Abstract

Pemanfaatan air bersih yang berasal dari tanah di DKI Jakarta semakin meningkat, akan tetapi sangat disayangkan tidak diikuti dengan upaya pengembalian ke dalam tanah. Peningkatan jumlah penduduk menyebabkan kebutuhan lahan untuk tempat tinggal semakin meningkat sehingga berdampak pada semakin menurunnya lahan terbuka untuk peresapan air hujan. Dampak yang terjadi dari kondisi ini adalah penurunan muka air tanah dan intrusi air laut. Keadaan ini perlu segera diatasi dengan teknologi tepat guna dan aplikatif yaitu instalasi pengolahan limbah air wudhu terintegrasi dengan sistem akuaponik. Selain itu instalasi ini juga dapat dimanfaatkan untuk menampung air hujan. Masjid dan mushola sangat berpotensi untuk menerapkan teknologi instalasi yang terintegrasi dengan akuaponik. Oleh sebab itu masjid dapat berperan sebagai penyangga ekologis bagi lingkungan sekitarnya. Sistem akuaponik dapat menjadi contoh program ketahanan keluarga dari aspek kesehatan khususnya untuk kecukupan zat gizi. Potensi fungsi lain dari masjid adalah sebagai solusi terhadap masalah sosial dan ekonomi terutama dalam menghadapi dampak pandemi Covid-19. Hal yang ditemukan di masyarakat terkait dampak tersebut adalah kemiskinan, kasus kekerasan rumahtangga, perceraian, kriminalitas dan kenakalan remaja. Untuk mengatasi permasalahan ini perlu adanya pemberdayaan masyarakat yang di pelopori dari masjid yang merupakan basis umat. Sejauh ini, peranan masjid sebagai penyangga ekologis, fungsi ketahanan keluarga, solusi bagi permasalahan sosial dan ekonomi masyarakat belum optimal khususnya untuk memberdayakan zakat, infak, sedekah dan wakaf (Ziswaf). Mitra dalam pengabdian masyarakat adalah masjid yang berada di DKI Jakarta yaitu At Tawab di Cakung, Al Hikmah di Cijantung dan Abu Bakar Ash Shiddiq di Pasar Rebo, Al Ikhwan Semanan di Cengkareng dan Istiqomah di Tanah Kusir. Metode yang digunakan adalah program ketahanan keluarga yang terdiri atas penerapan instalasi terintegrasi dengan akuaponik, dan program psikoedukasi termasuk peningkatan ekonomi masyarakat melalui zakat, infak, sedekah dan wakaf, komunikasi dan hukum. Program ini akan dilaksanakan selama 3 minggu. Luaran yang ditargetkan adalah 1 artikel ilmiah, dan 8 video yang terdaftar sebagai HKI.Kata kunci: air bersih, air wudhu, masjid, instalasi, ketahanan keluarga

Abstract

Osteosarcoma is a highly malignant primary tumor on bone that mainly attacks children and young adolescents. Until now, osteosarcoma therapy still combines some high costs and invasive therapy modalities that may give side effects, such as pain and nausea. Our previous studies suggested that non-contact electric field has anti-proliferative effect on breast cancer cells, in vitro and in vivo. In this study, we were interested studying alternating current electric field effects on osteosarcoma cells progression as well as its potential cytotoxic effects. MG-63 human osteosarcoma cells were cultured and treated with 200 kHz for 6 days. Several genes of interest including p53, p21, MDM2, caspase-3, caspase-8, and caspase-9 were analyzed using real-time qPCR method. Apoptotic index was measured using flow-cytometry assay. Apoptosis was observed through p53-independent p21 pathway (p = 0.011). Cells undergoing apoptosis through internal pathways were shown by the increase of caspase-3 (p = 0.015) and caspase-9 (p = 0.001) levels, but not caspase-8 (p = 0.080). This treatment has successfully reduced the number of living osteosarcoma cells by 14.7% (p = 0.000) and increased cell death up to 4.26% (p = 0.055). Apoptotic index was markedly increased to 16% (p = 0.001). 200 kHz non-contact electric field exposure can disrupt osteosarcoma progression through disruption of normal cell cycle via p53-independent p21 pathway and induction of apoptosis.

Abstract

The qPCR method requires an oligonucleotide pair to prime the amplification process. With the variety of qPCR reagent and primer options available, in silico and laboratory experimental validation approach was needed to validate the most suitable primer for prior use. This article aims to provide in silico analysis of actin alpha-2 smooth muscle (Acta2), fibroblast activation protein (Fap), hypoxanthine phosphoribosyltransferase-1 (Hprt1), platelet-derived growth factor subunit B (Pdgfb), phosphoinositide-3-kinase regulatory subunit-1 (Pik3r1), and vascular cell adhesion molecule-1 (Vcam1) qPCR primer with qPCR and electrophoresis validation. The procedure used in this approach was in silico analysis of primer from published articles and newly designed primer. The analysis was done with Primer-BLAST for gene specificity, Primer-Dimer, OligoCalc for hairpin formation, BLAST Nucleotide for identical sequence screening, and Clustal Omega for product length validation. Experimental validation was done using qPCR for optimal annealing temperature, priming ability, and amplificon specificity, and electrophoresis for product length validation. This assessment resulted in in silico and laboratory experimental validation of Acta2, Fap, Hprt1, Pdgfb, Pik3r1, and Vcam1 primer pairs producing suitable amplicon for qPCR using Rattus norvegicus cDNA with SYBR annealing temperature range of 60-65°C with three mM MgCl2. The primer pair can be used for further qPCR analysis under similar conditions and the procedure stated can be used as starting point for qPCR Primer preparation.

Abstract

Background: Cancer therapy based on alternating current electric field exposure, Electro-Capacitive Cancer Therapy (ECCT), has been pre-clinically tested for its effectiveness in breast tumor inhibition. However, concerns regarding the safety of electric field (EF) exposure to vital organs have been raised. In the kidney and spleen, IL-10 and TNF-α play roles in the response to inflammation. Objective: This research aimed to evaluate the safety of electric field exposure to vital organs, by assessing the expression of IL-10 and TNF-α mRNA in the kidney and spleen of rats, both with and without breast tumors. Methods: Female rats were divided into four groups; non-induction-non-therapy (NINT), non-induction-therapy (NIT), induction-non-therapy (INT), and induction-therapy (IT). Rats were induced by 7,12-dimethylbenz[a]anthracene (DMBA) at a dosage 20 mg/Kg body weight and exposed to electric fields for 10 hours, followed by a 2-hour rest period. Total RNA from the kidney and spleen was isolated, cDNA was synthesized by reverse transcription PCR, and analysis of IL-10 and TNF-α mRNA expression was performed by RT-qPCR method. Data were analyzed using the Livak formula, GraphPad Prism 9.0 software, and significance test ANOVA (α = 0.05). Results: Alternating current electric field exposure had no effects on the expression of TNF-α and IL-10 mRNA in the kidney and spleen of rats. Specifically, no effect was observed on TNF-α mRNA expression in all groups, and there was no effect on IL-10 mRNA expression in the spleen. Only the INT group showed a significant increase in IL-10 mRNA expression in the kidney (p < 0.05). Conclusion: Exposure of an alternating current electric field did not affect the relative mRNA expression of TNF-α and IL-10 in the kidneys and spleen of rats.
