Abstract

Neurodegenerative diseases represent a growing global health challenge, particularly in aging populations. This review examines the neuroprotective potential of Robusta coffee (Coffea canephora) and its bioactive compounds in neurodegenerative conditions. Robusta coffee contains a rich profile of bioactive compounds, including caffeine, chlorogenic acids, and other polyphenols, which demonstrate significant antioxidant, anti-inflammatory, and neuroprotective properties. Evidence from preclinical and epidemiological studies suggests that these compounds can modulate multiple pathways involved in neurodegenerative processes, including oxidative stress, neuroinflammation, and protein aggregation. This review synthesizes current findings on the molecular mechanisms underlying the neuroprotective effects of Robusta coffee components in conditions such as Alzheimer's disease, Parkinson's disease, and other neurodegenerative disorders. Recent advances in delivery systems, such as nanoparticle formulations, are also discussed as potential approaches to enhance the bioavailability and efficacy of coffee bioactives. By identifying the therapeutic potential of Robusta coffee components and highlighting gaps in current knowledge, this review provides a foundation for developing novel preventive and therapeutic interventions for neurodegenerative diseases based on these naturally occurring compounds.

Abstract

Hiperurisemia adalah gangguan metabolisme akibat tingginya kadar asam urat dalam darah, dengan prevalensi 15% di Indonesia dan 34,2% secara global. Diperkirakan, kematian akibat gout meningkat 55% pada 2060. Kondisi ini umumnya disebabkan oleh peningkatan aktivitas xantin oksidoreduktase (XOR) di hati atau gangguan ekskresi ginjal. Penghambatan XOR terbukti efektif menurunkan kadar asam urat. Indonesia memiliki lebih dari 9.609 spesies tanaman obat, beberapa di antaranya berpotensi sebagai antihiperurisemia melalui mekanisme penghambatan XOR. Allopurinol, obat standar untuk kondisi ini, dapat menimbulkan efek samping serius bila digunakan berlebihan. Oleh karena itu, terapi herbal menjadi alternatif yang menjanjikan. Review ini bertujuan membahas tanaman yang secara empiris mampu menurunkan asam urat dengan menghambat xantin oksidase. Pencarian artikel publikasi dilakukan di beberapa mesin pencari, seperti ResearchGate, ScienceDirect, Google Scholar menggunakan kata kunci "Antioksidan", “Asam Urat” "Hiperurisemia", "In Vitro", "Metabolit Sekunder" dan "Tanaman Obat" berdasarkan kriteria inklusi dan eksklusi, yang menghasilkan 15 artikel. Pencarian literatur dibatasi pada artikel yang diterbitkan dalam sepuluh tahun terakhir, dari 2015 hingga 2025. Hasil review menunjukkan bahwa tanaman tanaman tersebut memiliki potensi sebagai antihiperurisemia dengan mekanisme penghambatan enzim xantin oksidase.Kata Kunci: Antioksidan, Asam Urat, Hiperurisemia, In Vitro, Metabolit Sekunder, Tanaman Obat, dan Xantin Oksidase
