Abstract

Serangan cendawan Thielaviopsis paradoxa dapat menurunkan kualitas buah salak selama penyimpanan. Salah satu cara penanganan pascapanen untuk mempertahankan kualitas buah salak pondoh ialah dengan penambahan bahan tambahan pangan kalsium klorida dan bahan pelapis kitosan. Penelitian ini bertujuan menguji pengaruh CaCl2 dan kitosan untuk mengendalikan T. paradoxa, penyebab busuk hitam pada buah salak pondoh. Perlakuan terdiri atas CaCl2 6%, kitosan 1.5%, CaCl2 6% + kitosan 1.5%, dan kontrol. Hasil penelitian menunjukkan bahwa kitosan 1.5% lebih efektif dalam mempertahankan kualitas buah salak selama 12 hari penyimpanan dibandingkan dengan perlakuan lainnya. Perlakuan ini menghambat peningkatan populasi mikrob, persentase kerusakan daging buah, persentase susut bobot, dan penurunan nilai organoleptik dibandingkan dengan kontrol pada suhu ±28 °C dan kelembapan relatif 65%–75%. Kitosan 1.5% dapat direkomendasikan kepada petani salak untuk memperpanjang masa simpan buah salak sampai dengan 12 hari.

Abstract

ABSTRACTAntrachnose on chilli fruit caused by Colletotrichum capsici can reduce yield and quality of chilli fruit. Biological control agent can be used as an alternative to control C. capsici. Yeast is one of biological control agent which is potential to control the pathogen. This study was aimed at testing antagonistic potential of yeast on fruits and vegetables against C. capsici. Twenty two yeast isolates were isolated from banana, rambutan, red chilli, tomato, and eggplant fruits. Screening for antagonistic yeast using well test showed that 5 isolates of yeast (CMM-1, CMM-3, CMM-4, TMM-1, and EMM-11) completely inhibited the growth of C. capsici. Based on the result of biocontrol assay of the pathogen in vivo, four yeast isolates (CMM-3, CMM-4, TMM-1, and EMM-11) completely inhibited C. capsici in vivo. Identification using morphological and molecular characteristics showed that these four yeast isolates were Issatchenkia orientalis.Keywords: antagonistic yeast, antrachnose, biocontrol, Issatchenkia orientalis ABSTRAK Antraknosa pada buah cabai yang disebabkan oleh Colletotrichum capsici dapat menyebabkan penurunan produksi dan kualitas buah cabai. Penggunaan agens pengendalian hayati dapat menjadi salah satu alternatif untuk mengendalikan C. capsici. Khamir merupakan salah satu agens pengendalian hayati yang berpotensi mengendalikan C. capsici. Penelitian ini bertujuan menguji potensi antagonistik khamir pada buah-buahan dan sayuran terhadap C. capsici. Sebanyak 22 isolat khamir diisolasi dari buah rambutan, pisang, cabai merah besar, tomat, dan terung ungu. Seleksi khamir antagonis menggunakan uji sumur diperoleh sebanyak 5 isolat khamir, yaitu isolat CMM-1, CMM-3, CMM-4, TMM-1, dan EMM-11 menghambat total pertumbuhan C. capsici. Isolat CMM-3, CMM-4, TMM-1, dan EMM-11 menghambat total pertumbuhan C. capsici in vivo. Berdasarkan hasil identifikasi secara morfologi dan molekuler, isolat CMM-3, CMM-4, TMM-1, dan EMM-11 adalah Issatchenkia orientalis.Kata kunci: antraknosa, Issatchenkia orientalis, khamir antagonis, pengendalian hayati

