Abstract

This study aims to determine the best dose of propolis to treat Carassius auratus infected with Aeromonas hydrophila seen leucocyte differentiation from C.auratus. The method used was experimental method with Completely Randomized Design (CDR) with one factor consisting of five treatment levels and three replications. The treatment was Kn (not infected with A. hydrophila and untreated propolis), Kp (infected with A. hydrophila but not treated with propolis), and fish infected with A. hydrophila treated propolis with dose P1 (700 ppm), P2 (800 ppm), and P3 (900 ppm). Treatment is done by injection of propolis in fish infected by A. hydrophila in intramuscular. The fish used are 8-10 cm in size and kept for 14 days post-infection. The results showed that propolis solution has been able to treat C. auratus from infected A. hydrophila. The dose of propolis 800 ppm is the best dose to treat C. auratus infected A.hydrophila, seen from leukocyte differentiation (lymphocyte 80,25%, monocytes 10,09% and neutrophils 9,66%).

Abstract

Penelitian ini dilaksanakan pada bulan Desember 2019 s/d Februari 2020 bertempat di Laboratorium Parasit dan Penyakit Ikan Fakultas Perikanan dan Kelautan Universitas Riau. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan larutan daun kersen (Muntingia calabura L.) terhadap gambaran leukosit dan mendapatkan dosis terbaik penambahan larutan daun kersen ke dalam pakan. Metode yang digunakan adalah metode eksperimen dengan menggunakan Rancangan Acak Lengkap (RAL) dengan lima taraf perlakuan yaitu Kn= Kontrol negatif (pemberian pakan tanpa penambahan kersen), Kp= Kontrol positif (pemberian pakan tanpa penambahan kersen), P1= penambahan larutan daun kersen dengan dosis 1 mL/kg Pakan, P2= penambahan larutan daun kersen dengan dosis 2 mL/kg pakan dan P3= penambahan larutan daun kersen dengan dosis 3 mL/kg pakan. Hasil penelitian menunjukkan larutan daun kersen berpengaruh nyata terhadap gambaran leukosit ikan jambal siam (Pangasianodon. hypophthalmus) dilihat dari adanya peningkatan total leukosit, diferensiasi leukosit, dan aktivitas fagositosis (P<0,05). Dosis terbaik terdapat pada perlakuan P3 (3 mL/kg pakan) dengan rata-rata jumlah leukosit 9,38x104 sel/mm3, aktivitas fagositik sebesar 31,67%, dan tingkat kelulushidupan mencapai 86,67%. Sedangkan persentase limfosit 83,00%, monosit 9,00% dan neutrofil 8,00%.

Abstract

Ikan mas (Cyprinus carpio) merupakan jenis ikan air tawar yang banyak dikonsumsi. Salah satu penyakit yang biasa menyerang ikan mas adalah Motile Aeromonos Septiceamia yang disebabkan oleh Aeromonas hydrophila. Vaksinasi adalah pemberian vaksin yang tujuan utnuk meningkatkan kekebalan ikan terhadap bakteri A.hydrophila. Penelitian ini bertujuan untuk menganalisis profil darah merah ikan mas yang diberi vaksin A. hydrophila pada pakan dengan dosis yang berbeda, dan mendapatkan dosis vaksin A. hydrophila terbaik ditambahkan ke pakan. Metode yang digunakan dalam penelitian ini adalah metode eksperimen dengan menerapkan Rancangan Acak Lengkap (RAL) satu faktor dengan empat taraf perlakuan, yaitu adalah Kn (Pemberian pakan tanpa diberi Vaksin A.hydrophila), P1 (Pemberian Vaksin A.hydrophila dengan Dosis 3 mL/Kg pakan), P2 (4 mL/Kg pakan), dan P3 (5 mL/Kg pakan). Ikan dipelihara dalam keramba berukuran 1x1x1 m sebanyak 12 unit. Ikan yang digunakan berukuran 8-10 cm, dipelihara selama 30 hari. Hasil penelitian menunjukkan bahwa ada pengaruh pemberian A.hydrophila pada pakan terhadap profil darah merah ikan mas yang dipelihara di keramba (P<0,05). Dosis terbaik pemberian vaksin A.hydrophila pada pakan yaitu 5 mL/kg pakan (P3), dilihat dari gambaran darah merah seperti rata-rata total eritrosit 2,12×106sel/mm3, hemoglobin 8,67 g/dL, kadar hematokrit 34,33%, tingkat perlindungan relatif 66,67%, dan kelulushidupan 97,33 %.

