Abstract

Cimanggu progeny clove has been distributed to almost of centre production, such as in Sumedang. However, their genotypes are still unidentified. Therefore, clustering cloves accession population in Sumedang is necessary. The objectives of this study is to cluster and determine phylogenetic relationship of Cimanggu progeny clove population in Sumedang base on morphological characters. The experiment was conducted in Sumedang (West, Java) from 2013 until 2014, by using 10 selected accessions (healthy, more than 40 years old, productivity is ≥ 20 kg dried flower). Variables were observed include morphological leaf, flower and flower production. To distinguish genotype from each accession was conducted by cluster analyzed, while to identify the variables which were related to the clusters formation, used correspondence analysis. The results indicated, that clove population Sumedang can be classified into three clusters as followed: the first cluster are Syar 43 and 46 accessions, characterized by greenish orange and greenish purple of young leaves tip with flower tube greenish red (GR 180B), the second cluster are Syar 44 and 45 accession, characterized by tip reddish purple young leaves and greenish red (GR181B) flower tubes color, and the third cluster are Syar 47, 49, 50, 51 and 52 accessions, characterized by orange young leaves tips and greenish red (GR180C) flower tubes. This information can be utilized to support releasing Cimanggu clove variety.Keywords: Syzygium aromaticum, clustering, characters, morphology

Abstract

Strong opinions have been expressed toward oil palm plantation industry in Indonesia, and these opinions portray that oil palm plantation development in Indonesia has caused the destruction of the environment. One of the efforts currently undertaken by the Indonesian government to ensure the sustainability of the palm oil industry is establishing sustainability standard called the Indonesian Sustainable Palm Oil (ISPO) which is mandatory. ISPO is "guidance" for sustainable oil palm development as well as a commitment based on the laws and regulations aplication of some licenses in Indonesia. The purpose of this study was to determine the capability of oil palm plantation companies in the regional to meet the standards of the Indonesian Sustainable Palm Oil (ISPO) and to identify the problems faced in achieving these standards. This research was conducted in East Kalimantan province on the period June 2012-May 2013. Evaluation of the capability of the companies to achieve the ISPO standards was performed by the audit method, assessment results of all parameters that had been established in accordance with the Principles, Criteria and Indicators in the provision of ISPO which were assessed in a percent unit. The research results showed that the capability of the plantation companies in East Kalimantan in meeting the ISPO standards reached 79,14%, and this capability can be improved up to 100% by increasing efforts to comply with the principles, criteria and indicators that are still not in accordance with the provisions of ISPO are as follows: 1). Licence system and plantation management, 2). Aplication guidance cultivation technics and processing palm fruit, 3). Management and monitoring invironment, 4). Resposibility to workers, and 5). Social resposibility and community. The determantion factor performance on ISPO standard is the commitment of plantation company as a business stakeholder that supported by capable human resources to create the sustainable plantation development and also the goverment action in charge as the regulator in supervising the policy that has been set. Socialization and training about principles and criteria ISPO standard to plantation company needs soon and more intensively done by the government association with ISPO commission to accelerate the application of the ISPO. Socialization and training also are required in order to overcome the constraints in attainment of principles and criteria of the Indonesian Sustainable Palm Oil standard, because of the mentioned efforts are the part of success factors to apply the ISPO.Keyword: ISPO, Management, Palm Oil, Plantations, Sustainable.

Abstract

There are several ways to eliminate virus, suach as the application of thermoterapi and chemotherapy technique, and also the apex and meristem culture. One way to control this disease is the use of virus-free seedlings. The objective of this study was to find out the effect of combined treatment between thermotherapy or chemotherapy with apex or meristem culture to mosaic virus elimination of sugarcane. There were four steps in this research: (1) Virus detection of mother plant, (2) Application of thermotherapy at 50oC and chemotherapy by using Ribavirin 25 µg/l combined with apex culture, (3) Application of thermotherapy and chemotherapy combined with meristem culture, and (4) Evaluation of virus elimination. The plant materials used were PS862 from Cirebon (PS862-Crb), PS881 from Surabaya (PS881-Sby) and PSJK922 (PSJK922-Bgr) from Bogor. Virus detection was conducted by TEM and RT-PCR analysis. The temperature for thermotherapy was 500C and the antiviral agent was Ribavirin (0 and 25 μg/l). The result showed that thermotherapy or chemotherapy combined with apex culture could not eliminate virus infection. The combined treatment of chemotherapy and meristem culture could eliminate SCSMV in variety PS862-Crb based on RT-PCR assay, however TEM analysis still detected the viral particle. It was suggessted to udertake virus indexing of large number of samples to see the rate of virus elimination.Keywords: Saccharum officinarum L., Ribavirin, Potyvirus, TEM, RT- PCR

