Abstract

Fishing effort enhancement would put pressure on blue swimming crab resources and its ecosystem. The objective of this study was to determine the spatial distribution based on the size of the carapace width (CW), abundance, temperature, salinity, depth, and fishing intensity in Pati waters which was divided into two zones (zones 1=nearshore, zone 2=offshore). The result showed that the average of carapace width in zone 1 to zone 2 was significantly different (p <0.05), as well as between males and females. Average number of individual (abundance) was significantly different according to fishing areas and sex (p <0.05). The average of water temperature in zone 1 was higher than in zone 2, salinity in zone 1 was smaller than in zone 2, and the water depth in zone 1 was shallower than in zone 2. The higher the crab catch intensity leaded to the smaller the abundance and size of carapace width. Alternative management strategies were proposed such as to divert fishing grounds from coastal waters to offshore during low productivity season, to uphold rules on minimum legal size for catching (Lm = 107 mm),  to release of berried females catch, and to reduce accretion rate of gear and fishing fleet mainly in the coastal zone waters.Keywords:  blue swimming crab, carapace widht (CW), abundance, intensity of fishing

Abstract

The objective of the research was to determine the best enrichment of rotifer and the naupli of Artemia as feed  for mud crab, S. paramamosain larvae,  also to know their increasing of the DHA/EPA ratio and vitamin C content. Four treatments were tested, namely : A) Mud crab larvae every two days were given  rotifer and naupli of Artemia enriched with HUFA and vitamin C also added RICA-1 probiotic at 2 mg/L,  B) Mud crab larvae every two days were given rotifer and naupli of Artemia enriched with HUFA  and vitamin C,  C).  Mud crab larvae every two days were given rotifer and naupli of Artemia enriched with vitamin C, and D).  Mud crab larvae every three days were given rotifer and naupli of Artemia enriched with vitamin C. Result showed that the crablet production from the highest to the lowest was as follows: treatment A (177.5+17.6 ind./tank), treatment B (160+14.1 ind./tank) (A and B showed no significant different results (P>0.05)), treatment C (136+5.6 ind./tank), and treatment D (106+8,5 ind./tank) (C and D results indicated a significantly different (P<0.05) with treatment A).   Higher level of DHA/EPA ratio of 69.23% was found on rotifer enriched with HUFA, while on naupliofArtemia only 28,72%. Higher level content of vitamin C was 50.56% in rotifer enriched with vitamin C, while in naupli of Artemia was only increase to 35.56%.Keywords:  Scylla paramamosain larvae, enrichment,  HUFA, vitamin C, crablet

Abstract

Quality deterioration of fish is caused by various factors, one of them is due to the activity of cathepsin enzyme. Cathepsin enzyme activity can be inhibited by enzyme inhibitors. Inhibitors of the enzyme can be obtained from fish meat tissue by separating the enzyme-inhibitor complex with heat treatment. The purpose of this research was to extract and apply the cathepsin inhibitors to inhibit deterioration of fish quality. This research was conducted in three  steps: (1) the determination of the optimum extraction temperature for extraction of cathepsin inhibitors, (2) the determination of the most potent cathepsin inhibitors with various concentrations, and (3) the application of cathepsin inhibitors in inhibiting the deterioration of fish quality. The results showed that the cathepsin inhibitors was optimally extracted at 80°C. Cathepsin inhibitors diluted using buffer with a ratio of 1: 1 had the same relative activity without dilution. The process of quality deterioration of milkfish  control in chilling temperatures occured after 504 hours (21 days) storage with a 24-hour observation time interval. The complete process during observation was pre-rigor (0 hour ), rigor mortis (96 hours), post-rigor (360 hours), and deterioration (504 hours). The process of milkfish quality deterioration treated with soaking in  cathepsin inhibitors occured after 624 hours (26 days) storage with the detailed process as follows: 0 hour (pre-rigor), 144 hours (rigor mortis), 408 hours (post-rigor), and 624 hours (deterioration). Abstract

