Abstract

Study on determinant factors on success of Sumatran orangutan reintroduction was done to all orangutans in reintroduction station (84 individual + 1 infant orangutan). This study was conducted from August to December 2006 in the Station of Orangutan Reintroduction Centre located in Buffer Zone of Bukit Tiga Puluh National Park. The purpose of this study was to measure the effects of determining factors to the success of orangutans reintroduction to their natural habitat and to identify other factors that give impact to the reintroduction success. Data were collected through direct observation of activities and daily behavior of orangutans using scan sampling and continuous recording methods as well as through interviews with officials. Data analysis was performed descriptively and quantitatively using statistical tests (chi–square) with 95% confidence interval. The results showed that there is a match between percentage of success of the reintroduction of the six criteria of reintroduction success. The results of chi–square indicates that determinant factors, such as age class, orangutan life history, treatment and duration of reintroduction stage, are correlated to the success of reintroduction activities.

Abstract

Study on the effect of dose management on water irrigation and the effect of pruning corn lower leaves on productivity of Lamuru variety grown on dry land of dry climate area has been done. The experiment was organized at Naibonat Experimental Station, East Kupang Subdistrict, Kupang, East Nusa Tenggara, from June to October 2011. The area used was 72 × 32 m using split plot design methods, employing two treatments. The first treatment was conducted on the main plot with water irrigation dose given 100, 80, and 60% the dose control was according to the farmer’s customary. The second treatment was applied by pruning or not pruning the lower leaves at the generative phase on the determined submain plot. The result showed that dose of water irrigation of 80 and 60% gave significant effect on corn productivity, that is 7.3 and 5.3 ton/ha, but not significant to the control (6.6 ton/ha). On the other hand, the treatment with and without leaf pruning did not give significant effect on the growth component because pruning was conducted during the generative phase. The Anova test result of corn productivity was not significantly different between the treatment of with and without pruning. The treatment of 80% dose of water irrigation could save water until 842 m3 or 20% per planting season per hectare. Meanwhile, the farmer’s customary wasted the water up to 2.105 m3 or 50% per planting season per hectare. Based on the results, the optimizing of water irrigation interval is seven times more efficient in one planting period as compared to 14 irrigation times in one planting period.

Abstract

This study aims to determine the presence of the Streptococcus equi causes Strangles in horses. Inspection carried out by isolation and identification of bacteria from 20 samples nasal swabs of suspected horse disease Strangles. Isolation and identification of activities performed by the method of bacterial culture, Gram stain, catalase test, and with sugar test. Results obtained from a series of tests to identify the bacteria Streptococcus equi of 20 horses suspected by clinical strangles, there is a horse infected by  Strangles disease.

Abstract

Global fish stock has been significantly declined over the past 3 decades, especially in coastal waters. Over-exploitation of fish resources and habitat destruction has been considered playing important role in declining the fish stock. This study was conducted to restore sea cucumbers stock in Pulau Seribu. A base line study was needed to evaluate the status of seagrass ecosystems in restocking locations. The results showed there were 8 seagrass species identified in both study sites, covering 0-65% area, consisted of 20.45 ± 11.28% in Pulau Pramuka and 6.00 ± 6.56% in Pulau Kelapa Dua, respectively. Seagrass ecosystem in Pulau Pramuka indicated better condition than that in Pulau Kelapa Dua and statistically was significantly different (global R = 0.193; p < 0.001). However, to support the restocking efforts of sea cucumbers, the seagrass habitat in both locations need to be restored.

Abstract

Dry fractionation will produce a fraction of olein and stearin with different composition of high melting and low melting as well the distribution of triacylglycerol (TAG) in the form of trisaturated (St3), disaturated (St2U) and monosaturated (StU2). This research aimed to study the effect of critical cooling rate and crystallization temperature on the composition and profile of TAG changes as well the melting properties of the fractionated coconut oil products. Coconut oilwas heated at 70°C then cooledat differentcooling rate to reach various crystalization temperatures. The oil was then stirred at 15rpm and allowed to crystallize at different period of time, and finally fractionated by vacuum filtration using Whatman #40 paper. Fractionation temperatures were the same as crystalization temperatures. The results showed that the critical cooling rate was proportional to the solid-like to liquid-like ratio, the content of St3 and SFC profile of olein fraction but inversely proportional to the content of St2U and StU2 TAG. Interval crystallization temperature between 21.30 and 21.73 °C produced the S/L ratio, the content of St3 TAG and SFC profiles of olein fractions lower and the content of St2U and StU2 TAG higher than the temperature interval below or above it. Coconut oil fractionation more effective in higher crystallization temperature or lower critical cooling rate. In these cooling treatments, St3 TAG which has high melting pointwould be concentrated at stearin fraction, while St2U and StU2 TAG and MCTwould be at olein fraction. Therefore, it will increase melting properties of stearin fraction and decrease olein fraction.

