Abstract

Disease caused by the bacterium A. hyrophila known as MAS (Motile Aeromonad Septicaemia) decreases survival and growth in Catfish. Phytopharmaca is a potential solution to replace antibiotics. One of the phytopharmaca which has the potential to be effective, inexpensive, safe for humans, and environmentally friendly is avocado (Persea americana) with active compounds such as alkaloids, flavonoids, saponins, and tannins. The potential for prevention of bacterial diseases with avocado leaf extract on the clinical symptoms of fish in MAS disease needs to be scientifically proven to be applicable to aquaculture economically and environmentally sustainable. This study uses an experimental method with several stages, namely testing LD50, in vitro and in vivo tests. The LD50 concentration of A. hydrophila is at a density of 102 cfu/ml. The filtrate of avocado leaf maceration showed that a clear zone diameter is equivalent to penicillin in the in-vitro test. Fortification of avocado leaf extract on feed is quite effective in preventing MAS disease in the in-vivo test. The fastest of MAS disease clinical symptoms recovery is at a dose of 100 ppt and 300 ppt on the 7th day. The highest Survival Rate (SR) was found at a dose of 100 pp (100%) which was equivalent to negative control and followed by a dose of 200 ppt (73.33%), 300 ppt (66.67%) and positive control (46.67%).

Abstract

Giant freshwater prawn is one of Indonesia's freshwater prawn that has advantages such as fast growth and good meat quality. The continuous crossing of giant freshwater prawn that are closely related causes a decrease in its genetic diversity. In addition, the domestication process can also influences changes in the number and shape of chromosomes. This study is to determine the chromosome characters of domestication giant freshwater prawn (Siratu)  and those derived from natural catches (Mahakam) which are cultured in the Unit Kerja Budidaya Air Payau Balai Budidaya Udang Galah (UKBAP BBUG) Samas. Research on the characterization of chromosome of giant freshwater prawn from both natural and domesticated populations needs to be done to obtain genetic information, especially chromosome characters. This information is useful to further support the process of breeding and for the certification process of giant freshwater prawn. Chromosome length measurements were carried out using the Image Raster 3 program. The shape of the chromosomes was determined by following the classification of Levan et al. (1994) based on the Sentromer Index Value obtained. The results showed that the number of chromosomes in the population of Siratu and Mahakam were 2n = 118. The giant freshwater prawn of Siratu population had the karyotype formula 2n = 118 = 94 m + 24 sm, while the giant freshwater prawn Mahakam population had the karyotype formula 2n = 118 = 118 m.

Abstract

Microalgae diversity research in the Lok Ulo River in Kebumen Regency, Central Java was conducted in September 2018. This study aimed to determine the diversity of microalgae in the Lok Ulo river. This research was conducted by field survey method. Determination of water sampling was chosen from 3 stations based on considerations to represent waters including: Station I (upstream part of the river), station II (middle part of the river) and station III (downstream part of the river). Each of the three water samples is taken. Water samples are taken to environmental laboratories for physical, chemical and biological analysis. The results showed that microalgae species found in the upstream, middle and downstream of the Lok Ulo river numbered 9 species, namely Oscillatoria sp., Cymbellahelvetica, Cymbellatumida, Cymbellanaviculiformis, Diatomaelongatum, Fragilaria sp, Synedraaccus, Synedraaffanis, Tabellaria sp. The relationship between TDS on microalgae diversity is shown through linear equations Y = 2.404-0.00014X. Based on these equations, TDS (X) in the Lok Ulo river is able to influence the diversity of microalgae with TDS is negative, every increase of 0.1 mg / l, TDS will reduce diversity by 0.00014 microalgae. The magnitude of the effect of TDS on the diversity of microalgae is indicated by the value of the correlation coefficient (r) of 0.972 with a significance of 0.000 there is a TDS relationship with diversity. The closeness of the relationship between TDS and the diversity of microalgae is indicated by the coefficient of determination (R2) of 94.5%. This means that the magnitude of the diversity of microalgae is influenced by TDS with closeness of 94.5.

