Abstract

Implementation of practical work in biology laboratory needs supporting media including laboratory equipments and materials. In genetics and molecular biology, the routine materials are mostly molecular resources, which are produced by certain company. To get the materials, we must order and pay for a relative high price.These molecular resources in biology laboratory is not only beneficial for practical activity, but also useful for research activity in related fields.This research works as an application of methods applied in laboratory to result in a product that is valuable in practical works, i.e. the pGEM plasmids. The methods that were employed involved the transformation of E. coli with pGEM plasmid, blue-white screening by using X-gal, and plasmid isolation by alkaline lysis method. The transformation results showed a relative low efficiency at 1,2 x 103cfu/µg DNA. The electrophoresis in 0.8% agarose gel demonstrated that the plasmid DNA was successfully isolated with some thin bands at position 3000-4000bp, despite of a large chromosomal DNA and some smear RNA were still observed. The DNA obtained is pure enough, characterized by the ratio of A260/280 at the range of 1,8-2,0.

Abstract

The purpose of this research to develop environmentally chemistry lab kits to support laboratory of environmental friendly chemistry (green chemistry laboratory). The development refers to the model of Education Research and Development. The product was a chemistry lab KIT containing environmentally friendly lab materials, tools, and practical procedure that can be carried in the classroom. Results validisi expert on chemistry lab procedures showed very valid in either category. The response of students to 95.6% expressed a good product.

Abstract

Penelitian tindakan kelas ini bertujuan untuk mengetahui motivasi dan hasil belajar peserta didik pada mata pelajaran Pendidikan Kewarganegaraan (PKn) sebelum menggunakan media surat kabar dan setelah menggunakan media surat kabar. Subjek penelitian ini adalah peserta didik kelas VII B SMP Negeri 1 Turi tahun pelajaran 2014/2015. Instrumen penelitian yang digunakan berupa lembar observasi, tes tertulis, dan narasi angket. Penelitian tindakan kelas ini mengacu pada Kemmis dan Mc.Taggart, yaitu dilaksanakan dalam 2 siklus dan setiap siklus meliputi 4 tahapan yaitu perencanaan (planning), tindakan (action), observasi (observation), dan refleksi (reflektion). Hasil penelitian menunjukkan bahwa terdapat peningkatan motivasi peserta didik kriteria sangat aktif dan aktif yang awalnya 9% pada pra siklus (sebelum menggunakan media surat kabar) bertambah  menjadi 87% pada siklus II (setelah menggunakan media surat kabar). Hasil belajar peserta didik  kriteria cukup, baik, dan sangat baik (nilai hasil belajar > 65) pun naik, dari awalnya 9% pada pra siklus bertambah menjadi 97% pada siklus II. Penelitian ini menyimpulkan bahwa penggunaan media surat kabar mampu meningkatkan motivasi belajar dan berpengaruh pada peningkatan hasil belajar peserta didik pada mata pelajaran PKn.

Abstract

This study aims to determine whether there is influence between mastery of the use of a dissecting microscope to the value of laboratory techniques in management practicum student of biology education courses 2015 academic year 2015/2016. This research is a quantitative research, the independent variable is the mastery of the use of a binocular microscope, while the value of the dependent is a practical value. Collecting data using the method of documentation and testing methods such as pre-test and post test. To know domination influence of binoculer microscope to value practicum at this research applies correlation analysis and one way anova. From one way anova analysis can be concluded that the 5% significance level there is a real effect between mastery binocular microscope to count the value of his lab where F = 3,292 with significant value 0.046. From inferential one way anova analysis that with level of signifikasi 5% there is real influence between dominations of binocular microscope to value practicum where F calculate is = 3.292 with value signifikasi 0,046

Abstract

Prolonged drought impacts very significant for some areas of Indonesia causes the limited availability of surface water. Increasing need for clean water is not commensurate with the available surface water supply. Cemara village, Suboh District of Situbondo is one example of a water crisis in the dry season. Only rely on spring water from the mountains to meet the water needs of citizens. To improve the supply source of clean water, some alternative activities had been done, one of which is to identify subsurface aquifer layer with resistivity geoelectric method utilizing Schlumberger configuration. The research activities include measurements and observations in the field, data processing, and data interpretation. The results of processing and interpretation of the data concluded that the rock layers in the study site is in the form of soil or rock composed of volcanic tuff, breccia, sandstone, and sand as aquifer layers. At each point geoelectric identified aquifer layer that is able to store and distribute groundwater well at different depths. As a recommendation to carry out drilling , geoelectric point 2 has more potential aquifer layers . Aquifer layer was detected at a depth of 60 meters and 135 meters .

