Abstract

Background: Periodontitis is the most severe complication in oral cavity to diabetes mellitus. Stichopus hermanii contain GAG, a polysaccharide which very useful in wound healing. Hyperbaric oxygen therapy is effective repairing ischemia, hypoxia, and is used as adjuvant therapy. Purpose: To determine effect of administration of Stichopus hermanii (SH) 3% gel and hyperbaric oxygen therapy 3x30 minutes with 5 minute intervals in 7 consecutive days to the number of fibroblasts in ligament periodontal diabetes mellitus rats with periodontitis. Materials and Methods: The experiment design is factorial design. Rats are inducted with STZ single dose 65 mg/kgBB to make them suffer DM and are inducted with P.gingivalis 1x109sel/ml for 5 days straight to make them suffer periodontitis. 20 male wistar rats are divided into 5 groups. K- (control), K1 (STZ+P.Gingivalis), K2(STZ+P.Gingivalis+SH), K3(STZ+P.Gingivalis+HBO), K4(STZ+P.Gingivalis+SH+HBO). After 52 days treatment, rats is terminated to make a HPA Sample from the mandible and fibroblast is counted with 400x magnification. Result: One-way Annova here were significant differences (p<0,05) in K- (32.300 ± 11.381), K1 (31.725 ± 5.590), K2 (54.225 ± 7.165), K3 (49.725 ± 10.088), K4 (72.075 ± 9.342). Conclusion: administration of Stichopus hermanii 3% gel combination and 2.4 ATA hyperbaric oxygen 3x30 minutes a day with 5 minutes intervals in 7 consecutive days can increase the number of fibroblasts in diabetes mellitus rats with periodontitis.

Abstract

Background: Periodontitis is complication of DM in chronic state. Periodontitis susceptibility increase threefold in patient DM compared non-DM. Debridement with SRP alone is less effective in DM. Hyperbaric Oxygen Therapy (HBO) has the effect of increasing insulin sensitivity and synthesis of collagen. Collagen is the largest extracellular matrix supporting periodontal tissue. Stchopus hermanii contains condrotin sulfate serves to inhibit the formation of mixed periodontopatogen bacteri. Purpose: To determine the therapeutic effects of HBO 2,4 ATA 3x3’ interval 5’ 7 days and stichopus hermanii against periodontal ligament collagen density in the healing process of periodontitis with DM. Materials and Methods:The experiment design is factorial design. Rats are inducted with STZ 65 mg/kgBB to make them suffer DM and are inducted with P.gingivalis 1x109sel/ml for 5 days straight to make them suffer periodontitis, 20 male Wistar rats were divided into 4 groups. K0 (control group), K1(STZ+P. gingivalis), K2(STZ+P. gingivalis+Stichopus hermanii), K3(STZ+P. gingivalis+HBO), and K4(STZ+P.gingivalis+stichopus hermanii HBO) after treatment on day 52, all groups were sacrificed and done making preparations for the HPA of the mandible bone. Results: Data were analyzed using chi-square, Kruskal wallis, and Mann Whitney U with 95% significance level (p = 0.05). increase level of collagen significantly K1(1 ± 0.00) - K2(2±0.00), dan K1(1 ± 0.00) - K4(2.5 ± 0.289).Conclution:Therapy HBO combination of 2.4 ATA 3 x 30 with 5 minutes air breaks for 7 days and gel Stichopus hermanii3% effective on the Densityof collagen in the periodontal ligament healing process periodontitis with DM.

