Nypa fruticans Wurmb. is a rich source of bioactive phytochemicals, including flavonoids, phenolic, and tannins. This study aimed to evaluate the antioxidant activity of Nypa fruticans Wurmb. leaf extract and investigate its potential as a bioreducing agent for the green synthesis of AgNPs. The leaf extract was prepared using the maceration method, while antioxidant activity was determined using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. AgNPs were synthesized via a precipitation-based green synthesis approach and subsequently characterized by UV–Visible spectroscopy to monitor the formation of the surface plasmon resonance band. The extraction process yielded a crude extract with a recovery of 17%. The DPPH assay demonstrated a strong antioxidant activity, with an IC₅₀ value of 7 μg/mL. Optimization of the biosynthesis process revealed that an extract volume of 7 mL, a reaction temperature of 50 °C, and a reaction time of 90 min were the optimal conditions for the reduction of Ag⁺ ions, resulting in optically stable AgNPs. The successful formation of AgNPs was confirmed by an increase in SPR intensity accompanied by a red shift in the absorption wavelength, indicating nanoparticle growth. These findings suggest that Nypa fruticans Wurmb. is not only an effective source of natural antioxidants but also a promising eco-friendly bioreducing agent for the green synthesis of silver nanoparticles.