Abstract

Selulase merupakan salah satu enzim yang digunakan secara luas dalam industri dan kebutuhannya semakin meningkat. Untuk memenuhi permintaan industri, dibutuhkan medium berbiaya rendah untuk produksi selulase. Pemanfaatan limbah dapat menyediakan substrat alternatif dan membantu dalam memecahkan masalah polusi. Selulase dapat dihasilkan melalui proses fermentasi dengan bantuan mikroorganisme yang ditumbuhkan pada substrat yang mengandung selulosa. Penelitian ini menggunakan limbah dari pengolahan kayu jati yaitu serbuk gergaji kayu jati. Jumlah serbuk gergaji dari kayu jati melimpah dan pemanfaatannya belum optimal. Serbuk gergaji kayu jati diketahui memiliki kadar selulosa sebesar 46,5%. Kadar selulosa yang tinggi tersebut menjadikan serbuk gergaji kayu jati berpotensi sebagai substrat dalam produksi enzim selulase. Penelitian ini bertujuan untuk menentukan waktu inkubasi dan moisture content substrat yang optimal bagi Aspergillus niger untuk memproduksi selulase dari substrat serbuk kayu jati. Uji enzim dilakukan dengan metode CMCase. Hasil penelitian menunjukkan bahwa waktu inkubasi yang optimal untuk produksi enzim selulase oleh Aspergillus niger adalah 48 jam dengan kadar selulase 266,303 mg/L sedangkan moisture content substrat yang optimal adalah 50% dengan kadar selulase  197,394 mg/L.

Abstract

Salah satu daerah dengan aktivitas manusia yang memiliki potensi sebagai habitat bagi amfibi adalah lingkungan kampus Universitas Gadjah Mada (UGM). Penelitian ini bertujuan untuk mengetahui keanekaragaman, kemelimpahan, dan pola distribusi amfibi di wilayah kampus UGM di awal hingga pertengahan 2018. Metode yang digunakan dalam penelitian ini meliputi pendataan Anura dengan teknik sampling VES (Visual Encounter Survey) disertai tagging lokasi, kemudian dilakukan morfometri dengan 13 parameter. Data yang diperoleh kemudian ditabulasi.. Berdasarkan hasil penelitian, keanekaragaman dan kemelimpahan meliputi lima species anggota ordo Anura yang terdiri dari Duttaphrynus melanostictus (familia Bufonidae) yakni sejumlah 253 individu, Polypedates leucomystax (familia Rhacophoridae) yaitu 149 individu, Kaloula baleata (familia Microhylidae) yaitu 20 individu, Chalcorana chalconota (familia Ranidae) yaitu 39 individu, dan Fejervarya cancrivora (familia Dicroglossidae) yaitu 1 individu. Pola distribusi secara luas didominasi oleh D. melanostictus.

Abstract

This study aims to determine the biodiversity of butterflies, the relationship between the number of butterflies with the number of visitors and the strategy of butterfly conservation needed to manage the diversity of butterflies in Nature Recreational Park, Pananjung. This research was conducted by field survey method. Parameters observed at each site include vegetation diversity, direct collection and observation of butterflies. The parameters observed at each location include the index of diversity (H '), and regression analysis. Samples are taken from ecosystem which is divided into three stations ie many visitors station, station little visitors, station no visitors as much as thirteen times. The study was conducted at the end of December 2016 - end of March 2017 for 3 Months. The results of the study found 59 species of 1165 individuals. The variety of butterflies including high category (H '= 3.651) with fairness is still relatively high (E = 0.887). In addition, the amount of influence the number of visitors to the number of butterflies (r square) 0,398 or 39,8 % with regression equation is: yaitu :  Y = -0,0000079X3 + 0,005X2 – 0,828X + 44,083. The more visitors who come to ecosystem the more will decrease the number of butterflies. Furthermore, the conservation strategy that is needed in ecosystem is need to do in situ butterfly capture from Nature Recreational Park, Pananjung because of the discovery of endemic species of Tanaceia trigerta and rare species, Troides helena.

