Abstract

This research was conducted to prove that the leaves of avicennia marina contain ingredients that are potential as nutraceuticals, as well as to determine the comparison of the extraction results using water extract and ethanol extract. Avicennia marina leaves collected randomly and turn it into powder. The avicennia marina leaves powder processed with water extraction and ethanol extraction. The result of extraction process were tested by phytochemical screening tests to determine the content of flavonoids, alkaloids, tannins, steroids, triterpenoids, saponins, and examined for flavonoid levels by thin layer chromatography (TLC). The results of phytochemical screening test for ethanol extract of avicennia marina leaves showed positive results for the content of flavonoids, alkaloids, tannins, steroids, and saponins, but showed negative results for triterpenoid content. The water extract of avicennia marina leaves showed positive results for the content of flavonoids, alkaloids, tannins, and steroids, but showed negative results for saponins and triterpenoids content. At thin layer chromatography test, the flavonoid content showed that ethanol extract of avicennia marina leaves had 2.46%, while water extract of avicennia marina leaves had .80% of flavonoid content percentage. The results of ethanol extraction and water extraction from the leaves of avicennia marina show that they contain phytochemicals which are beneficial for health, and have the potential as a nutraceutical for medicinal purposes.

Abstract

Latar belakang Penyembuhan luka dapat mengalami gangguan sehingga perlu pengobatan. Sel fibroblas adalah salah satu faktor penting pada proses penyembuhan luka. Penggunakan growth factor efektif meningkatkan fibroblas tetapi memiliki efek samping, dan sulit diperoleh. Ekstrak daun mangrove avicennia marina dan kombinasinya dengan ascorbic acid memiliki potensi meningkatkan jumlah sel fibroblas. Tujuan Untuk mengetahui biokompatibilitas dan dampak ekstrak daun mangrove avicennia marina dan kombinasinya dengan ascorbic acid terhadap proliferasi sel fibroblas. Metode Uji MTT Assay pada kultur sel yang diberi perlakuan dan dibagi menjadi 7 kelompok yaitu kelompok Ascorbic Acid 200μg/mL (P1), Ekstrak Etanol Daun Mangrove 100μg/mL (P2), Ekstrak Etanol Daun Mangrove 200μg/mL (P3), Ekstrak Air Daun Mangrove 100μg/mL (P4), Ekstrak Air Daun Mangrove 200μg/mL (P5), Kombinasi Ekstrak Etanol Daun Mangrove 100μg/mL dan Ascorbic Acid 200μg/mL (P6), Kombinasi Ekstrak Air Daun Mangrove 100μg/mL dan Ascorbic Acid 200μg/mL (P7). Kultur sel diperiksa dengan mikroskop, dan dibaca nilai optical density dengan Elisa Reader dan dihitung persentase sel hidup. Hasil Data nilai optical density dianalisa statistik. Seluruh kelompok perlakuan (P1-P7) bersifat biokompatibel (sel hidup ≥ 60%). Semua kelompok perlakuan memiliki persentase sel hidup diatas kontrol sel, dan kelompok P1, P3, P4, P5, P6, dan P7 memiliki perbedaan signifikan (p≤0,05), tetapi kelompok P2 tidak memiliki perbedaan signifikan (p>0,05). Kesimpulan Terapi ekstrak daun mangrove avicennia marina dan kombinasinya dengan ascorbic acid bersifat biokompatibel dan mampu menstimuli proliferasi sel fibroblas.

Abstract

Background: Liver is an organ that performs a role in metabolism and detoxification of chemical substances. The use of immunosuppressive drugs excessively not only become predisposing factor for oral candidiasis, but also can cause damage and affect liver’s function. Hyperbaric oxygen treatment was known to preserve hepatocytes and prevent liver damage. This study was aimed to investigate the effect of hyperbaric oxygen treatment on liver hepatocyte damage in oral candidiasis immunosuppressed rats.Method: We divided fifteen Wistar rats randomly into three groups: G1 (healthy rats), G2 (immunosuppressed rats with oral candidiasis), and G3 (immunosuppressed rats with oral candidiasis given hyperbaric oxygen). The immunosuppressed condition was made by giving dexamethasone and tetracycline orally for 14 days. Induction of Candida albicans 0.1 cc was conducted on the fourth day. Hyperbaric oxygen treatment was given for five days continuously. The histopathological changes in the liver were measured by counting the amount of normal hepatocytes, pycnotic, karyolysis, karyorrhexis, and score of necrosis area using light microscope with 400x magnification.Result: The amount of pycnotic, karyolysis, and karyorrhexis hepatocytes increased in G2 and decreased significantly in G3, likewise the score of necrosis area. The statistical test using oneway Anova and LSD test showed significant differences (p<0,05) in pycnotic, karyolysis, and karyorrhexis hepatocytes between G1, G2, and G3.Conclusion: The hyperbaric oxygen treatment effect on liver hepatocyte damage in oral candidiasis immunosuppressed rats.
