Abstract

Patogen tanaman merupakan salah satu faktor utama penyebab penurunan produktivitas pertanian dan kerugian ekonomi yang signifikan di berbagai sistem budidaya. Identifikasi patogen secara cepat dan akurat menjadi langkah penting dalam pengendalian penyakit tanaman, khususnya untuk mendukung kegiatan karantina, pemantauan penyakit, serta penentuan strategi pengelolaan yang tepat. Metode diagnosis konvensional yang bergantung pada pengamatan gejala, isolasi, dan kultur patogen umumnya memerlukan waktu relatif lama dan bergantung pada keahlian peneliti, sehingga berisiko menimbulkan kesalahan identifikasi, terutama pada tahap awal infeksi ketika gejala belum berkembang secara spesifik. Perkembangan teknologi molekuler mendorong penggunaan pendekatan identifikasi berbasis DNA yang menawarkan sensitivitas dan spesifisitas lebih tinggi dibandingkan metode konvensional. Tinjauan ini membahas berbagai metode identifikasi patogen tanaman berbasis DNA, meliputi teknik polymerase chain reaction (PCR) dan turunannya, seperti nested PCR, multiplex PCR, quantitative PCR (qPCR), nano-PCR, dan droplet digital PCR (ddPCR), serta metode amplifikasi isotermal seperti loop-mediated isothermal amplification (LAMP), recombinase polymerase amplification (RPA), dan nucleic acid sequence-based amplification (NASBA). Teknologi lanjutan seperti next-generation sequencing (NGS) dan sistem CRISPR/Cas juga dikaji berdasarkan prinsip kerja dan potensi penerapannya dalam diagnosis patogen tanaman. Hasil tinjauan menunjukkan bahwa setiap metode memiliki keunggulan yang berbeda, di mana qPCR dan ddPCR unggul dalam sensitivitas dan presisi untuk deteksi target berkonsentrasi rendah, LAMP dan RPA menawarkan kecepatan analisis serta kemudahan aplikasi di lapangan, sementara NGS dan CRISPR/Cas memberikan resolusi tinggi untuk deteksi komprehensif dan diagnosis dengan spesifisitas sangat tinggi. Tinjauan ini menekankan pentingnya pengembangan metode diagnostik berbasis DNA yang efisien dan aplikatif untuk mendukung pengelolaan penyakit tanaman dan sistem pertanian berkelanjutan.

Abstract

Biology education at the high school level frequently faces conceptualization constraints, where students receive microbiology material theoretically without direct connectivity to their surrounding ecosystems. The lack of microscope utilization skills and inadequate laboratory facilities are often the main obstacles for teachers in designing practical work. This community service program aims to improve the competence of 35 teachers at SMA Negeri 3 Menggala, Tulang Bawang Regency, in conducting proper microscopic observations of bacteria and fungi using samples from the surrounding environment. The activity was conducted on January 28, 2026, through a participatory-educative approach comprising theoretical presentations, demonstrations of making fresh preparations, and microscopic observation practices. Evaluation results using pre-tests and post-tests showed a significant increase in participants' cognitive understanding and technical skills, jumping from an initial 45% to 87%. Participants' enthusiasm was remarkably high during the activity. The school principal felt greatly assisted by this program as it provided practical solutions to optimize contextual and discovery-based biology learning without relying on expensive manufactured practicum kits .

Abstract

White turmeric (Curcuma zedoaria) is one of the strategic bio-pharmaceutical plants commonly used as a cooking ingredient and as a family medicinal plant. In the Lampung region, turmeric ranks among the top five highest-yielding bio-pharmaceutical crops, making it an abundantly available commodity. This research aims to explore the endophytic bacteria present in turmeric and to investigate the functional abilities of the isolated strains. Endophytic bacteria are microorganisms that inhabit plant tissues and can act as plant growth promoters. In addition, these bacteria can solubilize phosphate, thereby supporting plant growth. Endophytic bacteria also produce the phytohormone IAA, which plays an important role in plant development. This study obtained three endophytic bacterial isolates from the leaves of white turmeric plants, namely KPD1, KPD2, and KPD3. Isolates KPD2 and KPD3 exhibited phosphate-solubilizing activity, as indicated by the formation of clear zones measuring 2.5 ± 0.58 mm and 4.0 ± 0.82 mm, respectively. In addition, all isolates were able to produce indole-3-acetic acid (IAA), with concentrations of 54.98 µg/mL (KPD2), 54.14 µg/mL (KPD3), and 25.05 µg/mL (KPD1). The abilities demonstrated by these endophytic bacterial isolates indicate their potential as plant growth-promoting bacteria (PGPB).