Abstract

This research was held from December 2020 – February 2021 at the Laboratory of Fish Parasites and Diseases, Faculty of Fisheries and Marine Science, Universitas Riau. The purpose of this study was to analyze and obtain the best dose of cherry leaf solution to prevent Motile Aeromonas Septicemia from total erythrocytes, hematocrit and hemoglobin levels of striped catfish (Pangasianodon hypophthalmus) fish. The method used to experiment with Completely Randomized Design (CRD) is one factor of five treatment levels. The doses used were negative control without soaking the cherry leaf solution and not tested against Aeromonas hydrophila, positive control without soaking the cherry leaf solution and tested against A. hydrophila, soaking the cherry leaf solution with a dose of 2800 ppm (P1), 3,200 ppm (P2), and 3,600 ppm (P3). Immersion was performed five times for 5 minutes with an interval of 7 days. The results showed that soaking with cherry leaf solution at a dose of 3,600 ppm (P3) was the best dose (P<0.05) with a total value of erythrocytes after the challenge test of 209,66 x 104 cells/mm3, hematocrit level of 37%, hemoglobin 11.86 g/dL, and the survival rate of the test fish was 86,67%.

Abstract

Probiotic containing Nitrosomonas sp and Nitrobacter sp play a role in the nitrification process to improve water quality in the media maintenance. This research was conducted in July – October 2021 at the Laboratory of Fish Parasites and Diseases, Faculty of Fisheries and Marine, Riau University. The purpose of this study was to obtain the best dose of Kp Super N probiotics for fish health. The method used is an experimental method using a one-factor with five treatment levels and three replications. The treatments applied were Cn (maintenance without probiotic Kp Super N and without being challenged with Aeromonas hydrophila), Cp (maintenance without probiotic Kp Super N and challenged with A. hydrophila), maintenance was given probiotic Kp Super N and challenged with A. hydrophila: P1 (2 mL/80 L of water), P2 (3 mL/80 L), P3 (4 mL/80 L). Fish were reared for 72 days. The results shown that the best treatment was P3 (4 mL/80 L) with a total erythrocyte of 2,70 x 106 cells/mm3, haematocrit value of 36,00%, and hemoglobin level of 8,17 g/dl, total leukocytes 8,17 x104 cells/mm3, leukocrite 3,17%, lymphocytes 63,00%, monocytes 4,00%, neutrophils 1,00%, and platelets 32,00%. Weight growth was 23,29 g and the survival rate 100%. The water quality temperature 26 – 29,7oC, pH 6 – 7,5, DO 4,9 – 5,2 ppm, ammonia 0,05430-0,00062 mg/L, nitrite 0,0549-0,1709 mg/L, and nitrate 0,2000-3,6670 mg/L. Based on the date obtain, it can be concluded that the probiotic Kp Super N is able to improve the health of Colossoma macropomum

Abstract

Vegetables in chicory and cabbage contain flavonoid compounds, alkaloids, saponins, glycosides, vitamin C, glucosinolates, glutamine, and sulforaphane, which have antimicrobial activity that can be used as immunomodulators that can strengthen the immune system. This research was conducted from February to April 2022 at the Laboratory of Fish Parasites and Diseases, Faculty of Fisheries and Marine, Universitas Riau. This study aimed to analyze the effect of fermenting mustard and cabbage waste flour in artificial feed on goramy hematology. The method used is an experimental method using a one-factor Completely Randomized Design (CRD) with five treatment levels and three replications. The treatments were P0 (Feed without added fermented chicory and cabbage and challenge with Aeromonas hydrophila), and fish were fed with fermented chicory and cabbage with doses of P1 (15%), P2 (25%), P3 (35%), P4 (45%) and challenged with A. hydrophila. Fishes were reared for 60 days with feed containing fermented chicory and cabbage, and the final data was 14 days of testing with A. hydrophila with the same feed. The results showed that the best dose of feed containing fermented chicory and cabbage was in treatment P2 (25%) with a total erythrocyte of 2.21 x106 cells/mm3, a hemoglobin level of 5.40 g/dL, and a hematocrit value of 38.67%. Tested with A. hydrophila bacteria, total erythrocytes 2.68 x106 cells/mm3, hemoglobin 6.60 g/dL, hematocrit value 39.00%, survival rate 96, 67%, absolute weight growth 1.06 g. Data obtained on O. goramy fed with fermented vegetable waste with cows' rumen showed an effect on improving fish hematology.

