Abstract

Breeding plan of cajuputi in Indonesia is aimed to increase plantation productivity of oil yield and 1.8 cineole content. Seed orchard of cajuputi at Paliyan, Gunungkidul, established using selected and genetically improved materials, has been producing seeds for operational plantation. This seed orchard would perform optimally if the mating systems of all individuals contribute to the inheritance of all genetic potential of the offsprings. Therefore, investigation of the mating systems of cajuputi was indispensible. The study has been carried out on 10 selected mother trees and the 24 offsprings of each mother trees using 8 microsatellite markers of nuclear DNA, namely Hin-2 (100-132 bp), Hin-4 (79-114 bp), Hin-5 (128-148 bp), Hin-7 (136-224 bp), Sal-1 (93-99 bp), Sal-3 (118-219 bp), Xho-1 (96-111 bp) and Xho-4 (150-216 bp), respectively. The result showed relatively high genetic variation of the offspring (HE=0.602, HO=0.594) originated from parent trees in the seed orchard. Parent trees tend to outcross(tm=0.951, ts=0.806), although seeds originated from biparental inbred (tm – ts = 0.145) and correlated paternity(rp=0.098) have also been observed. This genetically viable population could maintain its reproduction fi tness forshort term and adapt to the dynamic environmental changes for long term.Key words: mating system, cajuputi, seed orchard, microsatellite

Abstract

Salah satu aspek penting di kebun benih adalah informasi tentang sinkronisasi pembungaan. Pembungaan yang sinkron mendukung terjadinya perkawinan silang secara acak sehingga perolehan genetik yang dihasilkan  dapat optimal.  Penelitian tentang sinkronisasi  pembungaan  dilakukan  di kebun benih kayuputih di Paliyan, Gunungkidul, berdasarkan fenogram dan indeks overlap selama 2 periode pembungaan (2010-2011). Hasil penelitian menunjukkan lebih dari 75% famili penyusun di kebun benih memiliki pembungaan yang sinkron dengan puncak pembungaan terjadi pada akhir Januari sampai awal Februari. Sebanyak 90% famili memiliki nilai indeks overlap lebih dari 0,8 meskipun terdapat juga 2 famili dari Australia bagian barat dan Australia bagian utara yang memiliki nilai indeks overlap sangat rendah (<0,6). Sebagai implikasinya adalah bahwa benih yang dihasilkan dari kebun benih kayuputih di Paliyan mempunyai potensi genetik yang bagus karena merupakan hasil produksi dari 75% pohon-pohon di kebun benih Paliyan.

Abstract

Controlled pollination technique could be applied to generate gene recombinant from families in progeny test and to estimate genetic parameters. The early step in evaluation of controlled pollination is to estimate general combining ability and specific combining ability. the objectives of this research were to estimate generic parameters on full sib progeny test of melaleuca cajuputi subsp cajuputi cajuput and to identify gene action controlling growth and oil traits. The research was conducted on full sib progeny test at Gunungkidul established in an incomplete block Design with six treeplots and eight replications. Tree height and diameter stem growth were observed at seven years old. Oil yield and cineol content were examined using gas chromatography. Analysis of variance and genetic parameter were calculated for all measured variables. The result showed that individual heretabilities on growth traits (hi2 of stem diameter = 0,34 and tree height = 0,01, respectively) were lower than on oil traiths (hi2 of oil yield =0,6 and cineole = 0,4 respectively). This indicated that oil traits tend to genetically controlled. Analysis of variances on general combining ability (GCA) and Specific Combining Ability (SCA) effects from a serries of 15 sets of 5x5 half-diallel mating experiments showed that all traits (height, diameter, oil and 1,8 cineole yields) were controlled by non-aditive genes. Consequently, controlled pollination of selected plus trees should be carried out until heterosis of improved progenies could be found.

Abstract

Both wood and non-wood industries generate large amounts of residues from processes like bark peeling and removal. In the cajuput oil industry, the waste products include distillation residues and cajuput bark. The diverse extractives and chemical components found in cajuput bark have the potential for enhanced value through various applications. This study aimed to determine the chemical components, extractive content (EC), and percentage inhibition of antioxidant activity (%IAA) of cajuput bark extract to explore more effective uses of this byproduct. Cajuput bark (Melaleuca cajuputi Powell) was sourced from a 26-year-old cajuput stand in Paliyan, Gunungkidul. The bark powder was extracted using a methanol-water solvent mixture at concentrations of 40% and 80%. Extraction was conducted in water baths at temperatures of 60°C and 100°C for 120 minutes. GC-MS identified several chemical components in the methanol extract of cajuput bark, including sugars (erythritol, D-arabinose, D-lyxose, adonitol, d-galactose, D-glucitol, and L-rhamnose), fatty acids (lactic acid, glycerol, glyceric acid, malic acid, and palmitic acid), and phenolic derivatives (gallic acid and protocatechuic acid). The average extractive content of the methanol extract of cajuput bark was 4.07%, and the percentage inhibition of antioxidant activity was 40.26%.

Abstract

Kebun benih sebagai sumber benih pada tingkatan yang tinggi diharapkan dapat menghasilkan benih dan semai yang secara genetis memilki kualitas yang lebih bagus jika dibandingkan dengan tegakan biasa. Informasi tentang produksi benih dalam suatu kebun benih diperlukan untuk mengevaluasi kinerja kebun benih dan juga untuk menghitung ketersediaan benih dalam mencukupi kebutuhan benih untuk penanaman skala luas. Penelitian ini bertujuan untuk mengestimasi produksi benih kayuputih dari masing-masing provenan, serta korelasi antara produksi benih dengan penampilan morfologi tegakan dari kebun benih semai. Penelitian dilakukan di kebun benih semai kayuputih di Paliyan Gunungkidul. Pengamatan dilakukan terhadap produksi bunga, produksi benih dan ukuran tajuk tanaman (tinggi dan diameter tajuk). Hasil penelitian menunjukkan produksi benih antar provenan berbeda. Individu-individu dalam kebun benih semai dan kebun benih klon menunjukkan kinerja yang spesifik. Pada kebun benih semai provenan dari Masarete, Buru menunjukkan produksi benih paling banyak, demikian juga pada kebun benih klon. Secara umum produksi benih dari kebun benih semai lebih bagus dibandingkan dari kebun benih klon.
