Abstract

Acinetobacter baumannii is a multidrug-resistant pathogen causing severe nosocomial infections, primarily associated with alterations in penicillin-binding proteins (PBPs) that reduce the effectiveness of β- lactam antibiotics. Despite increasing interest in plant-derived bioactive compounds, the potential of oxygenated sesquiterpenes from Aquilaria malaccensis as inhibitors of PBPs remains underexplored, indicating a clear research gap. This study aimed to evaluate the antibacterial potential of three oxygenated sesquiterpenes—Agaruspirol, Eudesmol, and Sinenofuranol—against PBP1a (3UE3) and PBP3 (3UDF). A systematic literature review (SLR) integrated with an in silico approach was conducted, including ligand retrieval from PubChem, ADMET prediction using ADMETlab 3.0, molecular docking using CB-Dock2, interaction analysis with UCSF ChimeraX and PLIP, and antibacterial activity prediction using PASS Online. The results showed that all compounds satisfied Lipinski’s rule of five and exhibited favorable ADMET profiles. Molecular docking demonstrated stable interactions with both PBPs, dominated by hydrogen bonding and hydrophobic interactions. Agaruspirol exhibited the strongest binding affinity toward PBP3 (–6.9 kcal/mol) and PBP1a (–6.5 kcal/mol), along with the highest predicted antibacterial activity (Pa = 0.364; Pi = 0.039). These findings suggest that oxygenated sesquiterpenes, particularly agaruspirol, have potential as lead compounds for the development of antibacterial agents targeting PBP-mediated resistance in A. baumannii, and provide a scientific basis for further in vitro and in vivo validation.

Abstract

Gaya hidup perilaku remaja yang kurang  baik dapat berpengaruh terhadap kesehatan terutama kebersihan daerah kewanitaan karena dapat terinfeksi salah satunya Trichomonas vaginalis. Trikomoniasis merupakan infeksi yang disebabkan oleh protozoa Thricomonas vaginalis dan merupakan infeksi menular seksual non-virus paling umum ditemukan diseluruh dunia. Kegiatan ini dilaksanakan bulan Agustus dan sebanyak dua kali. Sampel dari kegiatan ini yaitu siswi SMK BIM PPMU Mojosari sebanyak 15 orang. Adapun metode dalam kegiatan ini yaitu ceramah dan pemeriksaan urine. Hasil kegiatan ini yaitu sebanyak 46,67% responden diikuti oleh berumur 15 tahun; 80% responden memiliki perilaku menjaga kebersihan organ kewanitaan dengan baik; 53,3% responden mengerti cara membasuh organ kewanitaan; 66,67% responden memiliki perilaku menjaga kesehatan organ kewanitaan dengan baik sehingga hasil pemeriksaan urine tidak ditemukan adanya Trichomonas vaginalis sebanyak 100%. Kesimpulan dari kegiatan ini yaitu sebagian besar siswi SMK BIM PPMU memiliki pola hidup dalam menjaga kebesihan dan kesehatan organ kewanitaan dengan baik sehingga dalam tidak ditemukan adanya infeksi parasit tersebut.

Abstract

Abstract : Gout is one of the non-communicable diseases commonly found in the community and is closely associated with dietary patterns and lifestyle. Limited public knowledge regarding risk factors and preventive measures contributes to suboptimal gout management. This community service activity aimed to improve community knowledge on the prevention and management of gout through education on healthy dietary patterns and lifestyle modification. The activity employed a one-group design with health education delivered through interactive lectures, discussions, and educational media, conducted in the Tenggilis Mejoyo area, Surabaya. Knowledge evaluation was carried out using pretest and posttest questionnaires consisting of ten questions assessing understanding of gout, risk factors, high-purine foods, and the role of diet and lifestyle in gout prevention. The results showed an improvement in participants’ knowledge after the educational intervention. Prior to the activity, most participants had low (56.3 percent) and moderate (34.4 percent) knowledge levels; following the intervention, the proportion of participants with low knowledge decreased to 18.8 percent, while moderate and good knowledge levels increased to 53.1 percent and 28.1 percent, respectively. Participants also demonstrated better understanding of high-purine foods, the importance of physical activity, and sustainable gout management. In conclusion, community-based health education is effective as a promotive and preventive approach to improving public knowledge on gout prevention and management.

Abstract

Intestinal worm infections remain a public health problem in tropical areas, especially those caused by Soil Transmitted Helminths (STH) with the species Ascaris lumbricoides, Trichuris trichiura, and Hookworm. Accurate stool examination methods are essential to support diagnosis and control programs. Objective: This study aimed to evaluate the effectiveness of variations in centrifugation speed with the 15% formaldehyde sedimentation method–5% detergent in detecting STH eggs. Method: The study was conducted at the Pharmaceutical Biology Laboratory of Anwar Medika University, Sidoarjo with a descriptive experimental design. Three types of suspension samples were examined with 92 replications. Microscopic examination used the 15% formaldehyde sedimentation method–5% detergent with variations in centrifugation speed of 2000, 3000, 4000, and 5000 rpm for 5 minutes. Identification of worm eggs was carried out based on the number and typical morphology of A. lumbricoides, T. trichiura, and hookworm. Data analysis used the Shapiro-Wilk test (p = 0.05) to assess initial differences between groups, then continued with ANOVA (p = 0.05) if the data met the assumptions of normality and homogeneity. Results: The results showed that variations in centrifugation speed affected the number of eggs detected. A speed of 3000 rpm for 5 minutes provided the most optimal results with higher sensitivity compared to other variations. In Data Analysis The data normality test using Shapiro-Wilk showed that all groups of centrifugation speed variations had a normal distribution (p > 0.05). Furthermore, the results of the homogeneity of variance test with Levene's Test also showed that the data were homogeneous (p > 0.05). Based on the ANOVA test, a p value <0.05 was obtained, indicating a significant difference between variations in centrifugation speed and the number of worm eggs detected. Further analysis showed that a speed of 3000 rpm for 5 minutes produced a more optimal number of eggs compared to other speed variations (2000, 4000, and 5000 rpm). Conclusion: The 15% formaldehyde–5% detergent sedimentation method with a centrifugation speed of 3000 rpm for 5 minutes increases the sensitivity of stool examination for intestinal worm infections, so it can be used as a more accurate alternative method for laboratory diagnosis.
