AbstrakAtenolol adalah pemblok selektif β-1 yang memberikan efek antihipertensi. Atenolol memiliki permeabilitas yang kurang baik dalam saluran cerna sehingga untuk meningkatkan bioavaibilitas dibuat sediaan dalam bentuk patch bukal mukoadhesif. Penelitian ini bertujuan untuk mengetahui pelepasan atenolol dan karakteristik fisik patch bukal mukoadhesif dengan variasi kadar Na-CMC dan PVP K-30. Sediaan patch bukal dibuat dengan lima variasi kadar (Na-CMC:PVP K-30) yaitu formula I (90:10); formula II (70:30); formula III (50:50); formula IV (30:70) dan formula V (10:90). Patch dibuat dengan menggunakan metode solvent casting. Evaluasi sediaan patch meliputi mutu fisik, keseragaman bobot, ketebalan patch, daya tahan lipatan, pH permukaan, swelling index, keseragaman kandungan, dan pelepasan atenolol secara in-vitro menggunakan alat sel difusi Franz. Hasil penelitian memperlihatkan ketebalan patch 1,12-1,16 mm, bobot patch 0,22 – 0,23 gram, pH 6,71-6,81 dan ketahanan lipatan > 300 lipatan. Pelepasan terbaik Atenolol mencapai 10,72 % pada Formulai I yaitu dengan perbandingan konsentrasi Na-CMC dan PVP K-30 (90:10)Kata kunci : atenolol, patch bukal, Na-CMC, PVP K-30AbstractAtenolol is β-1 selective bloker that widely used for the treatment of hypertension. Atenolol has poor permeability in the gastrointestinal tract, to increase the bioavaibility it was made as mucoadhesive buccal patches. This study was aimed to know the release of atenolol and physical characteristics with variations in the base levels of Na - CMC and PVP K-30. Buccal patch preparations were made with five levels of variation consisting of formula I (90:10); formula II (70:30); formula III (50:50); formula IV (30:70) and the formula V (10:90). Patches were prepared by solvent casting method. Patches were evaluated includes physical quality, patches thickness, mass uniformity, folding endurance, surface pH, swelling index, drug content uniformity and release of atenolol in-vitro using Franz diffusion cells. The results showed thickness of patch 1,12 – 1,16 mm, weight patch 0,22-0,23 gram, pH 6,71-6,81 and folding endurance > 300 folds. The best sustained release Atenolol up to 10,72% was show at Formula I which is a concentration ratio of Na-CMC and PVP K-30 (90:10).Keywords : atenolol, buccal patches, Na-CMC, PVP K-30
