Abstract

Gelatin is a biopolymer obtained through the partial hydrolysis of collagen and is widely utilized in the food, pharmaceutical, and cosmetic industries. This study aimed to evaluate the effects of acid type and concentration during the pretreatment process on the physicochemical characteristics, gel strength, and FTIR profile of gelatin extracted from Spanish mackerel (Scomberomorus sp.) bones, as well as to determine the optimum pretreatment condition for producing high-quality gelatin. Spanish mackerel bones collected from a fish processing industry in Ampenan Village, Mataram City, West Nusa Tenggara, Indonesia, were subjected to pretreatment using two types of acids, namely hydrochloric acid (HCl) and citric acid, each applied at three different concentrations. The evaluated parameters included yield, moisture content, ash content, pH, viscosity, gel strength, and functional group characterization using Fourier Transform Infrared (FTIR) spectroscopy. The results demonstrated that pretreatment with 4% HCl produced the highest quality gelatin, with a yield of 9.48%, moisture content of 7.72%, ash content of 0.86%, pH of 4.82, viscosity of 6.20 cPs, and gel strength of 163.63 g Bloom. All measured parameters complied with the requirements of the Indonesian National Standard (SNI 06-3735-1995) for gelatin. FTIR analysis confirmed the presence of amide A, B, I, II, and III absorption bands, indicating the characteristic collagen-derived structure of gelatin. Therefore, pretreatment with 4% HCl was identified as the optimum condition for extracting gelatin from Spanish mackerel bones.

Abstract

Granola potato (Solanum tuberosum L.) is a strategic horticultural crop in Indonesia, and its productivity largely depends on the availability of high-quality planting materials. In vitro tissue culture through shoot multiplication is an effective approach for the mass propagation of disease-free seedlings. Young coconut water contains natural cytokinins, auxins, gibberellins, amino acids, and minerals that have the potential to replace or enhance synthetic plant growth regulators in Murashige and Skoog (MS) medium. This study aimed to evaluate the effects of different concentrations of young coconut water on the morphogenesis of Granola potato plantlets, identify the optimum concentration for efficient shoot multiplication, and generate quantitative data on in vitro plantlet growth. The experiment was arranged in a completely randomized design with five concentrations of young coconut water (0, 50, 100, 150, and 200 mL/L) and four replications. The observed parameters included the number of shoots, plantlet height, number of nodes, number of leaves, and percentage of rooted explants. The results showed that supplementation with 100 mL/L of young coconut water produced the best morphogenetic response, with an average of 4.85 shoots per explant, a plantlet height of 6.42 cm, and 91.25% rooted explants. Concentrations above 150 mL/L tended to inhibit plantlet growth. The findings demonstrate that young coconut water at an optimum concentration is an effective natural source of cytokinins for supporting the in vitro multiplication of Granola potato plantlets.

Abstract

Critical thinking and problem-solving are essential twenty-first-century competencies that remain insufficiently developed through conventional instructional approaches, particularly in learning the human digestive system. This study aimed to examine the effect of the Group Investigation (GI) learning model oriented toward Socioscientific Issues (SSI) on senior high school students' critical thinking and problem-solving skills in the human digestive system topic, analyze differences in learning improvement between the experimental and control groups, and evaluate the effectiveness of the model based on the N-Gain index. The study was conducted at a public senior high school in Pekanbaru using a quasi-experimental pretest-posttest control group design. A total of 62 eleventh-grade science students participated as research subjects. Data were collected using validated critical thinking and problem-solving tests. The results revealed that the experimental group achieved higher posttest mean scores in critical thinking (74.06%) and problem-solving (72.20%) than the control group (61.42% and 59.07%, respectively). The N-Gain values of the experimental group were classified as moderate (0.57 and 0.55), whereas those of the control group were categorized as low (0.34 and 0.33). An independent samples t-test confirmed that the differences between the two groups were statistically significant (p < 0.001). These findings demonstrate that the SSI-oriented Group Investigation model is more effective than conventional instruction in improving students' critical thinking and problem-solving skills.

Abstract

The study of Protista in senior high school biology requires direct observation of microscopic organisms. However, limited microscope operation skills and low levels of students' science process skills often hinder effective learning. This study aimed to evaluate the effectiveness of a digital microscopy-based experimental method in improving science process skills (SPS) and psychomotor skills in learning Protista, compare its effectiveness with conventional light microscopy, and analyze the statistical differences between the two instructional approaches. The research was conducted at a public senior high school in Ternate using a quasi-experimental pretest-posttest control group design. The participants consisted of 63 tenth-grade science students, with 32 students assigned to the experimental group and 31 students to the control group. Data were analyzed using the N-Gain index and an independent samples t-test. The experimental group achieved mean posttest scores of 74.28% for science process skills and 76.27% for psychomotor skills, with N-Gain values of 0.59 and 0.62, respectively, both categorized as moderate. These results were significantly higher than those of the control group, which obtained N-Gain values of 0.37 for science process skills and 0.39 for psychomotor skills. The independent samples t-test confirmed significant differences between the two groups for both variables (p < 0.001). These findings indicate that digital microscopy is more effective than conventional light microscopy in enhancing students' science process skills and psychomotor performance in learning Protista.

Abstract

Aspergillus flavus is a major contaminant fungus of food commodities that produces aflatoxin B1, a highly potent carcinogenic compound that poses serious risks to food safety. The increasing resistance of A. flavus to synthetic antifungal agents has encouraged the exploration of natural alternatives, one of which is liquid smoke derived from mundu wood (Garcinia dulcis). This study aimed to identify the bioactive compounds present in three fractions of mundu wood liquid smoke using Gas Chromatography-Mass Spectrometry (GC-MS), evaluate the antifungal activity of each fraction against A. flavus, and determine the Minimum Inhibitory Concentration (MIC) and Minimum Fungicidal Concentration (MFC). Liquid smoke was produced through the pyrolysis of mundu wood at temperatures ranging from 350–400°C and subsequently fractionated sequentially using hexane, ethyl acetate, and water as solvents. GC-MS analysis identified 12 major bioactive compounds, with guaiacol (18.67%) as the dominant component in the ethyl acetate fraction and acetic acid (21.54%) as the major compound in the aqueous fraction. Antifungal activity assessed using the disk diffusion method showed that the 15% ethyl acetate fraction produced the largest inhibition zone (15.6 ± 0.26 mm), categorized as strong antifungal activity, with an MIC value of 2% (v/v). All fractions exhibited fungistatic properties. The ethyl acetate fraction of mundu wood liquid smoke demonstrated promising potential as a natural antifungal agent against A. flavus.
