Abstract

Durian fruit peel is one of the natural ingredients that are considered environmental waste by the people of Indonesia. The use of durian skin is starting to be sought, especially in the treatment of degenerative diseases, one of which is diabetes. Testing of activity as antidiabetic on durian fruit peel needs to be done. Durian fruit peel fraction was made by liquid-liquid partition which was then carried out to make compounds and in vitro antidiabetic potency test. Identification of compounds in the methanol fraction by qualitative TLC and UV-Vis spectrophotometry quantitatively showed high levels of rutin flavonoids. The flavonoid-rich fraction of rutin from durian peel was found to be effective in inhibiting alpha-glucosidase. The methanol fraction of durian fruit peel inhibited the activity of the alpha-glucosidase enzyme significantly (IC50 of 24) and did not differ much from conventional drugs, namely acarbose (IC50 of 23). This study showed that the methanol fraction of durian fruit peel which contains high routine flavonoid compounds has an active ability in in vitro antidiabetic testing.

Abstract

Red ginger is one of the natural ingredients that can be used to treat diabetes mellitus instead of synthetic drugs, which can have too many side effects. The compounds gingerol and shogaol obtained from red ginger have the ability to inhibit the alpha-amylase enzyme's function. Red ginger extract employs a liquid-liquid extraction method with a separating funnel for methanol fractionation and n-hexane fractionation, using a 96% ethanol solvent with an extraction method. The TLC method is utilized to identify the compounds of gingerol and shogaol, and UV-Vis spectrophotometry is employed to measure the inhibition of alpha-amylase enzyme activity. Utilizing the Rf value computation and the IC50 value computation results, data analysis was performed. The result of the methanol fraction of  5,585 µg/ml had the highest IC50 value followed by ethanol extract of 5,778 µg/ml and finally the n-hexane fraction of  5,783 µg/ml.   Keywords:          Fraction, extract, red ginger, inhibition, alpha-amylase enzyme   Abstrak Isi abstrak Pengobatan diabetes melitus dapat menggunakan bahan alam yaitu dengan jahe merah agar dapat mengurangi penggunaan obat-obatan sintesis yang dapat memicu terjadinya efek samping yang berlebih. Jahe merah memiki kandungan senyawa gingerol dan shogaol yang dapat melakukan penghambatan pada aktivitas enzim alfa-amilase. Ekstrak jahe merah menggunakan pelarut etanol 96% dengan metode maserasi, untuk fraksinasi n-heksana dan fraksinasi methanol menggunakan metode ekstraksi cair-cair yang menggunakan corong pisah. Identifikasi senyawa gingerol dan shogaol menggunakan metode KLT untuk uji penghambatan aktivitas enzim alfa-amilase menggunakan spektrofotometri UV-Vis. Analisis data dilakukan dengan menggunakan perhitungan nilai Rf dan hasil perhitungan nilai IC50. Adapun nilai IC50 tertinggi yaitu pada hasil fraksi  metanol sebesar 5.585 µg/ml diikuti dengan ekstrak etanol 5.778 µg/ml dan terakhir fraksi n-heksana sebesar 5.783 µg/ml.   Kata Kunci:         Fraksi, ekstrak, jahe merah, penghambatan, alfa-amylase