Abstract

This study examines the relationship type of monitor with eyestrain computer users in the computer lab of the Faculty of Science and Technology UIN Sunan Kalijaga. The study involved 192 respondents who use computers in the computer lab. Measurement of eyestrain performed by questionnaire to the respondent. The results showed that there has been eyestrain experienced by 160 respondents (83.3 %) . Pain in the neck, back and shoulders (13.7 %) is a sign of eyestrain often occur. Using chi square statistical test showed relationship between the type of monitor to eyestrain computer users (p-value = 0.043) . CRT monitor has a refresh rate that is smaller than the LCD monitor. The flicker on a CRT display make the eyes become tired quickly.

Abstract

Industrial workplace accidents are accidents that occur in the workplace, especially in industrial environments, where there is no element of intent or sabotage elements in it. Hazard is the activity, objects, components that are considered to cause damage or disruption of the process / activity therein to accidents. WL ALUMUNIUM as SMEs need sustainability production which can not be achieved in case of work accidents or worker safety is not assured due to the existing hazard. Therefore, this research wanted to determine the types of hazards that occur in these SMEs and risk analysis of existing hazard. Hazard Identification (HAZID) is a process of identifying hazards as a first step of determining the risk analysis. There are two main objectives in the hazard identification to obtain a list of hazard and the order based on the level of risk as well as to obtain a qualitative assessment of hazards and ways to reduce the hazard risk level. Based on the processing and analysis that has been done can be seen these SMEs have a hazard which are in the category of Low, Moderate, High and Unacceptable.

Abstract

The quality of an electrical connection engine shaft which is not straight and not exactly center can damage the engine crankshaft and the rotated engine. The Power used will be enlarged and the temperature of the equipment will increase. Increasing temperatures will shorten the life of the equipment. Previously, to test the quality of the connection shaft, we use a waterpas. Disadvantage of this method is a waterpas, only measures the flatness of the field in one or two areas only. Watpasdroid application developed as a quality improvement solution for shaft connection which is cheaper and easier. With this app, android phones can be used to measure how much the value of the tilt on the axis x, y and z. The value-added applications are high level of accuracy and simplicity. Without the need to add another device, the Android phone quite put on the field to be measured. Watpasdroid already used in Electric Drive laboratory in Electronic Engineering Polytechnic Institute of Surabaya (PENS), to check the connections of the engine shaft on motor loading test. Application developed using Basic4Android program (B4A), which uses the Basic programming language, which is easier than the other developer programs based on Java.

Abstract

Integrated Laboratory UIN Sunan Kalijaga Yogyakarta is a testing laboratory accredited by the National Accreditation Committee (KAN) with number LP-635 IDN. One parameter is the viscosity testing. Integrated Laboratory UIN Sunan Kalijaga Yogyakarta implementing viscosity test method MU01 / VIS / 2011. This test method validation necessary in order to have the valid test results. Validation is conducted every year to obtain a quality assurance test results. Validation carried out because the test method developed by the laboratory. Validation is done by measuring the level of precision and accuracy using standard materials. Measurement of standard materials does as much as 12 times the measurement. From the measurement results obtained at the price of precision is under 0.05% and accuracy between 99 % -100 %. In the test method validation can be concluded that the test method MU01 / VIS / 2011 fit for use as a test method. Validation testing methods viscometer above, still meet the requirements at a price of precision and accuracy.

Abstract

The Atwood machine is the equipment consists of two masses connected with string or rope on a pulley fixed. In the Atwood best practical implementation by using a stopwatch timekeeping is done manually lead time calculations are inaccurate because it depends on the precision of the practitioner. Manufacture of electronic equipment by utilizing light-sensitive sensor that can automatically record the time interval in experiments using the calculation time is expected to minimize these inaccuracies. This electronic equipment uses the principle of the switch transistor, where the transistor is in two circumstances, namely the state of saturation and discontinuity. Specifications of this electronic circuit when the infrared light is not blocked by an object, the resulting output voltage of 11.07 volts and when the infrared light is blocked by an object, the output voltage drops to 1.2 volts using input voltage of 12 volts. From the research could have made an electronic device that can be used to record time lapse automatically by using a light-sensitive sensor based phototransistor. Series that has been created can be utilized in the best lab Atwood. The result of the calculation of the acceleration of gravity by using the self-timer on the plane practicum Atwood is 9.791 m / s2.
