Abstract

This study investigates the phenotypic alterations in Dendrobium 'Transient White Rika' (RKA) and 'Florenza' (FLO) orchids resulting from colchicine-induced polyploidy during in vitro cultivation. Colchicine, a known mitotic inhibitor, was applied at varying concentrations (0 ppm, 4000 ppm, 5000 ppm, 6000 ppm) to induce chromosome doubling and assess its impact on the vegetative and generative phases of the orchids. A factorial Randomized Block Design (RBD) was employed to analyze the effects of these treatments. The study revealed significant variations in leaf length and color, particularly at higher colchicine concentrations. Morphological changes were most pronounced at 6000 ppm, where the leaf length decreased, and flower morphology exhibited unique features compared to the control. The study also observed a reduction in stomatal density as colchicine concentration increased, correlating with an enlargement of leaf epidermal cells. These findings indicate that colchicine treatment can effectively induce phenotypic changes in Dendrobium orchids, contributing to the development of hybrids with superior ornamental qualities. However, the study also notes that the response to colchicine is variable, with certain parameters like the number of new shoots and leaf width not showing significant changes. The results underscore the potential of colchicine-induced polyploidy in orchid breeding, though further research is necessary to optimize treatment protocols and ensure the stability and viability of the resulting hybrids. These findings may serve as valuable material for genetics-based learning modules in biology.

Abstract

This study aimed to identify mycorrhizal fungi, primarily from the genera Rhizoctonia and Tulasnella, on orchid roots and develop leaflet-based teaching materials using the Research and Development (R&D) model with the 4D approach (Define, Design, Develop, Disseminate). Fungal identification was conducted through microscopic examination and molecular techniques (ITS rDNA sequencing) to ensure accurate classification. The 4D model involved defining student needs, designing leaflet content focused on fungal symbiosis, developing draft materials, and disseminating the final leaflet for classroom use. The effectiveness of the leaflet was evaluated with 40 students through pre- and post-tests measuring knowledge of fungal roles in orchid growth. Results showed the average pre-test score increased from 48.5 to 85.2 in the post-test, with an N-Gain score of 0.722 (72.29%), indicating moderate effectiveness. Improvements were most notable in students’ understanding of fungal identification and symbiotic functions. These findings demonstrate that leaflet-based teaching materials developed from precise fungal identification can significantly enhance student comprehension of Kingdom Fungi concepts.

Abstract

This study aims to analyze the effect of the Project Based Learning model integrated with the Merdeka Curriculum on the learning outcomes of Grade X students in Biotechnology material at SMK Islam Al-Azhar Jember. The study employed a quasi-experimental approach with a pretest-posttest non-equivalent control group design. The sample consisted of 34 students who were given pretests and posttests to measure their biotechnology learning outcomes, using a learning outcome test as the instrument. The results showed a significant improvement in students’ learning outcomes after the implementation of the Project Based Learning model integrated with the Merdeka Curriculum, with the experimental group achieving a posttest average score of 89.73, compared to 76.21 in the control group. The N-Gain score for the experimental group reached 0.73 (73%), which is categorized as high, while the control group scored 0.56 (56%). A t-test indicated a significant difference between the two groups (p = 0.000 < 0.05). These findings indicate that the Project Based Learning model integrated with the Merdeka Curriculum is proven to be effective in improving Biotechnology learning outcomes.

Abstract

This study aimed to develop interactive learning videos using the Prezi application on the topic of photosynthesis to improve the learning motivation of 11th-grade students at SMK Darul Muklisin, Jember. The research employed the Research and Development (R&D) method with the 4D model (Define, Design, Develop, Disseminate). Data were collected through expert validation and student motivation questionnaires. Validation results from media and subject matter experts showed high feasibility, while a limited trial with 10 students indicated a significant increase in motivation scores, from 2.35 (low) to 3.52 (very high). These findings demonstrate that Prezi-based videos are effective in enhancing student motivation and facilitating understanding of complex material such as photosynthesis. In conclusion, interactive video media developed with Prezi is both feasible and effective, making it a potential innovative solution for digital-based classrooms.
