Gayo buffalo is a source of local livestock germplasm that has the potential to support local resource-based food security. This study was conducted to identify polymorphisms of the Gayo buffalo growth hormone gene. The PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism) procedure was developed to determine the polymorphism of the growth hormone gene. DNA was isolated from blood samples by the salting out method. The 7 buffalo blood samples used from Central Gayo were carried out by PCR technique using the HaeIII restriction enzyme. DNA isolation was carried out using a pair of primers 5'-GTA AGG ATG GCT CTG CCA GGT3' and GH reverse 5'-TGC ATG ATG CTG TCC CTC TGG A-3', The PCR product was digested by the HaeIII restriction enzyme. The amplification result was a specific single band with a 451 bp fragment and produced 100% AA genotype where the GH gene in Gayo buffalo is monomorphic. This initial genetic information can be used as a basis for managing and preserving Gayo buffalo genetic resources in supporting the sustainability of the provision of animal-based food based on local livestock.