Abstract

Background: The prevalence of diabetes mellitus (DM) is quite high in the world and Indonesia. Diabetes mellitus with poor glycemic control periodontitis as a complication the largest in the oral cavity which causes damage to the alveolar bone. Osteoblasts synthesize osteoid collagen to form bone as raw material. Oxygenation high pressure is able to increase the amount of alveolar bone osteoblasts in patients with DM. Sea cucumber contains growth factors that play a role in the differentiation of osteoblasts to bone formation. Purpose: To determine the effectiveness of combination hyperbaric oxygen therapy 2.4 ATA and sea cucumber gel 3% in the increasing of osteoblasts in rats with DM induced P.gingivalis. Materials and Methods: The study design was factorial design group. Rats are inducted with STZ single dose 65 mg/kg BB to make DM and are inducted with P.gingivalis 1x109 sel/ml to make them suffer periodontitis. Twenty male Wistar rats were divided into five groups. K-(without treatment); K1 (Streptozotocin + P.gingivalis); K2(Streptozotocin + P.gingivalis + sea cucumber gel); K3 (Streptozotocin+ P.gingivalis + HBO); K4 (Streptozotocin +P.gingivalis +HBO + sea cucumber gel). Result: One-way ANOVA here were significant differences (p<0.05) in K2 (24.97 ± 4.99), K3 (22.80 ± 6.45), K4 (28.47 ± 3.20) that compared with K1 (9.22 ± 4.95). Conclusion: Each hyperbaric oxygen therapy doses of 2.4 ATA 100% O2 3x30 minutes with 5 minute intervals for 7 days and sea cucumber gel (Stichopus hermanii) concentration of 3% and a combination of both therapies showed an increase of osteoblasts significantly in rats with diabetes mellitus induced P.gingivalis.

Abstract

Background: Diabetes mellitus (DM) with Periodontitis cause severe alveolar bone resorption. Osteoprotegerin prevent osteoclastogenesis process so that inhibited alveolar bone resorption. Stichopus hermanii powder and hyperbaric oxygen therapy (HBO) have content and good effects in wound healing. Purpose: The aim of this reseach was to analyze the effect of Stichopus hermanii powder and HBO therapy in the increasing of osteoprotegerin expression on DM with periodontitis. Materials and Methods: The research was an experimental laboratories post test only control group design. Twenty Wistar rats were divided into 5 groups. K0 was negative group while K1 was positive group K1-K4 groups were induced with 65mm/kg STZ single dose and 2ml bacterium P. gingivalis., K2 was treated with 3%golden sea cucumber powder in gel form. K3 was treated with OHB therapy 2.4 ATA for 7 days and K4 given a combination of both. Osteoprotegrin expression on osteoblast of alveolar mandible bone were examined by immunohistochemical staining. Data were analyzed by Kruskal-Wallis and Mann-Whitney test. Result: Kruskal-Wallis result showed significant differences from each treatment group (p <0.05). Mann-Whitney test showed the decreasion in the expression of osteoprotegerin between K1(2:50 ± 0.577) compare with K0 (p<0.05). Golden sea cucumber powder, HBO therapy and both combination had increased osteoprotegerin expression.significancy in K2(8.25 ± 1.258), K3 (5.75 ± 0.957) and K4(12.50 ± 2.082) (p<0,05). Conclusion: Stichopus hermanii powder 3% and 2.4 ATA OHB therapy for 7 days increased the expression osteoprotegrin on DM with periodontitis.

Abstract

Background: Porphyromonas gingivalis is the dominant bacteria that found in chronic periodontitis. Conventional antibacterial (antibiotic) especially tetracycline mostly used for periodontal treatment, but antibiotic ussage often causes resistant, allergy, and toxic. Shark Liver Oil Cenctrophorus sp species have antibacterial effect through its contents, squalene and squalamine that can be developed as an antibacterial adjuvant therapy in periodontal disease. Purpose: To examine the inhibition effect of Shark Liver Oil to the growth of Porphyromonas gingivalis. Materials and Methods: This study used a research design post test only control group design. The research subject is Porphyromonas gingivalis strain ATCC 33277 which have total amount 30 samples, divided into 5 groups. Three groups were given the shark liver oil with different concentrations of 10%, 15%, and 20%. Positive control were given the tetracycline and negative control were given the DMSO 1%. The inhibition was examined using diffution method and was inoculated on MH agar. The inhibitory effect has been observed by measuring the diameter of clear area on the disk using digital calipersin milimeter. Result: Shark liver oil could inhibit the growth of Porphyromonas gingivalis in every concentrations. Concentration 10% with 9,06 mm, concentration 15% with 11,31 mm, concentration 20% with 13,2 mm, while the negative control with 6,01 mm, and positive control with 30,09 mm. Conclusion: Shark liver oil could inhibit the growth of Porphyromonas gingivalis.

