Background: Faloak (Sterculia quadrifida R.Br.) is a medicinal plant native to East Nusa Tenggara that has potential as an antioxidant. Purpose: To determine total flavonoid, tannin, and phenolic contents of 95% ethanol and aqueous extracts of faloak root bark, stem bark, and twigs, and to evaluate antioxidant activity of selected extract. Method: This quantitative descriptive-exploratory research used maceration (95% ethanol) and decoction (water) methods. Metabolite contents were determined spectrophotometrically, while antioxidant activity was tested using the DPPH method ( ). Results: Highest secondary metabolite content was consistently found in the twigs. In the 95% ethanol extract, flavonoid, tannin, and phenolic contents of twigs were 0.7495, 38.8472, and 94.2777 mg/g extract, respectively, while in aqueous extract, they were 0.8093, 31.6775, and 59.7342 mg/g extract. 95% ethanol extract was selected as superior extract because it yielded higher metabolite levels than aqueous extract. DPPH testing showed values for root bark, stem bark, and twigs of 26.502, 36.110, and 21.524 ppm, respectively, classifying all samples into very strong antioxidant category. Conclusion: Flavonoid, tannin, and phenolic contents vary depending on the plant part and solvent type. Faloak twigs possess highest metabolite content and most potent antioxidant activity.
