Abstract

Penggunaan antibiotik dalam mengendalikan penyakit pada manusia telah banyak dilaporkan menimbulkan dampak negatif seperti resistensi mikroba dan reaksi alergi. Oleh karena itu diperlukan alternatif pengendalian lain yang lebih ramah dan aman. Bakteri endofit telah banyak dimanfaatkan sebagai agens biokontrol pada berbagai penyakit karena  dikenal sebagai sumber penghasil senyawa-senyawa yang bersifat antimikroba. Tanaman Ketapang (Terminalia cattapa) diketahui memiliki aktivitas antibakteri terhadap beberapa patogen penyebab penyakit dan krmungkinan bakteri endofitnya juga bisa menghasilkan senyawa antimikroba. Penelitian ini dilakukan dengan tujuan untuk mengetahui potensi antimikroba dari senyawa yang dihasilkan oleh bakteri endofit tanaman ketapang dalam menekan pertumbuhan patogen penyebab penyakit pada manusia yang diwakili oleh S.aureus dari bakteri gram positif, E.colli dari bakteri gram negatif, dan C.albicans dari golongan jamur. Percobaan dilakukan dengan metode kirby bauer dengan melihat zona hambat yang dihasilkan bakteri endofit dalam menekan pertumbuhan patogen, sekaligus sebagai sumber antibiotik baru. Hasil pengujian menunjukkan terdapat 2 isolat bakteri endofit BK II dan BK IV memiliki kemampuan terbaik dalam menghambat patogen Staphylococcus aureus dengan daya hambat  masing-masing sebesar 6,21 mm dan 8,23 mm namun tidak memiliki daya hambat terhadap E.colli dan C.albicans. Pengamatan mikroskopis memperlihatkan bahwa senyawa yang dikeluarkan oleh isolat-isolat bakteri endofit tersebut dapat menyebabkan terjadinya malformasi pada S.aureus. Diharapkan isolat bakteri endofit yang diperoleh bisa dikembangkan sebagai sumber antibiotik baru.

Abstract

Endophytic bacteria are bacteria that live in plants and produce compounds similar to their hosts, one of which is antimicrobial compounds. Several studies have shown that certain endophytic bacteria can produce chemical compounds that have health effects, especially endophytic bacteria isolated from medicinal plants. Red betel leaves are known to be able to cure various types of diseases. This plant contains secondary metabolites of the flavonoid, alkaloid, tannin, essential oil, and saponin groups where these compounds have antibacterial activity. This study aims to isolate and test the antibacterial activity of endophytic bacteria from red betel leaves against Staphylococcus aureus and Escherichia coli bacteria , as well as to conduct molecular identification using the 16S rRNA gene. The number of endophytic bacteria that were successfully isolated was 4 isolates, namely 1 shoot leaf isolate (PM), 2 young leaf isolates (MM1 and MM2) and 1 old leaf isolate (TM). The results of antibacterial activity testing were carried out on isolates that had been purified using the Kirby-Bauer method , which showed that each isolate had inhibitory activity against the test bacteria Staphyloccocus aureus , namely: PM: 4.03mm, MM1: 12.1mm, MM2: 3.84mm, all of which were classified as weak. While against the test bacteria Escherichia coli did not have antibacterial activity. The results of molecular identification with the 16S rRNA gene showed that the MM1 bacterial isolate was Bacillus thuringiensis .

Abstract

Background: Bay leaf (Syzygium nervosum DC) is one of the plants that has antibacterial activity that can be used as an active substance in hand soap gel preparations of bay leaf ethanol extract. To formulate a hand soap gel preparation from ethanol extract of bay leaf and evaluate its antibacterial activity. This study is experimental in nature. The research includes the preparation of a concentrated extract from bay leaves and its characterization, followed by the formulation of hand soap gel from the ethanol extract of bay leaves in four different concentrations: F1: 7.5%, F2: 10%, and F3: 12.5%. Evaluation of the physical quality of the formulations includes organoleptic properties, homogeneity, pH testing, viscosity testing, and stability testing. The antibacterial activity of the hand soap gel formulation is assessed using the replica method. The physical evaluation results of the hand soap gel meet the SNI standards. The average percentage decrease in the number of colonies was as follows F0 = 31.8%, F1 = 59.4%, F2 = 81.1%, F3 = 83.2% and the comparison preparation (brand B*) was 83.3%. Bay leaf ethanol extract hand soap gel is capable of inhibiting bacterial growth using the replica method.
