Abstract

Our previous study revealed that modified MAGSi (magnetic-silica nanopartivel beads) had various sizes and shapes that affected the DNA extraction process. In this study, we combined our MAGSi with several kinds of lysis buffer for DNA extraction to analyze the result. Human saliva DNA was extracted using three kinds of MAGSi. Each of them was made differently. We used ATL buffer, Triton Buffer, and without buffer for cell lysis. The concentration and purity of the extracted DNA were analyzed using spectrophotometry. Extracted DNA also was used as template DNA for DNA amplification using beta globin gene. This study revealed that our MAGSi-1 provided the best result for DNA concentration and purity. This study also revealed that DNA extraction using lysis buffer produced less concentration yield and purity value but it was more consistent. The concentration and purity value were different but consistent. All the extraction can be used as DNA template for amplification of human beta globin gene. Further studies are still needed to determine the effectiveness and consistency of the lysis buffer and our MAGSi.

Abstract

Black Soldier Fly larvae commonly known as maggot (Hermetia illucens), are currently gaining momentum to convert organic waste. Using bioconversion technology of organic waste with maggot, will not only help in decomposing waste but also the results of cultivation can improve the economy of society. This community service aimed to provide assistance in maggot cultivation for the cattle breeder community, Sangatta City, East Kalimantan province, so that they are familiar with and able to cultivate maggot, especially for producing high protein feed ingredients. The present community service activity was attended by a group of cattle breeders from the Sangatta area, East Kalimantan, and accompanied by a community service team from Mulawarman University. The activity lasted for three months and the implementation was carried out using the method of material presentation, discussion, question and answer, and the practice of maggot cultivation. The activity was carried out at the house of the Sangatta cattle farmer, East Kalimantan. As a result of the activity, cattle breeders have been able to start maggot cultivation and provide an evaluation that the activity is very important because it adds new insight and understanding about maggot cultivation, especially for food security in the area around the capital of the archipelago, Indonesia.  ---  Larva lalat tentara hitam atau dikenal awam sebagai belatung atau maggot dengan nama ilmiah (Hermetia illucens), saat ini popular dibudidayakan untuk merombak limbah organik. Dengan adanya teknologi biokonversi limbah organik dengan maggot, maka tidak hanya membantu dalam merombak limbah namun juga hasil budidaya dapat meningkatkan perekonomian. Pengabdian kepada masyarakat kali ini bertujuan memberikan pendampingan budidaya maggot bagi komunitas peternak sapi Sangatta, Kalimantan Timur, agar mengenal dan mampu membudidayakan maggot dan menghasilkan larva yang mengandung bahan pakan berprotein tinggi. Pengabdian diikuti oleh kelompok peternak sapi dari wilayah Sangatta, Kalimantan Timur dan didampingi oleh tim pengabdi dari Universitas Mulawarman. Kegiatan berlangsung total selama 3 bulan dan pelaksanaan dilakukan dengan metode presentasi materi, diskusi, tanya jawab dan praktek budidaya maggot. Kegiatan dilaksanakan di rumah peternak sapi Sangatta, Kalimantan Timur. Hasil kegiatan, peternak sapi telah mampu memulai budidaya maggot dan memberikan evaluasi bahwa kegiatan sangat penting karena menambah wawasan dan pemahaman baru tentang budidaya maggot terutama untuk ketahanan pangan di wilayah sekitar Ibukota Negara Nusantara, Indonesia.

Abstract

Increasing biology teachers’ comprehension of the principles of genetics is attainable through the implementation of specialized training sessions focused on chromosome and karyotype analysis. In this training, 21 high school biology teachers from Samarinda and Nears participated in a structured training program encompassing three stages. In the initial phase, participants were provided with materials on the fundamental aspects of genetics. Subsequently, in the second phase, hands-on training was imparted, focusing on acquiring skills in plant chromosome preparation techniques. Lastly, the third stage concentrated on refining skills to construct chromosome karyotyping. Completing all stages was required for participants, followed by submitting evaluation forms for assessment by the committee. The evaluation outcomes revealed that 84.30% of participants regarded the presentation and assistance provided by principal lecturer and assistants as highly commendable. At the same time, the remaining respondents expressed satisfaction with the qualityand adequacy of the training. This comprehensive training effectively equipped participants with specialized chromosome and karyotype analysis skills, and the participants notably absorbed the training materials. The anticipated outcome of enhanced teacher understanding, specifically in the knowledge of inheritance and chromosome biology, was to stimulate heightened levels of student engagement and learning interest within the biology classroom.

