Abstract

Bay leaves (Syzygium polyanthum) contain active compounds such as tannins, saponins, flavonoids, and steroids, which offer various benefits in skincare, including antioxidant, antimicrobial, anti-inflammatory, and natural cleansing properties. This study aimed to evaluate the antioxidant activity of bay leaf extract and assess the quality of lotion formulation containing the extract. Extraction was carried out using the maceration method with 70% ethanol as the solvent, and antioxidant activity was tested using the DPPH (2,2-diphenyl-1-picrylhydrazyl) method, yielding an IC50 value of 48.64 ppm, indicating moderate antioxidant activity. The lotion formulations were evaluated for organoleptic properties, homogeneity, pH, spreadability, viscosity, and emulsion stability. Results showed that formula F2 had superior quality compared to F1, with a spreadability of 6.5 cm (compliant with SNI 16-3499-1996), pH 6.70 (safe range 4.5–8), creamier texture, lower stickiness and greasiness, and faster absorption time (30 seconds). Formula F2 also demonstrated ideal viscosity (6805 mPaS) and shear thinning properties (torQ 68.1%), which ease application on the skin. Based on these findings, formula F2 meets the requirements for topical cosmetics according to SNI and the Indonesian Pharmacopoeia VI edition, especially in terms of pH, physical stability, and viscosity, and thus shows promising potential for development as a natural-based cosmetic product that is both safe and effective in protecting the skin from free radicals and irritation.

Abstract

Acne vulgaris is a common inflammatory skin disorder primarily caused by the proliferation of Propionibacterium acnes. In response to increasing demand for natural and safer therapeutic alternatives, this study investigated the phytochemical composition, total flavonoid content, and antibacterial activity of the ethanolic extract of Myristica fragrans flesh. Phytochemical screening revealed the presence of flavonoids, terpenoids, tannins, saponins, and alkaloids—classes of compounds widely recognized for their antimicrobial properties. The total flavonoid content (TFC), determined using the aluminum chloride colorimetric method, was found to be 0.1582 ± 0.0044 mg quercetin equivalents per gram of dry extract. Antibacterial assays using the disc diffusion method demonstrated dose-dependent inhibition of both Staphylococcus aureus and P. acnes, with greater efficacy observed against P. acnes. These results suggest that nutmeg flesh extract possesses significant antibacterial potential, particularly as a candidate for anti-acne cosmetic formulations. Moreover, this study highlights the underexplored value of nutmeg flesh—a byproduct of spice processing—as a sustainable source of bioactive compounds for skincare applications.

Abstract

Acne vulgaris is a common chronic inflammatory skin disorder primarily caused by Cutibacterium acnes infection. This study aims to investigate the phytochemical composition, total flavonoid content, and antibacterial activity of the ethanolic extract of Myristica fragrans fruit flesh against C. acnes and Staphylococcus aureus. The dried fruit flesh was extracted using ultrasonic-assisted ethanol extraction, followed by qualitative phytochemical screening and total flavonoid quantification via the aluminum chloride colorimetric method. Antibacterial activity was evaluated using the disc diffusion method at extract concentrations of 10%, 20%, and 30%, with standard antibiotics as positive controls. The extract contained key bioactive compounds including flavonoids, terpenoids, tannins, saponins, and alkaloids. Total flavonoid content averaged 0.15820 ± 0.00440 mg quercetin equivalent per gram of dry extract. The extract exhibited dose-dependent antibacterial effects, with more potent inhibition against C. acnes than S. aureus. Inhibition zones at 30% concentration reached notable diameters, confirming significant antibacterial activity based on CLSI standards. These findings highlight the potential of M. fragrans fruit flesh as a promising natural source of multifunctional bioactive compounds for anti-acne cosmeceutical development. Further studies involving compound isolation and in vivo efficacy testing are recommended to support its clinical application.
