Abstract

Abstrak Tingginya kadar glukosa pada Diabetes Mellitus memicu terjadinya stress oksidatif yang ditandai dengan ketidak seimbangan antara jumlah radikal bebas dan antioksidan. Penelitian sebelumnya ekstrak sirsak dan kelor efektif dalam bentuk tunggalnya mampu menurunkan kadar gula darah yang disertai dengan kemampuan untuk menurunkan dampak stress oksidatif. Penelitian ini dilakukan untuk mengetahui pengaruh kombinasi ekstrak sirsak dan kelor terhap kadar glukosa pada tikus yang diinduksi aloxan. Hewan uji sebanyak 36 ekor yang dibagi menjadi 6 kelompok yaitu: kelompok normal (akuades), kontrol negatif (CMC Na 0,5 %), kontrol positif (glibenclamide dosis 1,26 mg/kgBB), ekstrak sirsak 100 mg/kg BB, ekstrak kelor dosis 200 mg/kg BB dan kombinasi I (75:25), II (50:50)dan III (25:75) dalam persen. Semua Kelompok hewan uji, kecuali kelompok normal diinduksi aloksan dosis 160mg/kgBB pada hari ke-0, dilanjutkan dengan perlakuan hewan uji dari hari ke-1 sampai hari ke-14. Pada hari terakhir hewan uji dikorbankan untuk diambil organ liver dan dilakukan pengecakan kadar stress oksidatif yang meliputi lipid peroksidase dan glutathione.Penggunaan ekstrak daun kelor dan sirsak baik itu tunggal dan kombinasi mampu menurunkan stress oksidatif yang ditandai dengan turunya kadar LPO dan naiknya kadar GSH secara signifikan (p<0,05). Aktivitas optimal pada kombinasi sirsak kelor 50:50.Kombinasi ini memberikan hasil yang lebih bagus dibandingkan bentuk tunggalnya

Abstract

Hyperlipidemia is factor affected to coronary hearth disease, allow with increase of serum level cholesterol and tryglycerida. This research is to know effect combination of ethanolic extract of Curcuma xanthorrizha Roxb with low content of volletile oil and ethanolic extract Gynura procumbens (Lour.) on triglyceride level serum and its hyperlipidemia manifestation atherosclerosis. Forty eight male albino rat devide into 8 groups. Positive group receive gemfibrozil, CMC-Na 0,5 % (negatif) and treatment group combination of temulawak and sambung nyawa 100:0, 75:25, 50:50, 25:75, 0:100. Dose of ethanolic extract temulawak is 225 mg/kg BB and dose of ethanolic exstrac sambung nyawa 150 Methode of this research by give diet lipid and yolk egg for about 30 day on treatment group except normal group after this give sample test for about 30 day. Trygliceride level serum measure on day 0 (baseline), 30 and 60 with Trigliserida FS Diasys®. Hypotrigliceride activity describe into % of change triglyceride level and analyse with Mann-whitney with level of significane 95 %. Histopatology analyze used cardiac aorta. Hypotriglyceride activity combination of extract temulawak and extract sambung nyawa showing no significance difference both combination (p>0,05). Combination of temulawak:sambung nyawa (75:25) give the highest hypotrigliceride activity with decrease on triglyceride level serum for about 56,77 %. Combination both extract effective to avoid atherosclerosis

Abstract

Curcuma domestica and Phyllanthus niruri was two plants which has been shown to have an effect hepatoprotective. This research was conducted to determine mechanism study of hepatoprotective combination of Curcuma domestica and Phyllanthus niruri extract to animals induced by paracetamol. Animals devide into 8 groups in random order of 5 each rats. Normal groups were give aquadest, negative groups (Paracetamol) were give CMC Na 0,5 %, positive goups were give silimarin with dose 100 mg/kg, Curcuma domestica groups with a dose of 100 mg/kg, Phyllanthus niruri groups with 200 mg/kg BW as well as a combination of I-III with Curcuma domestica and Phyllanthus niruri extract comparison of each of 75: 50; 50: 100; and 25: 150 mg/kg in BW. Test preparations are given for 7 days. On day 7, all groups except normal induced with paracetamol single dose2.5 g/kg for peroral 30 minutes after administering the test preparations. Hepatoprotective activity were analyzed using liver histopathology. The results showed that the combination I can inhibited liver necrosis and gives better hepatoprotective activity comparable with single dose with optimal Curcuma domestica and Phyllanthus niruri extract dose of 75 and 50 mg/kg.
