Abstract

A total of 150 cattle consisting of 3 breeds (Bali, Madura and Ongole) of Indonesian native cattle were used in this study. The aims of the study were to identify and characterize polymorphisms in the growth hormone loci of those breeds of cattle. The genomic DNA was extracted using Wizard genomic DNA purification system from Promega. Two fragments of growth hormone gene were amplified using PCR and continued with RFLP using restriction enzymes of AluI and MspI. Polymorphisms were found at both loci of GH-L1 and GH-L2 of the growth hormone gene by PCRRFLP analysis in all breeds of those cattle.© 2002 Jurusan Biologi FMIPA UNS SurakartaKeywords: PCR-RFLP, growth hormone gene, polymorphism, Indonesian local cattle.

Abstract

The objectives of the research were : (1) to determine the distribution pattern, association, and interaction of plant species, (2) to determine the relationship between life form and the distribution pattern of species, and (3) to comparethe association chance of distribution patterns. Quadrate method was used in this project. Determination of speciesdistribution pattern was calculated using Poisson Distribution formula, and the determination of association was calculated using Contingency Table. The result indicated that: (1) The species richness were dominated by grasses, reaching 42.42%. Of the 33 observed species, 21 of them were in a clumped distribution, 7 species in a regular, and 5 species were at random pattern. (2) There are relationship between life form and the distribution of the plant species. The multi-plants tend to have a clumped distribution pattern, and single plant tend to have a regular or random distribution pattern. (3) The species of clumped distribution tend to have negative association to the species which of regular and random distribution pattern. However among the species which had the regular distribution pattern tend to have positive association. Abstract

The low production rate of shallot (Allium ascalonicum L.) in Indonesia could be caused by rarely excellent cultivars.Colchicine was one of the mutation agents frequently used in plant breeding in order to get polyploidy of cultivars. The aim of this research was to find out the differentiation of morphometric evidences and ploidy of shallot chromosomesinduced by colchicines 1%. Preparation was made by squash method and stained by acetocarmine. The results indicated that the amount, length and shape of chromosomes altered by the application of the agent. Abstract

Primary productivity dynamic of the water ecosystem was conducted faster in the last decades. This study was intended to find out the primary productivity of Cengklik dam Boyolali, Central Java to explain the ecosystem dynamic and to lead the maintenance of dam. This study used quantitative methods in completely randomized group design (CRD), and the data was analized by Analysis of Variance (ANAVA). Samples were taken horizontally in four sampling point, respectively in the riparian zone, around of the floating net (“karamba”), in the center of dam water and around of the ex-paddy fields. There were taken vertically in three-depth point in each of the sampling point, respectively 0.5 meter, 1.5 meter, and 2.5 meter. The results showed that the gross primary productivity of the dam was 11.122.500-22.545.600 mgC/m3/days, and the primary productivity differences in each of the point sampling caused by light intensity, nutrient supply, and abundance of the chlorophyll organisms.© 2002 Jurusan Biologi FMIPA UNS SurakartaKey words: primary productivity, Cengklik dam

Abstract

One of the biodiversity richness in Indonesia is ecosystem of Baluran National Park savanna. This type of savanna is similar to African savanna that is included in tropical savanna. There are two types of savanna, namely flat and undulating savanna. The savanna ecosystem that covers about 40% of the total area of Baluran National Park has important role on supporting herbivore animals such as wild cattle (Bos javanicus), deer (Cervus timorensis) and wild buffalo (Bubalus bubalis). The variety of grasses and other vegetation provide food for those animals, therefore, the reduction of the reduction of quality and quantity of the savanna would reduce in the population of the herbivores. The savanna in the Baluran National Park is about 10.000 ha, however, due to invasion of Acacia nilotica has resulted in the reduction of the savanna reaching about 50%. Moreover, illegal grassing inside the Park has brought negative impact to the quality of the area and the wild fauna. Overgrazing may influence on the reduction of the population of wild fauna. Pressure to the savanna has a great impact on the balance and preservation of whole ecosystem in Baluran. Efforts have been made in order to restore the Baluran ecosystem such as preventing the expansion of A. nilotica as well as preventing illegal grazing. Abstract

The aim of the experiment was to examine whether the morphological characters of eleven species of Ranunculuscollected from a number of populations were in agreement with the genetic data (isozyme). The method used in this study was polyacrilamide gel electrophoresis using peroxides, estarase, malate dehydrogenase, and acid phosphatase enzymes. The results showed that cluster analysis based on isozyme data have given a good support to classification of eleven species based on morphological groups. Abstract

The study of animal biodiversity that lived on the surface of soil under the stands forest surround Japan Cave BKPH Nglerak, North Lawu, Karanganyar has bee done. Observations were conducted in 6 stations of different stands of forest. Animals were caught by pit fall trap method. In each catching was found about 22 animals consisting of 6 families with Simpson’s diversity index of 0.5. The result of identification indicates that those animals belong to 4 classes: Insects (9 orders), Arachnids (2 orders), Diplopods (2 orders), and Crustacean (1 order).
