Abstract

Acne was a chronic skin disorder that was most often found in adolescents, so it frequently caused psychological effects that made the acne fighters look for ways to treat it quickly. Although topical antibiotics effectively treat acne, excessive long-term use can cause resistance. This has led to an interest in natural antimicrobials as an alternative treatment for acne. The natural antibacterial activity of the ethanol extract of bacang mango (Mangifera foetida L.) leaves has previously been reported to have antibacterial effects against Propionibacterium acnes, but only in the form of extracts not formulated into pharmaceutical preparations. Therefore, this study aimed to re-test the antibacterial activity of this extract, which has now been formulated into a gel preparation against the same bacteria. The gelling agent used in the gel formulation was Na-CMC with three different concentrations: 3%, 4%, and 5%. The Agar stab technique tests the antibacterial activity for three different extract concentrations. Those were 10%, 15%, and 20% for optimization of inhibitory activity before being formulated. Based on the results of testing the antibacterial activity of the ethanol extract of bacang mango (Mangifera foetida L.) leaves, an extract concentration of 20% had a more potent inhibitory activity than the others so that this concentration was chosen to be formulated into a gel preparation. After the formulation of the extracted gel, Na-CMC with a concentration of 5% produced a better gel base than other formulations. In addition, the three formulations provided the same inhibitory activity of 10 mm, so it can be concluded that the differences in gelling agent concentrations did not affect the antibacterial activity of the extracts in the preparations. However, a 3 mm decrease in inhibition occurred in the extract after it was formulated into a gel preparation. Even so, the extract gel preparation still has antibacterial activity against Propionibacterium acnes with antimicrobial inhibition zone activity, which is in the moderate category.

Abstract

Paparan sinar ultraviolet (UV) dari matahari yang berlebihan akan meningkatkan produksi Reactive Oxygen Species (ROS) dan mengaktifkan makrofag untuk melepaskan enzim matriks metalloproteinase (MMP) yang terlibat dalam degradasi matriks ekstraseluler kulit, seperti pelepasan kolagen dan elastin. Dalam mengatasi hal tersebut, lapisan epidermis kulit menyediakan berbagai senyawa penyerap sinar UV salah satunya yaitu melanin. Tetapi produksi pigmen melanin yang berlebihan dapat menyebabkan hiperpigmentasi yang menyebabkan tanda-tanda penuaan kulit. Sehingga, untuk mencegah hiperpigmentasi diperlukan tabir surya. Mikroalga Spirulina platensis menjadi salah satu bahan alam yang berpotensi sebagai bahan aktif tabir surya karena mengandung flavonoid dan senyawa fenolik yang melimpah. Kandungan tersebut bermanfaat untuk menyerap sinar UV. Penelitian ini bertujuan untuk mengetahui pengaruh variasi konsentrasi ekstrak etanol 90% Spirulina platensis yaitu 6%, 7%, dan 8% terhadap nilai SPF krim tabir surya dengan krim yang tidak diberi ekstrak, karakteristik fisik dan stabilitas sediaan krim tabir surya. Hasil pengujian SPF tertinggi secara in vitro dari krim dengan konsentrasi ekstrak 8% yaitu sebesar 22,05 dengan kategori proteksi ultra, dan krim yang tidak diberi ekstrak nilai SPFnya sebesar 0,5. Hasil evaluasi krim tabir surya berekstrak yaitu berwarna hijau pekat kecoklatan, bau khas wangi spirulina kuat, konsistensinya lembut, homogen, pH 6, tipe emulsi M/A, daya lekat 2-7 detik, dan daya sebar 5 – 6 cm. Uji stabilitas dengan cycling test menunjukkan hasil semua krim tabir surya stabil.

Abstract

Acne vulgaris is one of the most common dermatological diseases, affecting individuals of all ages, particularly during adolescence. One of the causes of acne vulgaris is the bacterium Cutibacterium acnes. Acne vulgaris is usually characterized by comedones, papules, pustules, or nodules. Factors influencing acne include genetics, psychological factors, hormones, bacterial infection, and sebaceous gland activity. One regularly used treatment for acne vulgaris is acne patches. With the aid of natural ingredients such as lemon juice and pure honey, an alternative treatment for acne vulgaris can be developed. Lemon juice (Citrus limon L.) can be utilized as an antibacterial agent against Cutibacterium acnes due to its flavonoid content, which has biological activity as an antibacterial. Meanwhile, pure honey (Honey bee) is an antioxidant that can kill bacteria and germs, causing itchy acne on the skin, and can help fade acne scars. The study employed a laboratory experimental design. This research aimed to formulate an anti-acne patch preparation containing a combination of lemon juice (Citrus limon L.) and an ethanol extract of pure honey (Honey bee) that would exhibit good physical stability. Evaluation of the anti-acne patch preparation included organoleptic tests, pH, weight uniformity, patch thickness, moisture absorption, and antibacterial activity tests against Cutibacterium acnes using the disk diffusion method. The results indicated that the anti-acne patch exhibited favorable physical properties. The combination of lemon juice and ethanol extract of pure honey enhanced antibacterial activity against Cutibacterium acnes. The antibacterial activity test results showed that the combination of lemon juice and ethanol extract of pure honey in the anti-acne patch at a concentration of 14% had the highest antibacterial activity, with an inhibition zone diameter of 51.38 mm.
