This research used a completely randomized design (CRD), which consisted of two factors with three replications. Concentrations of PEG as first factor consisted of four levels (0 mg/L, 5 mg/L, 15 mg/L, and 25 mg/L). Light conditions as second factor consisted of two levels (Dark and Light). The results showed that the concentrations of PEG (Polyethylene Glycol) did not significantly effect the time of callus appearance and diameter of callus on 4, 5 and 6 weeks after planting. Dark condition was the best conditions for callus induction of stevia. The texture of callus was compact on all treatments and the callus dominant color produced is golden brown.

Abstract

This research aimed to develop a learning module on biotechnology based on Sains Technology Society (STM) and to know the module quality according to the expert assessments. This research was a developments research (R & D) with 3-D (Define, Design, and Develop) model. The instruments in this research were questionaire and expert assessment sheets. Module was assessed from the feasibility aspects of content, sains technology society (STM), language, presentation and layout. The data from questionaires and assessment sheets was used to improve the module contents and structures. The result of this research was a learning module based on the sains technology society (STM) on biotechnology. The result assessment according to the expert showed that the modul was 93,8% with category very good and feasible to use as learning module for 12th grade of senior high school.

Abstract

This research aims to develop practicum worksheet based on Project Based Learning as mean as to support the entrepreneurial spirit of students and to determine the feasibility of the media through expert tests and user response tests. This research was conducted in October 2021-January 2022. The method used in this research was Research and Development (R&D) with a 3D model which consist of define, design and develop. The data collection used at the define stage were questionnaire analysis of the early-late analysis, student analysis, task analysis, concept analysis, and specification of learning objectives. The data collection obtained in the design stage were selecting the media, selecting the format and planning the first product design in form of storyboards, then data collection carried out at the develop stage is media feasibility assessment questionnaire by experts in terms of material and media. The results of the product assessment by material experts obtained an average value of 96,9%, while by media experts it was 97,4%. The results of the user response test obtained an average value of 85.2%. Based on the result of product assessment by experts and user respond test showed that the hydroponic practicum worksheet based on Project Based Learning obtained a very feasible category to be used as a practical guided that could support the entrepreneurial spirit of students

Abstract

This study aims to Develop Bio-Meta which integrates digital books with augmented reality and virtual reality in visualizing the concept of plant growth and Development, to Develop Bio-Meta as an interactive digital learning medium that can improve students' digital literacy, and to determine the feasibility of using it. Bio-Meta: Integrated Digital book With Augmented reality And Virtual reality from experts and samples. This research is a Development research (R&D). This research was conducted from January to September 2022 at Sultan Ageng Tirtayasa University and SMAN 2 Serang City. The research sample consisted of 20 students of class XII science. The process of Developing learning media is carried out with the stages of Analyze, Design, Develop, Implement and Evaluate (ADDIE). The applications used are Canva, Sketchfab.com, 3DVista, WebAR Zappar, and Heyzine Flipbooks. The results of the expert test analysis showed that the average score of the media expert team was 88.3% and the material expert team was 84.2% with the very good media category. The results of the student response test show that the media feasibility interpretation score from the use of the SUS instrument is 81.3 so that it meets the criteria for an interpretation score of more than 80.3 which means that Bio-Meta is a good media and students accept the Bio-Meta learning media as a suitable media to use. to help students understand the nervous system material.