Abstract

Biofloc is an agglomerate composed of various autotrophic and heterotrophic organisms as well as particles, which are mixed by aeration and circulation to form clumps that integrate quite well in water. This research was carried out in March- May 2021 in the Hatcheries and Experimental Ponds and Parasites and Fish Diseases Laboratory, Faculty of Fisheries and Marine, Universitas Riau. The study aimed to analyze the leukocyte profile of the Monopterus albus raired biofloc system with different stock densities. The research method used a one-factor, completely randomized design (CRD) with four treatments with three replications. The treatments applied were P1 (of 10 fish/25 L), P2 (of 20 fish/25 L), P3 (of 30 fish/25 L), and P4 (of 40 fish/25 L). M. albus were reared in a biofloc system for 40 days. The results showed that there was an effect of stock densities on the leukocyte profile of M. albus reared in biofloc systems with different stock densities, a significant effect (P<0.05). The best stock density was at P3 (30 fish/25 L), where the total leukocytes were 3.62 x 104 cells/mm3, leucocrite was 2.00%, phagocyte activity was 36.66%, while the percentage of lymphocytes was 83.00 %, monocytes was 16.00%, and neutrophils was 13.00%. Water quality during this study was at temperature 26-300C, pH 6-7, dissolved oxygen 4.4-6.8 mg/L, and ammonia 0.02-0.06 mg/L. It was concluded that there was an effect on the biofloc system of stock density on the leucocyte profile M. albus

Abstract

Aeromonas hydrophila bacteria that cause MAS (Motile Aeromonas Septicemia) disease. Cinnamon contains essential oils, tannins, saponins, flavonoids and phenols. The active compounds of cinnamon essential oil were cinnamaldehyde and cinnamic acid that can inhibit the growth of bacteria. The research was carried out from March to July 2022 at the Laboratory of Fish Parasites and Diseases, Faculty of Fisheries and Marine, Universitas Riau, Pekanbaru. This study aimed to determine the sensitivity of Cinnamomum burmannii in inhibiting the growth of Aeromonas hydrophila bacteria, Minimum Inhibitory Concentration (MIC), and Lethal Dose (LD50) concentration of C. burmannii solution against Cyprinus carpio by immersion technique for 24 hours. The research method used was an experimental method using the Kirby Bauer disc method with concentrations of 100%, 90%, 80%, 70%, 60%, 50%, 40%, 30%, 20%, 10%, 9%, 8%, 7%, 6%, 5%, 4%, 3%, 2%, and 1%. The results shown that the sensitivity of the C. burmannii solution to A. hydrophila bacteria ranged from 22.60 mm to 6.67 mm. The MIC value at a concentration of 4% was determined as the average number of bacterial colonies 143 CFU/mL. The LD50 value of C. burmannii solution against Cyprinus carpio by immersion for 24 hours was 3.64%. Based on the data abtain, it can be concluded that C.burmannii is able to inhibit the growth of A.hydrophila bacteria.

Abstract

Biofloc technology is one of the most profitable alternative solutions to the problem of aquaculture waste. The research was carried out from July to August 2020 at the Laboratory of Fish Parasites and Diseases, Faculty of Fisheries and Marine Affairs, Riau University. The purpose of this study was to isolate and identify candidate lactic acid bacteria (LAB) that have the potential as probiotics from the intestines Monopterus albus and to determine the growth rate of candidate probiotic bacteria and can suppress the growth of the pathogenic bacteria Aeromonas hydrophila. The method used is the experimental method with treatment of pH 2, pH 4 and pH 6, isolation was carried out from the best Monopterus albus rearing with a stocking density of 30 fish/25 liters of water. Candidate probiotic bacteria was found from the intestines of Monopterus albus as many as 6 LAB candidate isolates based on the results of physical, biochemical and pH treatments, the morphology of the probiotic candidate isolates consisted of one type of bacteria, namely Lactobacillus, from the 6 isolates which showed the widest zone of inhibition against the pathogenic bacteria Aeromonas hydropila was isolate derived from Monopterus albus intestine result shown that which grows at pH 2 is BL 2.1.1 with an inhibition zone diameter of 9,05 mm.

Abstract

Biofloc is a technology using good bacteria to convert organic waste to be food source by fish in the form of floc. This research was carried out in April-May 2021 in the Hatchery and blood observations at the Fish Disease and Parasite Laboratory, Faculty of Fisheries and Marine, University of Riau. The purpose of this study was to determine the health status of eels from the profile of erythrocytes cells and the best stocking density of eels cultivated in biofloc system. While the benefits of this study can be applied to biofloc cultivation and apply it in the field. The research method used one-factor completely randomized design (CRD) with four treatment levels and three replications (12 experimental units) for 40 days . The optimal stocking density of eels in clean water is 6 fish for 10 liters. The treatments in this study were P1: stocking density of 10 fish/25 L, P2: stocking density of 20 fish/25 L, P3: stocking density of 30 fish/25 L, P4: stocking density of 40 fish/25 L. The results showed that the erythrocytes profile of eel with different stocking densities had a significant effect (P<0.05) and the best treatment P3 (30 fish/25 L) with total erythrocytes 2.88x106 cells/mm3, hematocrit level 31.66 %, hemoglobin levels 17.40 g/dL, blood glucose 46.33 g/dL, specific growth rate 93.3%, and feed efficiency 67.46%. Water quality in this study was in normal range, that is temperature 25.7-27.3 oC, pH 7.0-7.8, DO 6.4-8.0 mg/L, and NH3 0.0003-0.00038 mg/L
