Abstract

Piperin adalah senyawa marker bagi tanaman dengan famili Piperaceae. Senyawa ini mempunyai rasa yang khas dan dijadikan rempah- rempah, sumber makanan, dan diaplikasikan sebagai pengobatan tradisional jamu dan senyawa ini berguna pada berbagai penyakit sebagai antiproliferative, antitumor, antiangiogenesis, antioksidan, antidiabetes, antiobesitas, kardioprotektif, antimikroba, antipenuaan, hepatoprotektif, anti-alergi, anti-inflamasi, neuroprotektif, dan efek imunomodulator. Pengujian kadar piperin sebagai kontrol kualitas diperlukan pada obat tradisional karena piperin merupakan senyawa aktif berkhasiat sekaligus sebagai senyawa mayor dalam beberapa tanaman khususnya genus Piper. Penelitian ini bertujuan untuk melakukan validasi metode analisis senyawa piperin dan mendeteksi marker senyawa piperin dalam obat tradisional menggunakan KLT-Densitometri. Penelitian ini bersifat eksperimental meliputi validasi metode analisis senyawa piperin dengan konsentrasi 10 ppm, 15 ppm, 20 ppm, 25 ppm, dan 50 ppm dan deteksi piperin dalam jamu CM, Jamu AU, dan jamu PS konsentrasi 100 ppm menggunakan KLT sebagai fase diam, dan heksan: etil asetat (2:1 v/v) sebagai fase gerak dan pada panjang gelombang 340 nm. Hasil penelitian menunjukkan koefisien determinasi pada untuk parameter lineritas yaitu 0,9985, simpangan deviasi (SD) 0,0004 – 0,0088, koefisien variasi 0,04-1,04, % recovery 92.46% - 105.65%, LoD = 2.93 ppm, dan LoQ = 9.79 ppm. Untuk pengujian kadar piperin dalam jamu, marker piperin dalam jamu dapat ditemukan dengan nilai Rf 0,37, dan kandungan kadar piperin terdeteksi dalam Jamu CM 66,49 ppm > Jamu AU 4,64 ppm > jamu PS 1,9 ppm. Penggunaan KLT Densitometri dalam validasi metode analisis dan kontrol kualitas senyawa dalam produk jamu dapat dilakukan dan menghasilkan nilai sesuai dengan syarat validasi dan mendeteksi marker senyawa dalam jamu dapat diaplikasikan dengan KLT-densitometri.

Abstract

Background: The yield of active compound in a plant can be affected by the extraction method, such as variety of solvent and extraction technique used. Objectives: This study aimed to optimize the yield of Lannea coromandelica leaf extract using several types of solvent and extraction techniques and evaluate the antioxidant activity of the extracts. Material and Methods: This study used a variety of solvent like distilled water, ethanol, n-hexane, and ethyl acetate with maceration, reflux, and sonication extraction techniques. The extract obtained was calculated for the percent yield and analyzed for their antioxidant activity using the DPPH method by UV -Vis Spectrophotometry with 523.5 nm maximum wavelength. The antioxidant activity was analyzed by factorial design analysis in Minitab 21 software. Results: The highest percentage yields was obtained from the extract with ethanol solvent, followed by distilled water, ethyl acetate and n-hexane. The highest extraction techniques was obtained from reflux and followed by maceration and sonication extracts. Analytical factorial design analysis showed that the linearity of solvent variation and extraction technique had a significant effect (P>0.05). From the analysis, it was found that there was a significant difference between the effect of the solvent on the antioxidant activities compared to the extraction technique. The extracts obtained have a very strong antioxidant activity against DPPH radicals. T Conclusions: L. coromandelica plant aquadest extract has potential to be developed in optimalization extraction method.

Abstract

Lannea coromandelica plant, commonly called tammate, is one plant that can be used as traditional medicine to treat various diseases. This plant contains metabolites that are active as antioxidants, antimicrobials, and antibacterials. This study aims to determine the fingerprint profile of L. coromandelica bark extract metabolite compounds with variations in densitometric and chemometric extraction methods. The extraction techniques used are maceration, reflux, and sonication, and the types of solvents used are aquades, 70% ethanol, ethyl acetate, and n-hexane. Each extract was analyzed by Thin Layer Chromatography (KLT)-Densitometry, and then the results were analyzed by chemometric analysis using Principle Component Analysis (PCA) and Cluster Analysis (CA). Densitometric results of almost all samples contain the same three metabolites, namely autogenerated1 (Rf 0.72, 0.76, and 0.77), autogenerated3 (Rf 0.90, 0.93, 0.94, and 0.95) and autogenerated5 (Rf 0.10, 0.11, 0.12, 0.13 and 0.15). From this study, it can be concluded that there is a similarity of L. coromandelica bark extract with a similarity value of 29,68% to 99,99%.
