Abstract

Experiment on the enlargement of individual mud crab Scylla serrata which caught by fisherman in the waters of Wael District still under economy worthy price. The purposes of this study is to see the crab growth rate through one of the crab fi sheries management strategies, there are maintenance and fattening. The weight size of individual mud crab used in this study between 100 gr to 325 gr ind-1. Male mud crabs was separated from females as a fattening treatment experiment and was placed on the two bamboo cages which each contain of 15 individuals then placed separately. Observation was made along 4 months. The result showed that the growth pattern of male and female mud crabs is still exponential, which male individu reach on average 532.3 gr weight change ind-1 and female with 499 gr ind-1 in 4 months. It concluded that female mud crabs has slower growth rate than males, and the fattening not reached an asymptotic value, which means they still can be fattened again in the cage.

Abstract

Female and male brooders were vaccinated by intraperitoneal injection and intramuscular injection respectively. Unvaccinated brooders were used as control. Fry which was produced by vaccinated and unvaccinated brooders were reared separately in paddy field pond for 15 days for the first nursery rearing period and another 15 days for the second nursery rearing period. These field experiment conducted in four locations, namely slmean (2 location) and magelang (2 location). Results of these experiments indicated that brooders vaccination could increase survival rates of the first and the second nursery rearing periods approximately 65.00% and 3.69% respectively. Average weight and total length of fry which produced by vaccinated broodstock increased 12.59% and 58.51% during the first nursey rearing period, 8.55% and 7.14% during the second nursery period.

Abstract

Acoustic technology for fisheries is one of technologies with the advantages to detect marine resources including estimating density and the presence of fish and relatively more accurate than conventional methods. The present study aims to assess the effectiveness of the operation of the lift-net with hydroacoustic technology in an effort to increase the productivity of the fisherman of boat liftnet. The study was conducted on fishing activity of boat liftnet in the Makassar Strait from April to May 2015 by utilizing the tools of acoustic technology in the operation of the boat liftnet. The analysis method is based on the productivity of the boat liftnet. The results showed that the productivity of fishing gear which is operated by using the tools of fisheries acoustic were improved compared with the fishing effort without acoustic tools. This result is expected to be a reference to research on the use of acoustic technology in a variety of fishing gear and as reference to research the use of science and technology in support of sustainable small-scale fishing effort.

Abstract

During January 2005 the holothurian community inhabiting littoral areas was assessed at five station on four ecological indices: density, species richness, diversity and evenness. It was revealed that the holothurian community consisted of 8 species in which three among them occurred in all locations at higher densities: Holothuria edulis, H. scabra and H. atra. Small variations of the ecological indices found in all location might indicated that the community characteristics among locations were relatively similar.

Abstract

The study aimed to determine the effect of pH and temperature on N-Acetylglucosamine (NAG) formation from shrimp shell chitin by Serratia marcescens PT-6 fermented in chitin broth medium. Parameters examined on NAG formation include bacteria growth, chitinase activity (U/ml), and NAG concentration in medium (mg/ ml). Bacteria growth was measured by turbidity (OD600), while chitinase activity and NAG concentration in the medium were analyzed quantitatively by colorimetric assay. The variation of initial pH examined in chitin broth medium was 5; 6; 7; 8, while temperature variation was 30°C; 37°C; dan 40°C. The results show that maximum concentration of NAG formed by Serratia marcescens PT-6 was 33.86 µg/ml on day-3 of fermentation, at pH 7 and temperature of 30°C. Chitinase activity on the same day was 0.002 U/ml, while OD600 value of the culture was 0.42 indicating the bacteria were in the log phase. This research implies that Serratia marcescens PT-6 is potential to be optimized further for the bioconversion process of NAG from shrimp shell waste through an enzymatic method by modifying its culture condition.

Abstract

The study of coral reefs in Rajaampat archipelago, district of Sorong, was carried out in September-October 2001, located at Boni Islands, Ayu Islands and Batang Pele Islands. The aims of this study were to know the percentage of live coral cover, coral diversity and species numbers of coral reefs in this area. Observations on coral reefs were conducted by applying the Rapid Reef Resource Inventory (RRI) and Line Intercept Transect (LIT). Results of Rapid Reef Resources Inventory (RRI) technique showed that the averages of live coral cover on the reef top and reef edge were 6.14 and 15.39% in Boni island (East Waigeo); 6.28 and 24.67% in Ayu island; 25.36 and 23.81% in Batang Pele Islands, respectively. Result of Line Intercept Transect (LIT) technique showed that the percentages of live coral cover were 35.02% in Boni Island, 51.07% in Ayu Island, and 40.86% in Batang Pele Islands. The value of evenness indexs were 0.872-0.934 in Batang Pele Islands; 0.758-0.861 in Ayu Islands; 0.805 in Boni Islands. The total number of species in three sites was 293 species, with the total numbers of spesies in each station were 153 species in Boni islands, 277 species in Ayu islands, and 205 species in Batang Pele Islands. The condition of coral reef at Ayu islands was categorized as good condition, while both of Batang Pele Islands and Boni Islands were categorized as moderate condition.