Abstract

Masyarakat di Pulau Kalimantan hingga saat ini masih banyak yang memanfaatkan keanekaragaman hayati tumbuhan sebagai obat tradisional, termasuk masyarakat Suku Kutai yang bermukim di Kecamatan Kenohan, Kabupaten Kutai Kartanegara. Pemanfaatan tumbuhan obat ini telah berlangsung lama, terutama oleh masyarakat yang tinggal di sekitar hutan. Penelitian ini bertujuan untuk mengidentifikasi jenis tumbuhan obat yang digunakan oleh masyarakat Suku Kutai, bagian tumbuhan yang dimanfaatkan, cara pengolahan, serta manfaatnya. Penelitian ini menggunakan metode survei deskriptif dengan teknik wawancara semi-terstruktur (semi-structured interview) yang dilengkapi dengan keterlibatan aktif peneliti dalam kegiatan masyarakat setempat melalui pendekatan Participatory Ethnobotanical Appraisal (PEA). Pengambilan sampel tumbuhan obat dilakukan dengan metode transect-walk sampling. Hasil penelitian mengidentifikasi 103 spesies tumbuhan dari 48 famili yang dimanfaatkan sebagai obat tradisional. Bagian tumbuhan yang paling banyak digunakan adalah daun sebesar 51%, diikuti oleh akar sebesar 16%, umbi sebesar 11, batang sebesar 10%, kulit batang sebesar 2%, getah sebesar 2%, buah sebesar 5%, dan biji sebesar 2% serta bunga 1%. Metode pengolahan tumbuhan obat yang umum dilakukan adalah dengan cara direbus sebesar 71%, ditumbuk sebesar 14%, diparut sebesar 9%, tanpa diolah sebesar 3%, dan diremas sebesar 3%. Tumbuhan obat ini dimanfaatkan oleh masyarakat untuk mengobati berbagai penyakit, termasuk penyakit degeneratif seperti diabetes, penyakit jantung, asam urat, dan hipertensi, serta penyakit tropis seperti sakit kepala, malaria, perawatan pasca-melahirkan, dan pendarahan.

Abstract

ARTICLE HIGLIGHTS- This research is the first study to provide data on the diversity of Citrus spp. from North Penajam Paser Regency (IKN) area, East Kalimantan for further development and breeding program. - This research offers new information on the distribution of Citrus spp. from the North Penajam Paser Regency (IKN) area, East Kalimantan. - This research provides information on the phenetic relationships among Citrus spp. from the North Penajam Paser Regency (IKN) area, East Kalimantan, and the morphological characteristics that play the most significant role in their clustering. ABSTRACTCitrus spp. is local Indonesian fruit with high diversity and widely distributed around Indonesia country. Diversity and relationship data of Citrus spp. from Penajam Paser Utara-East Kalimantan has never been studied before. This study aimed to provide the diversity and relationship data of Citrus spp. from Penajam Paser Utara using morphological characteristics which served as the basic data for further development and breeding. The research was conducted through observation and sampling, followed by characterization and scoring of morphological data, and lastly the data analysis. The research succeeded in finding 35 accessions of Citrus spp. consisting of C. limon “Jeruk Lemon”, C. suhuensis, “Jeruk Siam”, C. aurantiifolia “Jeruk Nipis”, C. sinensis “Jeruk Manis”, C. maxima “Jeruk Bali”, C. microcarpa “Jeruk Kalamansi”, and Citrus sp. "Jeruk Sunkist" spread in sub-district Penajam, Sepaku, Babulu, and Waru. The dendrogram to clustering Citrus spp. in PPU was divided into two large clusters. Cluster A was grouped based on the character of tree habitus, cordate leaf apex, entire leaf margin, pink mesocarp color, and seed absence. Cluster B was grouped based on the character of the shrubs habitus, acute and rounded leaf apex, crenate leaf margin, white to orange mesocarp color, and numerous seeds in the fruit. This diversity made Citrus spp. from Penajam Paser Utara had high potential value for further development through a plant breeding program.
