Abstract

Pewarnaan sediaan apusan darah tepi (SADT) pewarnaan yang digunakan untuk ekstraksi antosianin dengan berbagai varian kosentrasi menunjukkan kualitas gambar terbaik. Kemungkinan hal ini disebabkan karena jumlah senyawa antosionin sangat berinteraksi dengan sel darah eritrosit, leukosit dan trombosit secara baik. Tujuan dari penelitian ini agar bisa melihat morfologi sel darah eritrosit, leukosit dan trombosit pada manusia. Hasil penelitian yang telah diui terdapat pada pewarnaan alternatif alami umbi gadung ungu (Dioscorea hispida) dimana terdapat senyawa antosionin untuk pewarnaan, umbi gadung menunjukkan warna ungu ke pink dan terlihat jelas sel darah trombosit, leukosit dan eritrosit, sedangkan pada Giemsa menunjukkan warna biru pudar dtetapi masih terlihat jelas sel darah trombosit, leukosit dan eritrosit.

Abstract

Peripheral Blood Smear Preparation (SADT) is a smear preparation made using human blood that is colored for examination in hematology. This examination is done to see and assess peripheral blood cells such as leukocytes, platelets and erythrocytes. Giemsa has a drawback because methylene blue contained in giemsa can be inhaled causing cyanosis. Azur B content can also cause waste because the material contained is not easily decomposed and eosin content can cause allergic reactions.This study aimed to test the water extract of Cambodian flowers (Plumeria rubra L. cv. Acutifolia.) as an alternative dye of peripheral blood smear preparations. This research is in the form of qualitative research with a method of making purple dye for water extraction of frangipani flowers (Plumeria rubra L. cv. Acutifolia.). Microscopic observations of frangipani flower extract (Plumeria rubra L. cv. Acutifolia.) as a preparation of Edge Blood Smear (SADT) can be used as an alternative natural use besides Giemsa staining, because erythrocytes, leokocytes and platelets are able to bind colors well.

Abstract

Coffee husk is one of the resources that has the potencial to be used as raw material for tawes (Barbonymus gonionotus) feed. The addition of Aspergillus niger in this study is known to increase the protein content of coffee husk from 15.99% to 21.75%. This study aims to determine the effectiveness of giving fermented coffee husk using Aspergillus niger on the histology of Tawes intestine. This study used a completely randomized design (CRD) with 5 treatments of coffee husk addition, namely 0%, 10%, 20%, 30%, and 40%, and 4 repetitions. The results showed that the addition of 30% fermented coffee husk into the feed produced the best intestinal histology picture with an average value of villi height of 243.21 40.02 m, basal width of 79.77 30.77 m and apical width of 69.09 14.61 m.Keywords:FermentationCoffee HuskBarbonymus gonionotusTawesAspergilus niger