Abstract

An investigation into fecundities and eggs diameter of snakehead (Channa striata Bloch) in flood plain area of river basin of Musi, South Sumatera has been done from July to December 2002. The fish samples were collected from the fisherman. After measuring body length and weight, the gonads were collected and their maturation were observed. The fecundity counted by gravimetric method. Eggs diameter of fish which have TKG III, IV and V were also observed. The result showed that snakehead in the flood plain area of Musi river have fecundities ranged from 1141 to 16486 eggs with the body weight ranged from 60 to 640 g, gonad weight ranged from 1,15 to 17,04 g, and eggs diameter ranged from 0,65 to 1,34 mm.

Abstract

Monogenean is a parasite group which most often causes serious disease in fish due to its direct life cycle, high reproductive potential, and high pathogenicity. The aim of this study was to investigate infection level of Thaparocleidus sp.onstriped catfi sh which infected catfi sh fingerling. Striped catfi sh fi ngerling with 5–10 cm in total length samples from three diferent ornamental fish supplier in Makasar were  examined. Sampling was conducted weekly for three weeks and 10 fishes from each locations were collected. Samples were transported to laboratorium with aerated plactic bags and transported to Laboratory of Fish Parasite and disease for parasite examination. Organ which examined were gills. Prevalence of Thaparocleidus sp infection in all locations sampled was 100%. Mean intensity of the parasite infection was 98, 58 and 70 in location I, Location II, and Location III, respectively. The high prevalence and mean intensity of Thaparocleidus sp. infection indicated that health management in the aquaculture facilities was not properly implemented and this parasite existance is one of aquaculture problems to develop catfi sh fulfi lling ornamental or consumption demand .

Abstract

The experiment was carried out to evaluate the water and soil quality of pond, which were subjected to drying and aeration after inundation. The objective of the experiment was to find out which soil drying and water aeration treatments were the best for shrimp culture.The soil used was obtained from an intensive shrimp pond and sea water was collected  from pond area in jepara coast. The soil drying treatments consisted of drying the wet soil under the sun to air dry condition, field capacity condition and water saturated condition. The soil drying treatments were carried out for 64 days. The aeration treatments after inundation consisted of with aeration and without aeration. Soil less sea water also was prepared as control. The aeration treatments were carried out for 64 days. The soil and water quality were  observed during soil drying and water aeration treatments.Result of this experiment indicated that drying the pond bottom soil until field capacity condition gave the highest rate of organic matter decompotition followed by air dried soil, then water saturated oil. The decompotition of soil organic matter in the field capacity condition reached optimum on day 8 to 16. The pond bottom soil in water saturated condition contained lower NH3, NO2- and H2S than ind dried soil. Aeration gave better condition than the non aeration one. It seemed the best water quality condition for shrimp fry stocking occurred 4-8 days after inundation combined with aeration.

Abstract

Tujuan dari penelitian ini adalah untuk mengetahui aktivitas enzim pendegradasi kitin yang dihasilkan selama pertumbuhan dari lima isolat bakteri kitinolitik (SDI23, MDR23, SDI15, SDI13 dan BLT12) asal petis udang serta mengetahui karakteristik aktivitas enzim hasil pemurnian parsial pada berbagai suhu (30-55°C) dan pH (4-9). Kelima isolat ditumbuhkan pada medium koloidal kitin cair dengan suhu inkubasi 37°C selama 96 jam. Pengamatan terhadap jumlah bakteri (log CFU/ml) dan aktivitas spesifik enzim (U/mg) dilakukan setiap 12 jam. Aktivitas enzim pendegradasi kitin diukur secara kuantitatif, dengan mengukur jumlah N-asetilglukosamin yang dihasilkan dari hidrolisis koloidal kitin oleh enzim. Purifikasi parsial dilakukan dengan presipitasi ammonium sulfat bertingkat (20-80%) yang dilanjutkan dengan dialisis. Isolat SDI23, MDR23, SDI15 dan BLT12 menunjukkan waktu pertumbuhan optimum dengan jumlah sel tertinggi pada jam ke-48, sedangkan isolat SDI13 pada jam ke-24. Aktivitas spesifik enzim tertinggi ditunjukkan oleh isolat SDI23 sebesar 0,66 U/mg pada inkubasi selama 48 jam. Berdasarkanhasil tersebut, isolat SDI23 dipilih untuk memproduksi enzim pada tahap purifikasi parsial. Supernatan bebas sel dari media kultur isolat SDI23 disiapkan dari inkubasi SDI23 dalam medium koloidal kitin cair pada suhu 37oC selama 48 jam. Presipitasi dengan amonium sulfat pada fraksi 80% dan dialisis menghasilkan ekstrak kitinase kasar dengan nilai aktivitas spesifik enzim sebesar 0,072 U/mg dan 0,063 U/mg. Enzim hasil pemurnian parsial menunjukkan aktivitas optimum pada pH 6 dan suhu 45°C dengan nilai aktivitas spesifik enzim sebesar 0,09 U/mg.
