Abstract

The objective of this study was to investigate the effect of different types of Seaweed (Kappaphycus sp) and depth variation on the growth, production and carrageenan content of Seaweed using a top down cultivation method. Top down method combined the two methods of cultivation, the method of surface and off-bottom method. The types of Seaweed used were Kappaphycus alvarezii (Brown Maumere and Local Maumere) and Bali Seaweed (Kappaphycus striatum). This study was designed using a factorial experimental design, where is comprised of two factors: the types and water depth.  Parameters measured were daily growth rate, production and carrageenan content of Seaweeds.  The results showed that different types of Seaweed gave a significant effect (P<0.05) on daily growth rate and production of Seaweeds, however depth variation did not give a significant effect on daily growth rate and production of Seaweeds (P>0.05).  A combination of Seaweed types and depth variation did not give a significant effect on daily growth rate and production of Seaweeds (P>0.05). The highest carrageenan content was found in Local Seaweed (42.15%).  Brown Seaweed produced 40.59% of carrageenan content while Bali Seaweed produced 35.80%.

Abstract

Blue Spotted Ray (Dasyatis kuhlii) as the demersal organisms has a lot of accumulation of pollutants in the water that settles to the bottom. This study was conducted to look at the liver tissue alteration of Blue Spotted Ray caused by contamination of mercury (Hg). Performed on 5 tanks. Tank A as a control, tank B with concentration 0.025  ppm,  tank  C  with  concentration  of  0.05  ppm,  tank  D  with concentration  0.1  ppm  and  tanks  E  with concentration  0.2  ppm.  The  result  is  alteration  on  the  liver  tissue  in  the  form  of  atrophy,  cloudy  swelling, vacuolization degeneration and even necrosis or death of liver tissue already. The alteration can reduce and eveneliminate the function of the liver as a toxin absorbent and can lead to death.

Abstract

The objective of the present study was to use selected RAPD markers (Primer UBC-122 and YNZ-22) in parental selection to the bacterial Vibrio alginolyticus resistance in Tiger groupers (Epinephelus fuscoguttatus).  Eighteen fish parent samples were challenged with Vibrio alginolyticus and parental selection was examined by a PCR-RAPD method.   The results showed that from 18 fish samples, primer YNZ-22 generated 18 fragments while primer UBC-122 generated 8 fragments only (KHP, KB, HB, PB, MKH, MH, K and MK).  In addition, the average number of fragments from each samples generated from primer-YNZ was higher (3,2) than from UBC-122 (2,3). This indicates that primer YNZ-22 is better than primer UBC-122 to use in parental selection to the Vibrio alginolyticus resistance in Tiger groupers.  From the parental selection, five fish parents (KHP, PB, MH, K and  MK) were selected for their high resistance to the bacterial Vibrio alginolyticus.

Abstract

Feed eficiency is largely determined by its availability in suficient quantities and on timefeeding, s o it is necessary to study the frequency of feeding for the best production of soft shell crab. Four feeding frequency (1 time per day, 1 time per 2 days, 1 time per 3 days, and 1 time per 4 days) tested in this study. Feed with nutrient composition of 30.86% protein, 7.2% fat, nitrogen free extract (NFE) 48.89%, crude fiber 5.7% enriched with vitomolt 0.1041 5 mg/g of feed to the standard dose and 0.4166 mg vitomolt/g feed to high doses; or equal to 700 ng vitomolt/g  crab. Test crabsof Scylla  sp. reared in  crabs box and placed in  the pond. The results showed  the frequency of feeding 1 time per 2 days resulted in the highest percentage of molting (66.67%) and the lowest percentage of moulting was found at 1 per 4 times a day of feeding (36.67%). Growth param eters such as weight and carapace width, and feed eficiency did not difer among all treatments. Thus, feeding in the soft shell crab for the maximum production can be done with a frequency of 1 time per 2 days .

Abstract

This study aimed to determine and compare community structure and assemblages species of zooplankton at Kodingareng and Lanyukang Island. This study used randomly sampling method and the sampling was conducted on July, August and October 2012. Data was analysed using at PRIMER software program. The results showed that zooplankton at Kodingareng and Lanyukang Isla nd had 48 taxa. 47 taxa was found when sampling at dry season and 25 taxa was found when the sampling at wet season. The species was only found at Kodingareng Island were Cypris and Amphipod, while the species was only found at Lanyukang Island was  Luciferidae zoea, Tad  Pole  and  Tintinid.  The  highest  average  of  zooplankton  abundance  was  found  at  dry  season  sampling  at Lanyukang Island, account for 670 individu/L. The result of ANOSIM showed that the level of significance of zooplankton  community  structure  a t  Lanyukang  and  Kodingareng  Island  was  0.5%.  Based  on  nMDS  plot  of zooplankton assemblages species between Lanyukang and Kodingareng Island showed clear seperation. The SIMPER result showed that there was no species seperator of community structure between those islands due to there was the same dominance of species, such as Calanoida, Cyclopoida, dan Chaetognatha.

Abstract

Ocean acidification afected marine organisms especially calcifying organisms, such as Halimeda sp. This study was conducted on June to September 2012 at laboratory of Research Center and Development for Marine, Coastal and Small Islands, Hasanuddin University. The aim of study  to determine the efect of acidic level on Calcium concentration and growth rate of calcifying macroalgae, Halimeda sp. The experiment design was used completely  random  design  with  three  treatments  and  three  replicates.  Analysis  variance  was  used  for  data analysis with advanced respon test. Tukey test was used to compare the diference between treatments. Water quality parameters were analyzed descriptively. The results showed that there was a significant diference of pH treatments on calcium concentration an d growth rate of Halimeda sp. The highest calcium concentration was found  at pH 8. In  conclusion, increasing pH level of media disturbed the calcifying  process of Halimeda  sp especially at pH level of 6, while growth rate of Halimeda sp was not afected wit h the decreasing of pH.