Abstract

Root induction is an important step in the process of propagation of plants through tissue culture techniques. For root induction commonly used plant growth regulator auxin, which has an important role in plant growth and development, especially at the stage of root initiation. The purpose of this study is to obtain appropriate methods for root induction of oil palm in vitro and ex vitro. The experiment used completely randomized factorial design, consisted of three main activities: (1) Induction of rooting on solid media, using combinations of NAA (2 and 4 mg/l) and IBA (1, 2, 3 and 4 mg/l) concentrations with 5 replications; (2) Induction of rooting the liquid media, using three concentrations of NAA (0, 3 and 6 mg/l), each treatment was replicated 5 times; (3) Induction of rooting ex vitro, using rootone F or IBA (20, 40 and 60 mg/l) with 5 replications. Results indicated that the best medium for in vitro root induction on solid media was MS + NAA 4 mg/l IBA + 4 mg/l, while for liquid media was MS + NAA 6 mg/l. Ex vitro rooting induction showed 60% success of acclimatization by soaking vitro shoots base in NAA solution 40 mg/l and 60 mg/l for 1 hour.Keywords: Elaeis guineensis Jacq., ex vitro, IBA, NAA

Abstract

Vascular Streak Dieback (VSD) in cocoa caused by Oncobasidium theobromae. This is one of the important disease of cocoa in the world, including Indonesia. The knowledge on environmental effect and agronomical practices to the epidemics of VSD disease of cocoa, is important to control the diseases. The objectives of this research were to investigate environmental effect and agronomical practices to the epidemic VSD disease of cacao. The study was conducted on February to August 2014 in the three sub districts namely Tinondo (≥ 700 m asl), Mowewe (300-400 m asl) and Lambandia (≤ 100 m asl) East Kolaka Regency, South East Sulawesi. The study was carried out by colleting primary data included agronomical practices, the measurement of VSD disease severity in the field and soil nutrient, as well as secondary data. The results of this research showed that the environment and agronomical practices gave an effect to the epidemics VSD disease. Monoculture plantation had higher severity than mixed garden of cocoa. More condense the canopy of cocoa, more severe the VSD disease. More near the location of cacao plants to the river, more humid the enviroment and more severe the desease. Potassium deficiency in plants will cause susceptible to the disease, the high levels of Mg may limit K and Zn uptake at the soil. Recommendation for reducing VSD disease severity on cocoa plantations are cropping systems mixed with other plants, prunning, reducing the use of insecticides and herbicides, and do not plant the cocoa near to the river.Keywords: cacao, disease, epidemic, Oncobasidium theobromae

Abstract

The obstacle in developing coconut water-based product is its easily altered properties. Ultrafiltration and ultraviolet processing are potential to obtain a longer shelf life for coconut water drink without altering its nutritional values and unique organoleptic properties, unlike other processing techniques e.g. pasteurization and ultra high temperature. Shelf-life estimation experiment showed that ultrafiltration-and- ultraviolet-processed coconut water without any addition of food additives can be stored for 51 days in 00C. The objective of this research was to evaluate the effects of ultrafiltration and ultraviolet treatments on the nutritional, physicochemical and organoleptic properties of coconut water drink. The experiments were carried out at Food Analysis Laboratory, Indonesian Center of Agricultural Postharvest Research and Development, on January-April 2015. Coconut water was flown through the ultrafiltration membrane unit and ultraviolet light unit, samples were collected in three repetitions. Nutritional composition and physicochemical properties of fresh coconut water (FCW) and coconut water drink obtained from ultrafiltration and ultraviolet process (CUU) were evaluated and compared. Organoleptic analysis was done by 20 panelists, observations included quality hedonic (aroma, sweetness, saltiness, sourness and turbidity), and acceptance (preferance and ranking test), comparing FCW and CUU with commercial coconut water drink (CWD). CUU showed indistinguishable nutritional composition and physicochemical characteristics from FCW (p>0,05), except on total sugar (p=0,049), clarity (p=0,001), L* (lightness) (p=0,000) and b* (yellowish) (p=0,002). Panelists gave CUU a statistically equal rank to FCW, and better than CWD. The organoleptic characteristics of CUU were concluded as relatively same in saltiness and aroma as FCW, and less intense in sweetness and turbidity compared to CWD. After 10 days storage, panelists level of liking was higher for CUU compared to CWD in color (p=0,004) and general appearance (p=0,016).Keywords: coconut water, nutritional composition, organoleptic properties ultrafiltration, ultraviolet
