Abstract

A study on the Effect of Spermatoza in Bali Cattle Using Various Sources of Carbohydrates with Egg Skim Diluent has been carried out. The method used is Artificial Insemination to accommodate semen with 4 processes for making semen diluent by adding egg yolk skim, namely making skim buffer, making antibiotic buffer A which consists of 90%, skim buffer antibiotic with a mixture of egg yolks with a percentage of egg yolk as much as 10% of the total diluent A and making diluent B which consists of 72% buffer skim antibiotic 10% egg yolk 16% glycerol, and added to each treatment, namely 2% glucose and 2% fructose. From the results of the data obtained there was no significant difference (p 0.05) between the administration of 2% glucose, 2% fructose and 2% sucrose added to each egg yolk skim diluent on spermatozoa motility and the percentage of live spermatozoa. Key words: Artificial Insemination, Carbohydrates, Bali Cattle, Egg Skim, Spermatozoa AbstrakTelah dilakukan penelitian Pengaruh Spermatoza Sapi Bali dengan Menggunakan Berbagai Sumber karbohidrat dengan Pengencer Skim Telur. Metode yang digunakan yaitu Inseminasi Buatan untuk menampung semen dengan 4 proses pembuatan pengencer semen dengan menambahkan skim kuning telur yaitu pembuatan buffer skim, pembuatan buffer antibiotic A yang terdiri atas 90%, buffer skim antibiotic dengan campuran kuning telur dengan presentase kuning telur sebanyak 10% dari total pengencer A dan pembuatan pengencer B yang teridiri atas 72% buffer skim antibiotic 10% kuning telur 16% gliserol, serta ditambahkan dengan masing-masing perlakuan yaitu glukosa 2% dan fruktosa2%. Dari hasil data yang diperoleh tidak terdapat perbedaan nyata (p0.05) antara pemberian glukosa 2%, fruktosa 2% maupun sukrosa 2% yang ditambahkan ke dalam masing-masing pengencer skim kuning telur terhadap motilitas spermatozoa dan presentase spermatozoa hidup.

Abstract

This study aimed to analyze the effects of fructose administration and environmental conditions on the motility and viability of Bali cattle sperm. The study was conducted at the Artificial Insemination (AI) Service Unit over a two-month period, using an experimental method with a completely randomized design (CRD). Semen samples were collected from Bali bulls and then diluted using Andromed diluent at four different fructose concentrations: 0.1 ml, 0.3 ml, 0.4 ml, and 0.6 ml. Evaluations included analyses of sperm motility and viability before and after storage. The results showed no significant difference (p > 0.05) in sperm motility among the fructose treatments. The highest motility (70%) was recorded in the treatment with Andromed diluent + 0.4 ml of fructose; however, variations in fructose concentration did not significantly affect sperm motility. Furthermore, spermatozoa viability showed relatively similar results across all treatments, with the best viability observed at a 0.6 ml fructose concentration. Although fructose serves as an energy source for sperm, increasing the fructose concentration in the diluent did not significantly enhance motility. Environmental conditions such as temperature and humidity remain critical factors influencing sperm quality.
