Abstract

Beauveria mildew is known as useful natural control agent, due to it’s infection potential agents for controlling insect. The range of its host covers broad range of insects order including Lepidoptera, Coleoptera, Hemiptera and Hymenoptera. In the past, the behavior of B.bassiana was identified based on morphological character, which is have weakness because of environmental effect so the fenotype performance does not manifest the genotype value. Another identification method can be used to complete morphological approach is identification based on physiological behavior (toxin production) and biochemical (protein and isoenzim) where the protein analysis and isoenzim, is order to minimize the influence of environment factor for identification. The research was conducted in Laboratorium Biology UNM, there is 11 isolates B.bassiana used in this research represent location and host. Isolates were analyzed by physiological and isoenzim using electrophoresis. The result of physiological character showed that out of 11 isolates characterized, there are isolates produced beauvericin, interestingly the rest isolates produced unknown on identified metabolites. Based on total band protein pattern and isoenzim showed that there are variation, isolates from the same area or district have more similarity characters than isolates from different area.

Abstract

Kegiatan pengabdian masyarakat ini bertujuan untuk meningkatkan literasi dan kompetensi guru Biologi dalam memahami dan menerapkan teknik kultur jaringan sebagai bentuk penerapan bioteknologi modern, khususnya pada perbanyakan generatif buah naga (Hylocereus costaricensis) melalui pemanfaatan media kultur alternatif berbasis media pupuk Gandasil D. Pelatihan dilaksanakan pada tanggal 11-12 Agustus 2025 di Labratorium Pascasarjana Jurusan Biologi.  Melalui sesi teori dan praktik laboratorium yang meliputi pembuatan media kultur sederhana (air kelapa, gula, agar, dan variasi konsentrasi Gandasil D), teknik sterilisasi alat serta eksplan, hingga inokulasi biji buah naga secara aseptik di dalam laminar air flow. Melalui kegiatan ini, guru dibimbing untuk memahami konsep kultur jaringan, tahapan perbanyakan generatif, peran nutrisi dalam pertumbuhan eksplan, serta potensi penggunaan media alternatif yang lebih ekonomis dibandingkan media standar seperti MS (Murashige–Skoog). Hasil kegiatan menunjukkan bahwa guru SMA mampu mengikuti seluruh tahapan praktikum dengan baik, memahami fungsi setiap komponen media, serta dapat melakukan inokulasi biji buah naga secara mandiri. Selain meningkatkan kompetensi praktis, kegiatan ini juga memperkuat kemampuan guru dalam merancang pembelajaran bioteknologi yang aplikatif, menarik, dan relevan dengan konteks Kurikulum Merdeka. Pemanfaatan Gandasil D sebagai media alternatif memberikan nilai tambah karena mudah diperoleh, murah, dan dapat digunakan dalam pembelajaran laboratorium di sekolah dengan fasilitas terbatas. Kegiatan ini diharapkan menjadi langkah awal dalam penerapan bioteknologi sederhana di sekolah serta mendorong inovasi praktikum biologi yang lebih kontekstual dan berkelanjutan.

Abstract

Abiotic stresses such as drought, salinity, extreme temperatures, and oxidative stress are major factors limiting the growth and productivity of rice plants. The development of CRISPR-Cas9-based genome editing technology provides a new, more precise approach to modifying genes that play a role in plants' adaptive responses to environmental stresses. This study aims to synthesize the development of genome editing research in rice plants, focusing on the identification of target genes, genome editing strategies, and their impact on increasing abiotic stress tolerance. The method used is a systematic literature review of research articles published in international scientific journals. A total of 31 articles were analyzed in the literature review process, and 22 articles that explicitly reported target genes and genome editing strategies were used in the main synthesis. The results of the analysis showed that most studies targeted regulatory genes that play a role in regulating stress responses, especially groups of transcription factors such as NAC, DREB, bZIP, MYB, and WRKY. In addition, several studies also targeted genes related to ion homeostasis, hormonal signaling pathways, secondary metabolism, and antioxidant systems. The most dominant genome editing strategy was gene knock-out through CRISPR-Cas9-based mutations, while several recent studies have begun to apply multiplex genome editing to target more than one gene simultaneously. Genome-edited rice plants generally show increased tolerance to various abiotic stresses through improved ion homeostasis, increased water use efficiency, cell membrane stability, and strengthened antioxidant systems. These findings indicate that CRISPR-Cas9 is a precision genome engineering approach with the potential to support the development of rice varieties that are more adaptive to environmental changes.