Mud crab (Scylla serrata) is one of small scale fisheries commodity that have high economic value and typically associated with good mangrove ecosystem. A habitat degradation will cause a serious impact on the existence of mud crab population. The puropse of this study was to assess the ecological quality of mud crab habitat condition on Bintan Bay’s mangrove ecosystem. Ecological data collection was conducted by using plots line transect for mangrove and the mud carb data collection by using fishermen catch (fisher based survey). The results showed the value of habitat quality index (HQI) of mud crab (Scylla serrata) ranged from 52-82 which mean in the “moderate” category (index value 43-66) and "good" (index value 67-90). It showed that Bintan Bay’s mangrove ecosystems was good enough to support the viability of mud crab. Habitat quality had a linier impact on body weight gain (R2 = 99.78%) and carapace width (R2= 99.21%). This showed that higher the index value of habitat quality, the size of the body weight and carapace width will increases. Keywords: Scylla serrata, habitat quality, ecology, mangroves, and Bintan Bay

Abstract

Freshwater eel (Anguilla spp.) is a catadromus fish that migrates from freshwater into seawater for spawning and return to freshwater during larvae stage to grow. The objective of this study was to determine species composition of tropical freshwater eels based on morphology collected from several rivers flowing in to the Palabuhan Ratu Bay. Young, adult, and glass eels were caught in August 2014 until Desember 2014. Glass eels were only caught in Cimandiri River. Freshwater eels were classified by ano-dorsal length (AD) divided by total length (TL), cluster analysis, and discriminant analysis based on morphometric data. The results showed different AD/TL of young and adult, i.e., 1.93±0.51 for A. bicolor bicolor, and 16.99±0.65 for A. marmorata, and three different AD/TL of glass eels i.e.,1.72±1.05 for A. bicolor bicolor6.60±1.79 for A. nebulosa nebulosa, and 15.07±1.04 for A. marmorata. Cluster analysis showed three groups of population  characters of young and adult eels i.e.  A. bicolor bicolor (2 groups) and A. marmorata (1 group). Discriminant analysis showed three groups of glass eels population i.e., A. bicolor bicolor, A. nebulosa nebulosa, and A. marmorata. Morphological identification showed that young and adult eels in Cibareno  and Citepus rivers had two species i.e., A. bicolor bicolor and A. marmorata. Glass eels from Cimandiri river had three species i.e., A. bicolorbicolor, A. nebulosanebulosa, and A. marmorata. A. bicolor bicolor was found in each rivers and as a dominant species in Palabuhan Ratu Bay.Keywords: A. bicolor bicolor, A. marmorata, A. nebulosa nebulosa, species composition

Abstract

Runoff of nutrients (N, P, Si) from the mainland would disrupt the balance of nutrients in the waters resulted in changing microalgae communities composition, especially between diatom and dino-flagellate (Dia/Dino). This study was aimed to analyze the relationship of nutrient ratios (N/P, N/Si, Si/P) in the waters, to the density ratio of the Dia/Dino in the coastal and marine waters of Spermonde archipelago, South Sulawesi, Indonesia, which experience nutrient enrichment. Nutrients and phyto-plankton samples were collected from the coastal waters of Tallo, Maros, Pangkep, and seawaters of Spermonde in three seasons (transition, wet, and dry). The results showed the average value (± stan-dard deviation) of Dia/Dino ratio in the coastal and sea waters is 1422±1742 and 1174±1290 in the transition season; 238±205 and 1039±2264 in the dry season; and 153±270 and 72±71in the rainy season. Meanwhile, in the marine waters the ratio of Dia/Dino ranges between 1 and 9839. The ratio of Dia/Dino was not different significantly (p>0,05) by spatial and temporal, but Dia/Dino ratio was different significantly (p<0,05) on the ratio of nutrients in coastal waters, where the value of N/Si ratio tends to ignite monospesies enrichment diatom (Chaetoceros) with a variant of 70,4%. Keywords: nutrient ratios, diatom, dinoflagellate, coastal, Spermonde