Abstract

Background: Osteomyelitis is an bone infection caused by bacteria of Staphylococcus aureus. The use of clindamycin within long term may cause of resistance. A compound in Nannochloropsis oculata’s extract contain antibacterial, antiinflamation, antioxidant that can be used as therapy of osteomyielitis. Purpose: Knowing comparison of the effectiveness between the combination of clindamycin and Nannochloropsis oculata’s extract to increasing the density of collagen on the mandibular osteomyelitis. Materials and Methods: The experimental research’s type with a design post test only control group. Fourty male wistar rats as model of mandibular osteomyelitis has made by inducing bacteria of Staphylococcus aureus inserted in socket after an extraction of left incisive teeth on lower jaw and waiting for 28 days. The rats are divided into 5 group: negative control (K1), clindamycin (K2),clindamycin with 50mg Nannochloropsis oculata’s extract (K3), clindamycin with 100mg Nannochloropsis oculata’s extract (K4), clindamycin with 200mg Nannochloropsis oculata’s extract. After 7 days, all of group are euthanazed and then the density of collagen measured using histopathology slide used Masson’s Trichom and 100x enlargement, so then analyzing the density of collagen by Kruskal-Wallis and Mann-Whitney. Result: There were significant differences in collagen between K1 and K2 (p=0,037), K1 and K3 (p=0,013), K1 and K4 (p=0,002), K1 and K5 (p=0,000). and there were significant differences in collagen between K2 and K5 (p=0,013). But there were no significant differences in collagen between group K2 and K3 (p=1,000), K2 and K4 (p=0,288). Conclusion: The combination of clindamycin with 200mg Nannochloropsis oculata’s extract having an effect on increasing number of the density of collagen significantly on the healing of mandibular osteomyelitis.

Abstract

Background: Ulcers are common and cause pain, difficulty in speaking, eating and swallowing. Avicennia alba role in the process of wound healing and has been shown to accelerate the healing process of gastric ulcer. It contains flavonoid, saponin, NSLcomponents, minerals and vitamin C, which plays a role in wound healing. Purposes: To prove the effect of ethanol extract of Avicennia alba consentration 10%, 20%, 40% against traumatic ulcer. Materials and Methods: The subjects of this research were 30 wistar rats that were randomized into 5 different groups; K0, K1, P1, P2 and P3. (K0) control, (K1) were given hyaluronic acid, (P1) were given extract of Avicennia alba 10%, (P2) were given extract of Avicennia alba 20%, and (P3) were given extract of Avicennia alba 40%. The subject was wounded using amalgam stopper that has been heated before. Subject was given topical application once daily. The ulcer’s diameter was measured at the second day until the day cured using digital caliper. Data were analyzed using One way ANOVA test. Result: The result showed signification of p<0,05, showed that there were difference diameters betweentwo groups. The average diameter differences of traumatic ulcer among rat are: 0,75 mm (K0), 1,51 mm (K1), 1,01 mm (P1), 2,02 mm (P2), and 1,70 mm (P3). The results showed statistically significant value of p>0.05 which there was no difference in effectivenessAvicennia alba extract in the K1 group. Conclusions: Ethanol extract of Avicennia alba 10%, 20%, and 40% have effect in the healing of traumatic ulcer. The effectiveness of those were the same as giving a 0.2% hyaluronic acid. The concentration 20% of Avicennia alba was effective to the healing of traumatic ulcer