Abstract

Estuarine crocodile often considered as dangerous animal due to continual news of human crocodile conflict cases around the world.  This study aimed to analyze the perception of visitors about estuarine crocodile in captivity and facilities in two captive breeding sites. We conducted field observation and structured interview to 120 respondents in two local zoos: the Indonesia Jaya Crocodile Park (TBIJ or crocodile park)  in Bekasi and Ragunan Zoo (TMR or the zoo) in Jakarta. All visitors have low knowledge about estuarine crocodile, which they obtained mostly from mass media. Although most visitors know that estuarine crocodies are protected however in pratice their don’t have enough empathy to captive crocodiles. Visitors to both areas are local who lives near the vicinity of the park or zoo. Crocodile park visitors mostly came from Bekasi with an average number of visits 1 time and 1-hour length of visit. The reason for their visit were the closeness of the location and to learn crocodiles. The zoo visitors mostly came from Jakarta with an average number of visits more than 3 times and 6 hours length of visits. Respondents visited the zoo because of low cost and recreation. Visitors of the zoo provided  positive review for several aspects in the zoo, while visitors in crocodile park gave positive review only on physical condition of crocodiles.

Abstract

Pulai (Alstoniasp.), Kayumanis/Cinnamon (Cinnamomum burmanii, Cinnamomum iners), Pakanangi/Kisereh (Cinnamomum parthenoxylon/Cinnamomum porrectum), Kulilawang (Cinnamomum halmaherae), Agarwood (Aquilaria sp., Gyrinops versteeghii and Gyrinopsis cumingiana), and Pasakbumi (Eurycoma Longifolia) are some of  medicinal plants species which growing in plantation or natural forest. The exploitation to these species causes their existence to be scarce or almost not known, with the result that is needed a policy to save the existence of medicinal plants species. Industries of herbal medicine which exploit the raw material from nature need to build seedlings and enrichment area. The collaboration between medicinal industries with local people to build medicinal plants species plantation can be developed. This paper informs a field review of some medicinal plants species from several regions during the research activities in 2005 to 2009.

Abstract

Penelitian ini dilatar belakangi oleh belum terdatanya jenis-jenis pohon dan informasi ekologis hutan perlindungan di kawasan Bukit Cogong. Penelitian ini bertujuan untuk  mengetahui Indeks Keanekaragaman (H’) spesies vegetasi strata pohon dan mengetahui faktor lingkungan abiotik di areal hutan perlindungan kawasan Bukit Cogong. Penelitian ini menggunakan metode Point Centered Quarted, yaitu salah satu metode pengukuran yang tidak menggunakan plot atau metode plotless. Penelitian ini di buat 3 area kajian yaitu area kajian A, area kajian B, dan area kajian C. Secara keseluruhan luas area penelitian yaitu 17 ha atau 170.000 m2 yang diambil 20% dari keseluruhan luas areal hutan perlindungan yaitu 85 ha atau 850.000 m2, sehingga luas masing-masing area kajian 56.666m2. Pada masing-masing area kajian diletakkan 10 stand. Pada setiap stand penelitian diletakkan garis transek utama sepanjang 560 m. Berdasarkan hasil penelitian diperoleh rerata di area kajian A sebesar 1,38, area kajian B sebesar 1,14 dan area kajian C sebesar 1,36. Rerata Indeks Keanekaragaman jenis vegetasi strata pohon di areal hutan perlindungan kawasan Bukit Cogong secara keseluruhan tergolong sedang melimpah dengan kriteria H’ ≤1≤ H’ 3 dan faktor lingkungan abiotik yang terukur di area kajian A, B dan C pada areal hutan perlindungan kawasan Bukit Cogong hasilnya tidak berbeda jauh.

Abstract

The nocturnal parthenogenetic species, mourning gecko (Lepidodactylus lugubris), has been recorded in Sleman, Daerah Istimewa Yogyakarta, Java, Indonesia. There has yet no specific information of its distribution in Java and its behaviour. One out of three individues observed was successfully caught. A single specimen collected from The Oriental white-eye (Zosterops palpebrosus) cage is reviewed along with literature reports of the occurrence of the species. It is likely that population of this invasive gecko has been established in the region.
