Abstract

Obesity is a complex, multifactorial condition contributing to male infertility by increasing oxidative stress, which negatively affects reproductive hormones, sperm quality, and blood cholesterol levels. The Wedelia plant (Sphagneticola trilobata (L.) Pruski) is a natural herbaceous plant containing flavonoids and tannins, known for enhancing sperm quality and inhibiting cholesterol synthesis in the blood. This study aims to determine the potential of Wedelia in improving sperm quality and regulating blood cholesterol levels. The study used 24 male Wistar rats divided into four groups, consisting of C (no HFF, distilled water only), NC (HFF, distilled water), T1 (HFF and 100 mg/Kg BW Wedelia extract), and T2 (HFF and 200 mg/Kg BW Wedelia extract). HFF was administered from weeks 1 to 3, followed with extract treatments from weeks 3 to 5. On day 36, the rats were euthanized for blood collection using hematocrit into EDTA tubes. The rats were dissected for the epididymis, which was then incised in a 10% PBS to release sperm. Sperm motility and count were observed on glass slides, while sperm viability and morphology were analyzed with hematoxylin-eosin using an Optilab microscope (400x magnification). The observation parameters included sperm morphology, count, motility, viability, total cholesterol, LDL, and HDL levels. Data were analyzed using ANOVA with DMRT post hoc testing (p ≤ 0.05). The results showed that T2 exhibited an increase in sperm count, motility, viability, and morphology in HFF-induced obese rats. T1 and T2 also showed enhanced HDL cholesterol and reduced total and LDL cholesterol levels. In conclucions, a Wedelia leaf extract dose of 200 mg/kg BW optimally improves sperm quality and reduces total and LDL cholesterol levels.

Abstract

Wedelia plant, which has the scientific name Sphagneticola trilobata (L.) Pruski, is one of the plants classified as weeds and is found in various agricultural areas. The largest phytochemical compound content in wedelia leaves consists of flavonoids, tannins and alkaloids. The purpose of this study was to identify phytochemical compounds contained in wedelia leaves and to determine the levels of existing compounds. This research method consists of wet sorting, making extracts by maceration method using 96% ethanol. The ethanol extract was then analyzed using Fourier Transform InfraRed (FT-IR), High Performance Liquid Chromatography (HPLC), and UV-Vis Spectrophotometer. Data analysis to determine the line equation and correlation value using statistical analysis application SPSS Version 30.0 and analysis of phytochemical compound components using descriptive analysis. Based on the results of the analysis of phytochemical compounds using the FTIR method, several functional groups were obtained that were very identical to antioxidant compounds in the form of flavonoids, tannins, and alkaloids. After that, further tests on flavonoid levels using the HPLC method obtained levels of 215.435 mg/g (%b/b), and determination of tannin levels using a UV-Vis Spectrophotometer obtained levels of 128.35 mg/g (%b/b).

Abstract

Asap rokok merupakan salah satu sumber radikal bebas yang menyebabkan gangguan pada saluran pernapasan. Songgolangit (Tridax procumbens L.) merupakan salah satu tanaman potensial yang kaya antioksidan dan dapat menangkal radikal bebas seperti paparan asap rokok. Penelitian bertujuan untuk mengetahui potensi songgolangit untuk melindungi saluran pernapasan Tikus Wistar yang terpapar asap rokok. Penelitian menggunakan 25 ekor Tikus Wistar jantan, dengan 5 perlakuan yang terdiri dari K (pemberian akuades), KN (pemaparan asap rokok), P1 (pemaparan asap asap rokok dan pemberian ekstrak 100 mg/kg BB), P2 (pemaparan asap asap rokok dan pemberian ekstrak 200 mg/kg BB), dan P3 (pemaparan asap asap rokok dan pemberian ekstrak 300 mg/kg BB). Pemberian ekstrak dilakukan pada hari ke 1-21, sedangkan pemaparan asap rokok diberikan pada hari ke 8-21. Pada hari ke 22, tikus dieutanasi dan dibedah untuk diambil paru-paru dan trakeanya. Organ dipreparasi menggunakan metode parafin dan pewarnaan hematoksilin-eosin. Parameter pengamatan terdiri dari rasio organ paru-paru, luar inflamasi, jumlah sel edema, diameter paru-paru, tebal epitel trakea dan diameter trakea. Seluruh parameter data dianalisis menggunakan uji ANOVA dan uji lanjutan Duncan (p<0,05). Hasil penelitian menunjukan bahwa perbaikan paling signifikan pada rasio paru-paru, diameter alveolus, jumlah sel edema dan tebal epitel trakea terdapat pada dosis 300 mg/Kg BB (p<0,05), namun tidak ada perbedaan secara signifikan pada luas inflamasi dan diameter lumen trakea (p>0,05). Kesimpulan menunjukan bahwa pemberian dosis 300 mg/kg BB ekstrak songgolangit dapat melindungi struktur saluran pernapasan Tikus Wistar yang dipapar asap rokok.
