Abstract

Dendrobium lasinthera is an orchid that has considerable potential as a raw material for anti-cancer drugs and for the production of cut flowers. Therefore, this orchid is a plant with high economic value. This orchid has beautiful and long-lasting flowers. The demand for this orchid is increasing along with its diverse uses, so the propagation of this orchid needs to be increased. In vitro propagation is the most appropriate step to reproduce this orchid. One of the stages that determines the success of orchid propagation through tissue culture is the root induction stage before acclimatization. Using the right type and concentration of Growth Regulatory Substances (ZPT) is very important. This research aims to determine the effect of several NAA Growth Regulatory Substance (ZPT) concentrations and determine the best NAA concentration for Dendrobium lasianthera root induction. This study used a Completely Randomized Design (CRD) with several NAA concentrations (Control (A); 0.5 mg/L (B), 1 mg/L (C), and 1.5 mg/L (D)), on Murashige & Skoog (MS) media. The results of this study show that NAA has a significant influence on the percentage of root formation, root emergence time, and root length in the Dendrobium lasianthera orchid. NAA 1.5 mg/L is the best concentration for induction of Dendrobium lasianthera orchid roots.

Abstract

Dendrobium lasianthera (Orchidaceae) is one of the potential plants as ornamental plants because it has beautiful and long-lasting flowers. Propagation of orchid plants through in vitro culture technique has been widely done to meet market demand. One of the supporting factors in plant propagation through in vitro culture techniques is growth regulators. Cytokinin is one of the growth regulators commonly used in plant propagation, including orchids. The study aims to determine the effect of several cytokinins and find the best type for shoot induction of Dendrobium lasianthera. The method used a Completely Randomized Design (CRD) with 4 treatments and 6 replications with several types of cytokinis (control; BAP 1.2 mg/L, Kinetin 2 mg/L, and Thidiazuron 1 mg/L) in Murashige & Skoog (MS) media. The results showed that several cytokinins significantly affected the number of shoots, time of shoot appearance, and shoot length of D. lasianthera. Thidiazuron was the best cytokinin for in vitro shoot induction of D. lasianthera

Abstract

Cytokinins are growth regulators that play a crucial role in cell division and shoot growth. Thidiazuron, a potent cytokinin, was the focus of our research. The research aimed to determine the effect of several concentrations of Thidiazuron on the protocorm growth of Grammatophyllum stapeliiflorum and to identify the best concentration of Thidiazuron (mg/L) for the protocorm growth of G. stapeliflorum orchids in vitro. The research was conducted from January until March 2022 at the Plant Physiology Laboratory, Universitas Andalas, Padang. The research used a Completely Randomized Design (CRD) with 5 treatments and 5 replications. Several concentrations of Thidiazuron were added to MS media with various concentrations: 0 mg L-1 (A), 0.50 mg L-1 (B), 1 mg L-1  (C), 1.50 mg L-1(D), and 2 mg L-1  (E). The parameters in this research were the percentage of life explants, day of shoot appearance, number of shoots, shoot length, number of leaves, number of roots, and root length. The results showed that Thidiazuron significantly affected the protocorm growth of the G. stapeliiflorum orchid compared to the control treatment. Notably, 2 mg L-1  Thidiazuron emerged as the best concentration for all growth parameters of protocorm G. stapeliiflorum, a finding that could significantly impact our understanding of orchid cultivation and plant physiology.