Abstract

Abstract  Oil palm empty fruit bunches (OPEFB) are the ligno-cellulosic wastes from palm oil processing. They can be used to produce raw materials for value-added products. The purpose of this study was to determine the degradation capacity of JPA wood rot fungi and Trichoderma sp. S2-2 on OPEFB. The 500 g of substrates consisted of 81% of OPEFB, 15% bran, 1.5% lime and 1.5% gypsum were used for growing. The substrates were inoculated with five treatments i.e without isolate (K); with JPA isolate (JPA); with Trichoderma sp. S2-2 (T); with the two isolates (JPA + T); and with JPA isolate and after four weeks of incubation inoculated with Trichoderma sp. S2-2 [(JPA)+T]. All treatments were incubated for eight weeks. The results showed that JPA+T was the best treatment which the two isolates must be inoculated simultaneously for degradation of OPEFB. Lignin and cellulose content on JPA+T treatment respectively were 20.83% and 33.77%. C/N ratio of OPEFB degraded with JPA+T was lower than the C/N ratio of TKKS degraded with Trichoderma harzianum and TKKS degraded with EM4 in previous study. AbstrakTandan kosong kelapa sawit (TKKS) merupakan limbah lignoselulosa dari pengolahan minyak kelapa sawit. TKKS dapat dimanfaatkan untuk memperoleh bahan baku untuk produk bernilai tambah. Tujuan penelitian ini adalah untuk mengetahui kemampuan degradasi jamur pelapuk kayu isolat JPA dan Trichoderma sp. S2-2 pada TKKS. Sebanyak 500 g substrat terdiri dari 81% TKKS, 15% dedak, 1,5% kapur, dan 1,5% gypsum digunakan untuk per-tumbuhan. Substrat diinokulasi dengan lima perlakuan yaitu tanpa isolat (K); dengan isolat JPA (JPA); dengan Trichoderma sp. S2-2 (T); dengan isolat JPA dan setelah empat minggu inkubasi, diinokulasi dengan Trichoderma sp. S2-2 [(JPA+T)]. Semua perlakuan diinkubasi selama delapan minggu. Hasil percobaan menunjukkan bahwa perlakuan JPA+T adalah perlakuan terbaik yaitu kedua isolat tesebut harus diinokulasi secara bersamaan untuk mendegradasi TKKS. Kandungan lignin dan selulosa TKKS dengan  perlakuan  JPA+T   masing-masing  adalah  20,83% dan 33,77%. Rasio C/N TKKS hasil degradasi dengan JPA+T lebih  rendah  daripada  rasio C/N pada TKKS yang didegradasi dengan Trichoderma harzianum dan TKKS yang didegradasi dengan EM4 pada penelitian sebelumnya.

Abstract

The objectives of this study were to get informations on the population of A. flavus and aflatoxin B1 content of five processed peanut products collected from retailers in Kecamatan Bogor Tengah (Subdistrict of Central Bogor), Municipality of Bogor. A total of 129 samples of processed peanut products was collected. They consisted of roasted peanuts with skin pods (33 samples), flour-coated peanuts (33), siomay sauce (18), pecel/gado-gado sauce (33) and satai sauce (12). A sample each of 2 000 g roasted peanuts with skin pods as well as flour-coated peanuts, and a sample each of 1 500 g siomay sauce, pecel/gado-gado sauce as well as satai sauce was mixed homogenously. It was then divided two times manually to obtain working samples to determine A. flavus population, AFB1 content and a reserve sample. Peanut kernels of roasted peanuts with skin pods and flour-coated peanuts were obtained by peeling their skin pods and the batter coat of tapioca flour manually, respectively. Aspergillus flavus in peanut processed products was isolated using a serial dilution method, followed by pour plate method on Aspergillus Flavus and Parasiticus Agar (AFPA). AFB1 content was determined using Thin Layer Chromatography method. Two replicates were used for each sample. The results showed that the population of A. flavus in roasted peanuts with skin pods, flour-coated peanuts, siomay sauce, pecel/gado-gado sauce and satai sauce were 0.3, 0.1, 0.3, 13.2 and 0.4 cfu/g (wet basis), respectively. The highest AFB1 content of  processed peanut products (43.2 ppb) was found in roasted peanuts with skin pods, followed by flour-coated peanuts (34.3 ppb), satai sauce (23.2 ppb), pecel/gado-gado sauce (17.1 ppb) and siomay sauce (4.4 ppb).

Abstract

The objectives of this study were (a) to investigate the effect of postharvest handling (threshing and storage) methods on the quality of sorghum (Sorghum bicolor (L) Moench) grains variety Numbu, in terms of the percentages of damaged grains and seed germination, population growth of S. zeamais, F. proliferatum and F. verticillioides; fumonisin B1 and carbohydrate contents, and the percentage of weight loss during storage. The change of moisture contents of sorghum grains was also recorded. Threshing was conducted using a stick of wood and a paddy thresher. Sorghum grains were packed in hermetic plastic bags. The conditions inside of the bags were  airtight and normal conditions. Each bag with different conditions inside of the bag was introduced with 10 pairs of Sitophilus zeamais (1-14 days old). Sorghum was stored for one, two and three months of storage under warehouse conditions. The results showed, that during storage the moisture contents of sorghum were  lower than that its standard safe moisture content (
