Abstract

Pengujian kadar fosfat dalam bahan pakan merupakan kontrol kualitas yang penting dalam penyusunan ransum hewan ternak. Telah dilakukan pengujian kadar fosfat pada bungkil kedelai dan bungkil inti sawit menggunakan microplate Spektrofotometer UV-Vis dan penentuan hasil pengukuran fosfat pada bungkil kedelai dan bungkil inti sawit dengan parameter validasi. Destruksi sampel dilakukan pada microwave digester dengan larutan asam nitrat (HNO3) 65% dan asam klorida (HCl) 37%. Pengukuran serapan  ekstrak sampel dilakukan pada panjang gelombang maksimum 895 nm. Parameter validasi metode yang ditentukan hasilnys yaitu nilai linieritas (koefisien korelasi pengukuran), limit of detection (LoD), limit of quantitation (LoQ), akurasi, dan presisi. Hasil penelitian menunjukan kadar air dan fosfat pada bungkil kedelai dan bungkil inti sawit berturut-turut yaitu 9,92% dan 6,88% serta 0,77% bahan kering (BK) dan 0,49 % BK. Nilai linearitas yang didapatkan yaitu 0,999. Nilai LoD dan LoQ berturut-turut sebesar 0,49 mg/L dan 1,49 mg/L. Nilai presisi metode dinyatakan dalam koefisien variansi Horwitz pada bungkil kedelai dan bungkil inti sawit adalah 7,93% dan 8,42% dan akurasi penelitian sebagai persen perolehan kembali pada bungkil kedelai dan bungkil inti sawit berturut-turut adalah 85,46-89,49% dan 92,05-93,88%. Hasil ini menunjukan bahwa pengujian kadar fosfat dengan metode asam askorbat menggunakan microplate spektrofotometer UV-Visibel untuk analisa fosfat pada bahan pakan hewan ternak telah memenuhi persyaratan validasi dan dapat digunakan untuk analisa rutin.

Abstract

Phosphorus content in animal-derived foods is an important indicator of nutritional value and food safety. This study validated a method for phosphorus analysis in chicken meat and fresh cow’s milk using wet digestion based on microwave digestion and microplate UV-Vis spectrophotometry with the principle of molybdenum blue complex formation by ascorbic acid at a wavelength of 880 nm. Method performance was evaluated according to ISO/IEC 17025:2017. The validation parameters included linearity and linear range, sensitivity comprising the limit of detection (LOD) and limit of quantification (LOQ), precision comprising repeatability and intermediate precision, as well as accuracy. The calibration curve showed good linearity within the range of 2–12 ppm (r² = 0.998). The LOD and LOQ values were 0.29 and 0.97 mg/L for chicken meat, and 0.17 and 0.57 mg/L for cow’s milk, respectively. Precision testing resulted in repeatability and intermediate precision RSDs of 2.00% and 2.18% for chicken meat, and 2.62% and 2.14% for cow’s milk. Accuracy, determined by recovery analysis, showed values of 97.14% for chicken meat and 103.15% for cow’s milk. These results demonstrate that the validated method meets the acceptance criteria of ISO/IEC 17025. The method provides acceptable linearity, sensitivity, precision, and accuracy, and can be applied for routine phosphorus analysis in meat and milk matrices.