Abstract

The antibacterial activities of mangrove species, Avicennia marina, Sonneratia caseolaris (collected from Teluk Payo, Banyuasin, South Sumatera), Ceriops tagal, Rizhopora apiculata, and Sonneratia alba (collected from Sadai, South Bangka) were screened against Salmonella typhi and Listeria monocytogenes by agar disk diffusion assays. Extractions were conducted using organic solvents (methanol, ethyl acetate, and acetone, subsequently). Most of the extracts tested showed potential antibacterial activity against both pathogens. The methanol extracts of the bark from S. alba and the fruit from A. marina showed particularly large inhibition zones (15 mm) against S. typhi. The acetone extract of S. alba leaves showed the largest inhibition zone (14 mm) when tested against L. Monocy-togenes. Further partial purifications of selected extracts which showed strong inhibition were performed by silica gel column chromatography using various eluent compositions with different polarities. The third fraction of methanol extract from S.alba leaves eluted with chloroform:methanol (1:5) resulted in a remarkably large inhibition zone (23 mm) against S. typhi. The third and seventh fractions of acetone extract from S. alba leaves eluted with ethyl acetate:methanol (7:3) resulted in a large inhibition zones (15 mm) against L. monocytogenes. In addition, the sixth fraction of methanol extract from A. marina fruit eluted with chloroform : methanol (6:4) resulted in the largest inhibition zone (17 mm) against L. monocytogenes. These results indicated that mangrove extracts could be developed as potential biomaterials for biopharmaceutical as well as biopreservation industries.Keywords: antibacterial activity, mangrove, column chromatography

Abstract

An experiment to evaluate the efficacy of inactivated polyvalent bacterial vaccines to increase specific immunity of tiger grouper (Epinephelus fuscoguttatus) against bacterial infection was conducted in floating net cages located in Sumbawa Island, West Nusa Tenggara. Polyvalent bacterial vaccine was prepared by mixing three species of killed-bacteria (Vibrio harveyi, V. alginolyticus, and Photobacterium leiognathi) with ratio of 1:1:1. The density of bacterial vaccine was adjusted at 1010 CFU/ml. The vaccines were delivered by immersion. The control fish were immersed in sea water without vaccine. Fish with ± 6 cm of total length and ± 7 g of body weigth were used in this experiment. The fishes were reared in concrete tanks in hatchery of Gondol, Bali until they reached ± 9 cm of total length. Later, the fishes were reared for 6 months in floating net cages located in Sumbawa. Re-vaccinations were delivered 60 days and 120 days post initial vaccination. The results showed that titer antibody and survival rate of the vaccinated fish were 1/128 and 77.11%, higher than the control (titer antibody=1/4-1/8, survival=45.56%, respectively). The relative percent survival of the vaccinated fish was 57.95%. It was suggested that the vaccines were immunogenic and had protective ability against bacterial infection in tiger groupers. Keywords: bacterial disease, polyvalent bacterial vaccine, tiger grouper

Abstract

Sediment oxygen consumption level is an indicator of mineralization procces intensity and indicator of microorganism activity in the substrate, and also a description of dissolved oxygen consumption or utilization in a water body. This study aimed to evaluate factors of sediment quality which influenced sediment oxygen consumption level. The study was conducted at the experimental pond installation of Research Institute for Coastal Aquaculture (RICA), in Punaga, Takalar Regency, South Sulawesi, using two units of 4000m2 intensive ponds with density of 50ind./m2. Rearing period was approxi-mately in 4months. Pond management was carried out in accordance with standard operating pro-cedures. Homogeneity between experimental units was approached by the uniformity of pond management including pond preparation, shrimp seed management, feed management,and water management. Sediment quality variables measured were redox potential, pH, total organic matter, sediment oxygen consumption, total bacteria, and production aspect. The results showed that the factors which have close relationship with the sediment oxygen consumption were redox potential, organic matter, and total bacteria. The model could be used to predict the sediment oxygen con-sumption on white shrimp pond with the following regression equation Y=  0.0000496 + 0.00025 organic matter + 0.00037 total bacteria - 0.00000948 potential redox with R2 = 0.847.Keywords: sediment oxygen consumption, sediment quality, pond, Litopenaeus vannamei

Abstract

The growth of pearl shell oyster is influenced by nacre availability consisting of calcium carbonate. Nacre is identified as a protein metabolism product in pearl oyster. The aim of this study was to evaluate the effects of media level pressure in larva rearing tank on protease enzyme activity to promote growth performance and survival rate of Pinctada maxima larvae. The research was conducted from March 6th to May 7th 2015 in laboratorium of Mataram Marine Bio Industry Technical Implementation Unit, LIPI. “Chi-Square” test showed that survival rate of larvae at Umbo-veliger stage (planktonic) was significantly affected by water level pressure within 99% with ratio 1.00:1.61. In contrast, treatment with no water level pressure also significantly influenced the survival rate of larvae in spat stage within 99% confidence with ratio of 1.00:1.93. The highest average survival rate of larvae in umbo-veliger and spat stages were 13.10% and 2.60%, respectively.
