Abstract

Starfruit leaves, which belong to the Averrhoa family and contain tannin, saponin, flavonoid, and terpenoid compounds, have been widely used in traditional medicine due to their pharmacological benefits. This study aims to investigate the effect of solvent maceration on the extraction of total phenolics from starfruit leaves and to evaluate their antioxidant activity capacity. The study utilized Wuluh starf leaves obtained from Biofarmaka IPB University, Bogor, Indonesia, in February 023. The total phenolic content (TPC) and antioxidant tests, such as ABTS and FRAP, were influenced by the maceration stage using solvents, such as a combination of water, ethanol, and methanol. The Folin-Ciocalteu reagent and sodium carbonate were used to determine the total phenolic content, and antioxidant tests, such as the reaction with radical cations (ABTS) and the antioxidant reaction with reduction of ferri-trpyridytriazine (FRAP), were used to evaluate the antioxidant activity capacity. The results indicate that extraction with 100% acetone solvent has the FRAP method's highest phenolic content and antioxidant capacity, with 10.6325 mg GAE/g DW and 34.26641 ± 1.82 µmol TE/g DW, respectively. On the other hand, the 50:50 ethanol:acetone solvent had the highest antioxidant activity using the ABTS method, with 100.31 ± 4.80 µmol TE/g DW. The TPC-FRAP standard curve shows a positive correlation with R2 = 0.9388, while the TPC-ABTS and FRAP-ABTS standard curves do not significant correlation with the R2 values of -0.09045 and 0.08038, respectively.

Abstract

Guava (Psidium guajava) is a popular tropical plant widely used as processed food and medicine for various diseases. This plant contains highly bioactive compounds like quercetin, flavonoids, and ferulic acid. The bioactive content can be influenced by the extraction method. However, the solvent used for extraction can also affect the bioactive content. Therefore, this study aims to determine the best solvent or combination of extraction solvents to extract guava leaves based on total phenolic content (TPC) and antioxidant activity using simplex centroid design (SCD). This research used guava leaf samples with various solvent combinations of water, ethanol, and acetone. The determination of phenolic content was determined using the TPC method, while the determination of antioxidants was conducted using the FRAP and ABTS methods. The research results showed that the highest TPC levels were produced by the solvent water : ethanol : acetone. Then the highest levels of FRAP and ABTS antioxidants were produced by the water: ethanol solvent. The solvent optimization solution obtained was using water: ethanol: acetone with a ratio of 0.462:0.436:0.102, respectively.

Abstract

Parkinson's disease is a global health challenge affecting over 10 million individuals worldwide, leading to increased disability-adjusted life years (DALYs) and a rise in mortality rates. This study explores the potential anti-Parkinson's properties of Xylocarpus granatum, focusing on its interaction with key enzymes associated with the disease: catechol-O-methyltransferase (COMT), adenosine A2A receptor (A2AR), and monoamine oxidase-B (MAO-B). Using molecular docking and molecular dynamics approaches with YASARA Structure, the ethanol extract of X. granatum was investigated for its mechanism of action. Among 30 compounds, five demonstrated promising binding affinities. Structural flexibility analysis revealed minimal fluctuations in active-site residues, highlighting the stability of key complexes involving kaempferol, epicatechin, epigallocatechin, and native ligands. Molecular Mechanics Poisson–Boltzmann Surface Area (MM-PBSA) simulations provided insights into the binding energy of these complexes. Notably, kaempferol exhibited higher binding energy than the natural ligand, suggesting superior binding affinity. Analysis of the average radius of gyration (Rg) showcased control drug-MAO-B exhibited higher Rg values, indicating a more flexible protein conformation. Confirming mode stability with root mean square deviation (RMSD) analysis shows overall stability, except in the A2AR-bound complex. The study's collective findings underscore the structural stabilization of ligand-protein complexes, contributing valuable insights into the potential anti-Parkinson's properties of X. granatum. These discoveries hold promise for developing more effective therapies for Parkinson's disease and significantly contribute to the neurology field.

Abstract

Cardamom (Amomum cardamomum) is a herbal plant with various phytochemical compounds such as phenols, starch, tannins, terpenoids, flavonoids, and antioxidants. Cardamom of the same type but originating from different regions can produce variations in the content of metabolites. This study aimed to select cardamom accession containing high starch, chlorophyll, and antioxidant capacity. The research was conducted at Biopharmaca Experimental Garden, Cikabayan IPB University from March to August 2022 using four cardamom accessions, i.e., Bogor Hijau, Bogor Merah, Ciamis and Sukabumi. Three leaves were taken for each accession, with three replications. The leaves taken were numbers 5, 6, and 7 counted from the tops of the cardamom plant. The methods used in this study were the phenol-sulfuric acid method, the DMSO method, the DPPH method, and the CUPRAC method. The results showed that the highest starch content was produced by the cardamom of Bogor Merah accession, with an average of 3.63 g (100 g)-1 FW. The highest total chlorophyll content was found in the cardamom of Bogor Hijau accession, with an average of 1.9 mg g-1 FW. At the same time, the highest antioxidant capacity using the DPPH method was produced by the cardamom of the Sukabumi accession with an average of 1.28 μmol TE g-1 FW. In comparison, the highest antioxidant capacity of the CUPRAC method was produced by the cardamom of the Bogor Hijau accession with an average of 5.71 µmol TE g-1 FW. Thus, it is recommended to use Bogor Merah dan Bogor Hijau accession for further evaluation. Keywords: accession, antioxidant activity, cardamom leaves, chlorophyll, starch