Abstract

The color of natural dyes, as anthocyanins, is very easily changing as he temperature changes. The color change can be used as a color indicator to provide information about product deterioration due to its sensitivity toward temperature and light. This indicator is called smart label of color indicator. This film label can be made of chitosan and polyvinyl alcohol and extract of erpa (Aerva sanguinolenta) leaves. The extraction was done under various leaf:water ratios. The filming was done in 2 methods, i.e. (i) by mixing the extract into film solution and drying the film at 50 °C; and (ii) by brushing the extract on the film surface. The leaf extract had a pH of 5.09 and the total yield of 116.65 mg anthocyanin/100 g leaves. Filming of color indicator in mixing method and dried at 50 °C resulted a colorless film. Brushing method was the best method to produce smart label with quite stable property and 1 mL extract could be applied evenly per 40 cm2 film area. Thus, this film can be utilized as smart label with color indicator to detect the deterioration in temperature-sensitive products.

Abstract

Starfruits is one of exotic tropical fruits that have increasing its popularity both for domestic and global market due to its unique shape and taste, as well as their nutrition values. However, this fruit is considered perishable product with short self-life, thus the proper post harvest treatment should be done to prevent their quality after harvest. In general, the objective of this research was to determine optimum treatment of hot water treatment (HWT) and immersion period in CaCl2 solution using response surface method (RSM) for post harvest handling of starfruits in order to maintain its quality during storage. Among quality parameters tested, weight loss and hardness were found as the most responsive to the combination treatment. Parameter of weight loss rate showed a maximization response with stationary point of HWT’s temperature of 42 °C, expose period for 42 minutes, and immersion period in CaCl2 solution for 38 minutes with respond point of weight loss was 3.74%. Meanwhile, the hardness parameter achievement was the minimum with stationary point of HWT’s temperature of 48 °C, expose period for 35 minutes, and immersion period in CaCl2 solution fpr 35 minutes resulted respond point of hardness of 0.63 Newton. Interest level from the panelist were color, taste, texture and aroma, respectively, whereas the treatment combination that accepted by panelist were (45 °C, 65', 40'), (45 °C, 40', 15'), and (45 °C, 40', 40’).

Abstract

Anatomically, bamboo is constructed from many types of cells. Vascular bundles are the cells which give the highest contribution to the bamboo strength, thus the density distribution of vascular bundles could be used as the main variable for analyzing the layer system of bamboo strip and laminated bamboo. The density of vascular bundles distribution decreases gradually from the outer to inner part in a regular manner which could be fitted by linear or nonlinear function. Ratio of modulus of elasticity (E) which widely used in transformed cross section (TCC) method for analyzing the layer system are substituted by ratio of density distribution of vascular bundles with asumption that both are highly correlated. The data in this study proved that there are high Pearson’s corellation between the theoretical and the empirical result, and the paired t-student test also showed both are not significantly different; thus, this new method could be applied in very good result. There are 3 models applied in this study, namely linear, logarithm, and power. Power model is the best among others since its theoretical gives the nearest estimation to the empirical measurement.

Abstract

The aim of this study was to find chemical and microbiological characteristics of instant bandrek and nutmeg syrup from Sinarsari and Dramaga villages, Dramaga District, Bogor Regency. The analysis resulted that both products still fulfilled the quality standard of microbiological set by the Indonesian National Standard. The antioxidant capacity of instant bandrek and nutmeg syrup consecutively was equivalent with 256.12 mg AEAC per serving size and 775.62 mg AEAC per serving size, respectively. The toal phenolic of the instant bandrek drink and nutmeg syrup consecutively was equivalent with 89.81 mg GAE/L and 140.68 mg GAE/L, respectively. It can be concluded that the food processing did not remove the bioactive compounds that could function as antioxidant.

Abstract

Reverse transcriptase (RT) enzyme is a key tool in molecular biology for the synthesis of complementary DNA (cDNA) from messenger RNA (mRNA). Combining RT activity with PCR amplification has been a gold standard as the first step in cloning the coding region of any gene of interest. Evidently, RTs have been critical in advancing molecular biology, genetics and medicine to their current stage. In this study, we were developing the RTDRNase H recombinant enzyme isolated from serotype-2 simian betaretrovirus-2 (SRV-2) infected Indonesian Macaca fascicularis using Escherichia coli expression system. The study was conducted using RT SRV-2 gene expression using E. coli expression system, proteins purification, and application to RT PCR technique. The SDS PAGE expression analysis showed a specific band size of 32.7 kDa assumed as RT protein enzyme. Application of RT SRV-2 enzyme generated to the RT PCR technique of β-globin CDV and SRV-2 env gene target showed that the RT SRV-2 enzyme was capable to reverse transcribed mRNA into cDNA as indicated by the presence of specific DNA band compared with commercial RT enzymes. This RT SRV-2 enzyme showed its activity similar to that of commercial one, although the activity was lower.
